双通道PAM-100测量系统商品编码:9027500000 品牌:WALZ 型号:DUAL-PAM-100原理:利用荧光发射器发射出来的光学射线来检测植物叶片细胞内叶绿体中的荧光。
功能:单独或同步测量微藻、大藻、水生植物等的叶绿素荧光和P700。
检测对象:微藻、大藻、水生植物。
详细原理:细胞内的叶绿素分子通过直接吸收光量子或间接通过捕光色素吸收光量子得到能量后,从基态(低能态)跃迁到激发态(高能态)。
处于较高激发态的叶绿素分子很不稳定,在几百飞秒内,通过振动弛豫向周围环境辐射热量,回到最低激发态。
最低激发态的叶绿素分子可以稳定存在几纳秒,重新放出一个光子,回到基态,即产生荧光,是处于较低激发态的叶绿素分子释放能量回到稳定基态的几种途径之一。
色素分子处于氧化态和还原态时,或增加/减少亚基后,其吸收峰也会有变化。
双通道PAM-100测量系统采用独特的调制技术,检测来自于植物叶片细胞内叶绿体中的光合系统复合蛋白体的叶绿素荧光,通过测量活体叶片的叶绿素荧光和P700吸收变化来全面研究植物两个光系统(PS I和PS II)的活性变化对光合作用的影响。
双通道PAM-100测量系统相当于两台脉冲-振幅-调制叶绿素荧光仪,通过远红光LED发出的远红光来激发PS I在较短的时间内到达最高氧化态,从而测得PS I 的最大能力,通过荧光发射器来发射光照强度很低的光能,进而检测初始叶绿素荧光及其他时期的荧光,蓝色LED主要提供波峰在蓝光光区的光源,很多藻类对光能的吸收利用主要在蓝光光谱区。
双通道PAM-100测量系统既可以进行复杂的叶绿素荧光分析(PS II活性),还可以通过测量P700的吸收变化来检测PS I的活性,可用于光合作用机理研究、植物生理学、农学、林学、园艺学等领域。

4、试剂五应用液的制备:取试剂五贮备液用无水乙醇20倍稀释后使用,建议当天配制当天使用。
5、标准品:临用前加入1 mL 试剂三,配成20 mg/mL 的标准品溶液。
产品说明:维生素E (Vitamin E )是一种天然脂溶性抗氧化剂,能阻断机体不饱和脂肪酸的过氧化,维持不饱和脂肪酸细胞膜的完整性和正常功能,并可清除超氧阴离子自由基,具有延缓衰老、预防溶血性贫血等作用,在医药、化妆品、保健品、食品行业具有较高的应用价值。
VE 还原Fe 3+为Fe 2+,Fe 2+与1,10-菲罗啉产生有色络合物,在510 nm 有特征吸收峰。
技术指标:最低检出限:3.0152 μg/mL 线性范围:3.125-50 μg/mL注意:实验之前建议选择2-3个预期差异大的样本做预实验。
如果样本吸光值不在测量范围内建议稀释或者增加样本量进行检测。
需自备的仪器和用品:可见分光光度计、天平、台式离心机、1mL 玻璃比色皿、可调式移液枪、研钵/匀浆器、漩涡震荡仪、无水乙醇、正庚烷、蒸馏水和EP 管。
操作步骤:一、样本处理(可适当调整待测样本量,具体比例可以参考文献)1.组织样本试剂名称组织(g)0.1蒸馏水(µL)200试剂一(µL)300试剂二(µL)500匀浆后在漩涡混匀仪上震荡5min(充分抽提),于25℃,5000g离心5min,取上层正庚烷抽提液300µL加入到900µL无水乙醇中(上层抽提液:无水乙醇=1:3)混匀待测。

研究发现,在加标量为2~800 μg·kg-1时,粮食中6种真菌毒素的平均回收率为75.5%~102.5%,批内的相对标准偏差为3.7%~9.7%,均满足食品理化检测要求。
由此可知,色谱及质谱检测技术适用于对粮食中多组分真菌毒素进行定量与定性检测。
