Inhibitors, Agonists, Screening Libraries Data SheetBIOLOGICAL ACTIVITY:HSF1A is a cell–permeable activator of heat shock transcription factor 1 (HSF1).IC50 & Target: HSF1[1]In Vitro: HSF1A protects cells from stress–induced apoptosis, binds TRiC subunits and inhibits TRiC activity without perturbation of ATP hydrolysis. Genetic inactivation or depletion of the TRiC complex results in human HSF1 activation and HSF1A inhibits the direct interaction between purified TRiC and HSF1 in vitro. Moreover, fluorescence anisotropy experiments using FITC coupled to HSF1A demonstrates that HSF1A–FITC binds to a purified Tcp1 subunit of TRiC with an affinity of approximately 600 nM. This is validated qualitatively via titration of purified Tcp1 into binding reactions containing 500 nM Biotin or HSF1A–Biotin [1]. Quantification bycounting the number of cell containing aggregates as a function of the total number of cells reveals that at HSF1A concentrations as low as 2 μM, a reduced number of aggregate–containing cells are observed. The fraction of cells containing aggregates continued to decrease in a dose–dependent manner such that pretreatment with 12 μM HSF1A resulta in ~20% of the cells exhibiting aggregates visible by fluorescence microscopy [2].In Vivo: HSF1A enhances HSF1 activity, stabilizes HSF1 expression and minimizes Doxorubicin (DOX)–induced cardiac damage. WKY rats are challenged with DOX (accumulated dose: 30 mg/kgw), and DOX combined with HSF1A (100 mg/kgw/day). Supplementation with HSF1A significantly elevates cardiac functions back to the levels of the control group. HSF1A has been shown to stimulate human HSF1 nuclear translocation, elevate protein chaperone expression and ameliorate protein misfolding and cell death in aneurodegenerative disease model. 丙二醛(malondialdehyde,MDA)含量测试盒说明书(货号:G0109F分光法48样)一、产品简介:丙二醛(MDA)是由于生物体官衰老或在逆境条件下受伤害,其组织或器官膜脂质发生过氧化反应而产生的。
它的含量与生物体衰老及逆境伤害有密切关系。
MDA在高温、酸性条件下,与硫代巴比妥酸(thiobarbituric acid,TBA)缩合,生成红色产物,在532nm有最大吸收峰,进行比色后可估测样品中过氧化脂质的含量;同时测定600nm下的吸光度,利用532nm与600nm下的吸光度的差值计算MDA的含量。

四、丙二醛(MDA)的测定:建议正式实验前选取2个样本做预测定,了解本批样品情况,熟悉实验流程,避免实验样本和试剂浪费!1、样本制备:①组织样本:称取约0.1g组织(水分充足的样本可取0.5g),加入1mL提取液,进行冰浴匀浆。
4℃×12000rpm离心10min,取上清,置冰上待测。
【注】:若增加样本量,可按照组织质量(g):提取液体积(mL)为1:5~10的比例进行提取。
②细菌/细胞样本:先收集细菌或细胞到离心管内,离心后弃上清;取500万细菌或细胞加入1mL提取液,在4ºC或冰浴进行匀浆(或使用各类常见电动匀浆器)。
4ºC约12,000rpm离心10min,取上清作为待测样品。
【注】:若增加样本量,可按照细菌/细胞数量(104):提取液(mL)为500~1000:1的比例进行提取。
③液体样本:直接检测。
若浑浊,离心后取上清检测。

BIOLOGICAL ACTIVITY:
RG108 is a non–nucleoside inhibitor of DNA methyltransferase with IC 50 of 115 nM.
IC50 & Target: IC50: 115 nM (DNA methyltransferase)
In Vitro: RG108 effectively blocks DNA methyltransferases in vitro and does not cause covalent enzyme trapping in human cell lines.Incubation of cells with low micromolar concentrations of RG108 results in significant demethylation of genomic DNA without any detectable toxicity. Intriguingly, RG108 causes demethylation and reactivation of tumor suppressor genes, but it does not affect the methylation of centromeric satellite sequences [1]. In another study, the synthesis and in vitro analysis of a biotinylated RG108conjugate is investigated to evaluate the interactions with DNA methyltransferase enzymes [2]. In a recent study, it is shown RG108
can significantly reduce the DNA methyltransferases activity in SM derived iPS cells as compared to the native SMs [3].PROTOCOL (Extracted from published papers and Only for reference)
Kinase Assay:[1]The substrate DNA for the in vitro methylation assay is a 798 bp fragment (-423/+375 relative to the initiation codon)from the promoter region of the human p16Ink4a gene. The methylation reaction contains 350 to 400 ng substrate DNA and 4 units of M.SssI methylase (0.5 μM) in a final volume of 50 μL. Inhibitors are added to final concentrations of 10, 100, 200, and 500 μM,
respectively. Reactions are done at 37°C for 2 hours. After completion, the reaction is inactivated at 65°C for 15 minutes and the DNA is purified using PCR Purification kit. Three hundred nanograms of purified DNA is digested for 3 hours at 60°C with 30 units of BstUI and analyzed on 2% Tris–borate EDTA agarose gels.
