肝癌microRNA稳定性、个体差异性与性别相关性的分析

【摘要】目的:分析肝癌microRNA稳定性、个体差异性与性别相关性。方法:选取52例肝癌患者及29例健康志愿者作为研究对象,收集内镜下肝癌患者的肝癌组织、癌旁正常组织活检标本及所有研究对象的血清样本,利用应用免疫组化(SP) 法检测不同条件下肝癌组织、癌旁组织及血清样本的microRNA表达水平及稳定性;比较分析肝癌患者血清microRNA个体差异性与性别的相关性。结果:肝癌microRNA在不同温度、pH值及冻融次数等条件下均有扩增,具有抵抗降解及结构性质高度稳定;肝癌microRNA在肝癌组织的表达水平显著高于癌旁组织、血清及健康志愿者血清的microRNA表达水平(P<0.05)。肝癌microRNA表达水平的个体相关性分析R=0.998,P<0.05,microRNA的个体差异性极小;肝癌microRNA在性别差异性表达中具有显著性的相关性,R2分别为0.998和0.997。结论:肝癌microRNA的结构性质稳定,其表达水平可特异性区别肝癌患者与健康志愿者,个体差异性较差及与性别无相关性,可作为肝癌诊治的监测指标。【关键词】肝癌;microRNA;稳定性;个体差异性;性别相关性

【Abstract】Objective: To analyze mirnas stability of liver cancer, individual difference and gender.Methods: Selecting 52 patients with liver cancer and 29 cases of healthy volunteers as the research object, collect endoscopic hepatocellular carcinoma in patients with liver cancer tissue and normal tissue adjacent to carcinoma biopsy specimen serum samples and all the research object, by using immunohistochemical method (SP) test under different conditions of liver cancer tissue and normal tissue adjacent to carcinoma and the expression level of micrornas in serum samples and stability;Comparative analysis of the correlation of liver cancer patients serum mirnas individual diversity and

gender.Results: The liver cancer mirnas in different conditions, such as temperature, pH value and the number of freezing and thawing are amplification, highly resistance to degradation and the nature of the structure stability;Liver cancer mirnas in cancer of the liver tissue of

Conclusion: The liver cancer mirna structure properties of stability, and its expression level can specificity difference between liver cancer patients and healthy volunteers, individual difference is poor and has no impact on gender, can be used as a monitoring index of the diagnosis and treatment of liver cancer.

肝癌是指癌基因、抑癌基因及生长因子调控的粘膜上皮细胞增殖和凋亡平衡紊乱,导致肝脏或胆管上皮细胞、间叶细胞恶性分化的肿瘤;该病呈渐进性发展、多因素影响及病死率高[1-2]。目前分子生物学领域发现microRNA与肝癌的病变及诊断有关;因此,有必要从分子水平寻求诊断肝癌的肿瘤标记物,并研究其作用机制及标准化。microRNA作为内源性的单链 RNA,可直接促进其靶mRNA的降解或抑制其翻译表达,达到调控靶基因的转录及翻译表达。由于microRNA具有高度的特异性、时序性,调节癌基因及抑癌基因的平衡,在肿瘤的病发形成发挥重要作用[3]。杨桢[4]研究指出microRNA在肿瘤细胞的分化增殖、凋亡及转移等过程起到调控作用。对此,本文旨在分析肝癌microRNA稳定性、个体差异性与性别相关性。

1资料与方法

1.1一般资料

选取52例肝癌患者及29例健康志愿者作为研究对象;对于52例肝癌患者,其中男性患者28例(53.85%)、女性患者24例(46.15%);年龄范围26.5~78.7岁、平均年龄(56.5±5.5);活检病理分型为肝细胞癌32例、胆管细胞癌16例、转移性混合癌4例。排除标准:合并心脑血管疾病、其它组织器官肿瘤、呼吸、
