Collect 5.5 lbs (3kg) of ice from unit.Discard ice and Sani-Sponges (Fig. 11).ServiceIce machine operation (all models)Follett’s ice machine consists of fi ve distinct functional systems covered in detail as follows:• Water system• Electrical control system• Mechanical assembly• Refrigeration system• Bin fullThe Horizon ice machine overviewThe Follett Horizon ice machine uses a horizontal, cylindrical evaporator to freeze water on its inner surface. The refrigeration cycle is continuous; there is no batch cycle. The evaporator is fl ooded with water and the level is controlled by sensors in a reservoir. A rotating auger (13 RPM) continuously scrapes ice from the inner wall of the evaporator. The auger moves harvested ice through the evaporator into an ice extrusion canal. The ice is forced through a restrictive nozzle that squeezes out the water and creates the Chewblet. The continuous extrusion process pushes the Chewblets through a transport tube into a dispenser or bin.A solid state PC board controls and monitors the functionality of the ice machine. In addition to sequencing electrical components, the board monitors various operational parameters. A full complement of indicator lights allows visual status of the machine's operation. Additionally, the PC board controls the self-fl ushing feature of the ice machine. The evaporator water is periodically drained and replenished to remove minerals and sediment.A unique “bin full” detection system is incorporated in the Horizon ice machine. A switch located at the ice discharge port of the machine detects the position of the transport tube. When the bin fi lls up with ice, the transport tube moves out of the normal running position, and the switch turns the ice maker off. A domed housing at the end of the transport tube contains the ice extrusion loads during shut down.Harvest system diagramElectrical systemNormal control board operationThe PC board indicator lights provide all the information necessary to determine the machine's status. Green indicator lights generally represent “go” or normal operation; Y ellow indicators represent normal off conditions; Red indicators generally represent alarm conditions, some of which will lock the machine off.A fl ashing green light labeled POWER indicates power to the machine. There are two types of errors namely “hard” or “soft”. A hard error is one that shuts the machine off and will not allow restart until the reset button is pressed. Even cycling power will not reset a hard error. A soft error can either be automatically reset should the condition rectify, or if power is cycled. Should an error occur, consult the troubleshooting guide in this manual or a Follett service technician.Soft errors:HI AMPS: The PC board monitors the amperage of the auger motor. Should the gear motor experience current draw above the allowable limit, the machine will shut down and the TIME DELAY and HI AMP will be illuminated. After the time delay the machine will restart and the TIME DELAY and HI AMP will clear.LO WA TER: During operation, the water level cycles between the normal low and normal high sensors. Should the water be shut off to a running machine, a soft error will occur. The error sequence is as follows: During operation, the water level falls to the normal low sensor, and when it does the water feed solenoid is energized. If water is not detected at the normal low sensor within 10 seconds, a soft error will occur. The machine will shut down, but the water feed solenoid will remain energized. Should water return, it will fi ll to the normal low sensor and the machine will resume normal operation. The error will clear automatically.HI PRESSURE: Should the refrigeration pressure rise above 425 psi, the machine will shut down and the TIME DELAY and HIGH PRESSURE will be illuminated. After the time delay, and if the pressure has fallen back below the reset point of 295 psi, the machine will restart and the TIME DELAY and HIGH PRESSURE will clear.Hard error:DRAIN CLOG: The drain clog sensor, located in the chassis, underneath the rear drain pan, will detect the presence of water just below the top edge of the pan. If water does not properly fl ow out of the drain pan it will overfl ow into the chassis and rise to the sensor (especially during a self-fl ushing purge cycle). Turning the power switch off and then back on again will restart the ice machine.Relay output indication:Each relay on the board has an indicator light associated with its output. For example, when the relay for the water feed solenoid is energized, the adjacent indicator light glows green.Time delay on start-up:ATTENTION!Unit has 15 minute start up delay when power is applied toprevent circuit overload. Depressing the reset button on thecontrol board will bypass this delay.Flushing logicFlush on fl y: For every one (1) hour of ice making time, the machine will open the drain valve for a duration of 60 seconds. While the drain valve is open, the machine will continue to make ice and the water feed valve will cycle to maintain water level.Off cycle: At the completion of off-cycle time delay, the machine checks for a cumulative one (1) hour of ice making time since the last off-cycle fl ush. If the cumulative ice making time exceeds one (1) hour, the machine will open the drain valve for 60 seconds to drain the evaporator in its entirety. It will then refi ll with water and begin making ice. Install the three bolts: partially tighten each bolt,alternating bolts until the rear bushing is fully seatedand properly aligned (Fig. 22).4. When fully seated, remove the bolts.5. Install the reservoir with three bolts (Fig. 23).6. Install the reservoir insulation, tube clamp, andsolenoid lever with three screws.7.Install solenoid.1. Remove and inspect O ring seal. Replace ifdamaged in any way (Fig. 25.1).2. Clean O ring groove. Lubricate O ring with petrol-geland reinstall (Fig. 25.2).3. Use cardboard disc to press new mating ring intomain housing (Fig. 26.1).4. Lube the shaft with liquid soap in the area shown(Fig. 26.2) and slip on seal and spring (Fig. 26.3).Note: Do not touch the sealing surfaces with barehands. Contact with bare skin will cause premature seal failure.5. Install auger (Fig. 26.4).6. Install main housing:§Slide main housing onto auger shaft (Fig. 27.1).§Install main housing insulation (Fig. 27.2).§Use an allen wrench to install 3/16" allen screws (3) (Fig. 27.3).7. Apply a coat of petrol-gel to the auger shaft.8. Install gear motor:§Install main housing insulation (Fig. 28.1).§Slide gear motor onto the auger shaft (Fig. 28.2).§Install two 1/2" bolts (Fig. 28.3).§Install 11/16" auger nuts and washer (Fig. 28.4). 9. Use 3/16" allen wrench to orient auger shaft withkeyway in the upward position (Fig. 29.1).10. Install key into keyway (Fig. 29.2).11. Install washer and fi nger tighten 11/16" jam nut.12. Tighten jam nut with adjustable wrench while usingallen wrench to prevent auger from turning.13. It is the responsibility of the technician to ensure that these requirements are met. Recharging ice machine to other than factory specifi cations will void the warranty.Refrigerant replacement requirements1. Non-contaminated refrigerant removed from any Follett refrigeration system can be recycled and returned tothe same system after completing repairs. Recycled refrigerant must be stored in a clean, approved storage container. If additional refrigerant is required, virgin or reclaimed refrigerant that meets ARI standard 700-88 must be used.2. In the event of system contamination (for example, a compressor burn out, refrigerant leak, presence ofnon-condensibles or moisture), the system must be repaired, evacuated and recharged using virgin orreclaimed refrigerant that meets ARI standard 700-88.3. Follett Corporation does not approve of recovered refrigerants. Improper refrigeration servicing procedureswill void the factory warranty.EvacuationEvacuate the system to a level of 500 microns. When the 500 micron level is reached, close all valves. Allow the system to sit for approximately 20 minutes. During this period the system pressure should not rise. If the system pressure rises and stabilizes there is moisture in the system and further evacuation is needed. Calculated amount per 24 hours should be checked against rated capacity for same ambient and watertemperatures in Ice Production Tables.“Bin full” detection systemThe Follett Horizon ice machine incorporates a unique “bin full” detection system that consists of the shuttle and actuator. The shuttle incorporates a fl ag and sensor. Referencing the fi gure below, the normal running position of the fl ag is down, out of the sensor. When the bin fi lls to the top and ice can no longer move through the tube, the machine will force the shuttle fl ag up into the sensor, shutting the machine off. The shuttle actuator, located above the ice bin allows the ice to curl up within it when the bin is full. The absolute latest in available technology is utilized with 32-bit (40-bit extended) floating point processors and high performance 24-bit Analog Converters. The high-bit DSP prevents noise and distortion induced by truncation errors of the commonly used 24-bit fixed-point devices. A complete set of parameters include I/O levels, delay, polarity, 6 bands of parametric EQ per channel, multiple crossover selections and full function limiters. Precise frequency control is achieved with its 1 Hz resolution. Inputs and outputs can be routed in multiple configurations to meet any requirement. The 480M can be controlled or configured in real time onthe front panel or with the intuitive PC GUI accessed via the RS-232 interface. Software upgrade for CPU and DSP via PC keeps the device current with newly developed algorithms and functions once available. LightCycler480荧光定量PCR仪操作规程1.每次反应最后一步都必须做降温,降至40度,维持至少30秒。

2.在操作过程中,切忌戴有粉末的乳胶手套接触光学透性封板膜。

3.试剂加完样,封膜完毕后,在板式离心机中1500×g(3000rpm)离心两分钟,以排除气泡。

4.先开电脑在开仪器,先关仪器再关电脑。

打开总电源,打开连接LightCycler480荧光定量PCR仪的电脑,并打开LightCycler480 Software软件,然后打开LightCycler480仪器后方的电源开关,此时仪器会在与电脑软件接通后进行自检,仪器正面右边两盏桔黄色的灯会闪烁。

自检完毕后,左边的灯会变绿,右边的灯仍为桔黄色。

这时按一下灯右边带双箭头的开关,仪器的载板器会自动伸出,将准备好的反应板放入载板器中(反应板的切角方向应与载板器的切角方向相应才能将反应板正确放入,切勿用裸手接触反应板的光学透性封板膜)。

再按一下带双箭头的开关,仪器的载板器会自动缩入仪器内,此时仪器能自动检测到反应板,原来呈桔黄色的灯会变绿,LightCycler480 Software中Start Run按钮也同时变为激活状态。

5.设定完实验程序并编辑完样本后,点击软件中Start Run按钮,运行实验。

6.软件界面上出现Run Complete….标识时,说明实验运行完毕,按一下仪器上带双箭头的按钮,使载板器伸出,取下反应板,再按一下带双箭头的按钮,使载板器缩回,关闭LightCycler480仪器后方的电源开关,在电脑上分析实验数据。

7.分析完实验数据后保存结果,打印报告,并关闭电脑,关闭总电源。

实验员在离开PCR扩增区时带走用完的反应板。

维护SOPLightCycler 480是一台免维护的仪器。

做任何维护工作前,须确认主机电源已被关闭。

预防性维护:1.检查LightCycler 480主机两侧与背后空间,确保无任何阻碍空气流通的物品,包括书本、纸张或其他任何物品;2.若主机表面和桌面有灰尘,用不起毛的软布蘸少量清水擦拭干净。

L-A1.0软件使用说明为了能够使您尽享本系统操作的方便快捷,请您详细阅读本说明:一、系统安装说明:打开您的电脑,启动操作系统之后,将本系统光盘放入光驱,打开我的电脑,打开光盘,会显示带有软件名称的文件夹,双击打开该文件夹,点击运行SETUP文件,您就可以根据中文提示安装本系统了。

注:如软件安装过程中出现如下提示,请直接点击取消按钮。

此操作不影响软件的正常安装使用。

若您使用的是通过USB口传输的巡检器,请您在安装完本软件系统后再安装USB驱动,详细的安装程序请参考USB驱动安装说明。

二、软件操作说明:(一)软件登录、注销1、登录双击巡更管理系统图标,软件起动后,会出现以下登录界面,选择权限,然后输入相对应的密码,系统管理员初始密码为333;操作员密码为222;浏览者密码为111。

只要输入正确,便可登录成功;登录界面如下图:登录成功后进入软件主窗口,提示选择您所使用的产品型号,如图:请在此选择用户名请在此输入密码2、注销操作员的注销,即更换操作员,使新的操作员用新的权限登录。

注销方法(如下图),选择菜单“操作员 用户注销”来实现。

点击此处即可(二)资源设置资源设置分五部分组成:人员钮设置、地点钮设置、事件设置、棒号设置、系统设置,每一部分的设置如下:1、系统设置:在第一次进入软件后,应首先对系统进行设置。

系统设置分为基本信息写入和权限用户密码管理。

如下图,在此可输入公司名称、选择的串口号,并可在此对权限密码进行修改。

修改完毕点击保存即可。

在此修改密码注:巡检器与电脑是用串口进行数据传输的,默认使用的串口号为COM1,巡检器与电脑是用USB 口传输的,默认使用的串口号为COM3(具体情况可到设备管理器中查询),在系统设置完毕后请重新登录巡更系统。

2、棒号设置:在使用巡检器之前需要将巡检器的棒号输入到软件中,以便识别。

点击“资源设置—>棒号设置”。

将巡检器与计算机连接好,并且将巡检器打开。

点击采集数据,会出现如下图所示:1140为该棒号码。

压着开关另一手将要锁上的刀具对准主轴刀上的定位键(drive key),刀具插上后,放开手动
退刀开关,刀具即被锁紧在主轴上。
注意:为保持主轴的精度,将刀具锁上主轴之前请先将主轴刀柄座斜度部分及将锁上的刀柄 擦拭清洁再锁上。

冲屑开关 押此按钮开关启动冲屑用切削水;再押一次停止冲屑切削水。

蜂鸣器解除 警报产生时,蜂鸣器会响,操作者可按此开关先将蜂鸣器暂时解除,再做问题排除的工作。

当 X/Y/Z 任何一轴有超过行程,EM 指示灯亮起。持续押此按钮开关并使用微调模式将进入紧急停 止开关内之轴反方向移开 100mm 时再放开,紧急停止区域的情况即可解除。
第4轴 手动移动第 4 轴,可依第 4 轴按钮开关上之方向标示¸移动第 4 轴之方向。轴回到机械原点时,灯会亮。

Discard ice andSani-Sponges (Fig. 11).ServiceIce machine operation (all models)Follett’s ice machine consists of five distinct functional systems covered in detail as follows:§Water system§Electrical control system §Mechanical assembly §Refrigeration system§Bin fullThe Horizon ice machine overviewThe Follett Horizon ice machine uses a horizontal, cylindrical evaporator to freeze water on its inner surface. The refrigeration cycle is continuous; there is no batch cycle. The evaporator is flooded with water and the level iscontrolled by sensors in a reservoir. A rotating auger (13 RPM) continuously scrapes ice from the inner wall of the evaporator. The auger moves harvested ice through the evaporator into an ice extrusion canal. The ice is forcedthrough a restrictive nozzle that squeezes out the water and creates the Chewblet. The continuous extrusion process pushes the Chewblets through a transport tube into a dispenser or bin.A solid state PC board controls and monitors the functionality of the ice machine. In addition to sequencing electrical components, the board monitors various operational parameters. A full complement of indicator lights allows visual status of the machine's operation. Additionally, the PC board controls the self-flushing feature of the ice machine. The evaporator water is periodically drained and replenished to remove minerals and sediment.A unique “bin full” detection system is incorporated in the Horizon ice machine. A switch located at the ice discharge port of the machine detects the position of the transport tube. When the bin fills up with ice, the transport tubemoves out of the normal running position, and the switch turns the ice maker off. A domed housing at the end of thetransport tube contains the ice extrusion loads during shut down.Harvest system diagramDIP Switch SettingsError faults:The Horizon PC board monitors various operating parameters including high pressure, auger gearmotor amperage limits, clogged drain, and low water alarm conditions. There are two types of errors namely “hard” or “soft”. A hard error is one that shuts the machine off and will not allow restart until the reset button is pressed. Even cycling power will not reset a hard error. A soft error can either be automatically reset should the condition rectify, or if power is cycled. Should an error occur, consult the troubleshooting guide in this manual or a Follett service technician.Soft errors:HI AMPS: The PC board monitors the amperage of the auger motor. Should the gear motor experience current draw above the allowable limit, the machine will shut down and the TIME DELAY and HI AMP will be illuminated. After the time delay the machine will restart and the TIME DELAY and HI AMP will clear.LO WA TER: During operation, the water level cycles between the normal low and normal high sensors. Should the water be shut off to a running machine, a soft error will occur. The error sequence is as follows: During operation,the water level falls to the normal low sensor, and when it does the water feed solenoid is energized. If water is not detected at the normal low sensor within 10 seconds, a soft error will occur. The machine will shut down, but the water feed solenoid will remain energized. Should water return, it will fill to the normal low sensor and the machine will resume normal operation. The error will clear automatically.HI PRESSURE: Should the refrigeration pressure rise above 425 psi, the machine will shut down and the TIME DELAY and HIGH PRESSURE will be illuminated. After the time delay, and if the pressure has fallen back below the reset point of 295 psi, the machine will restart and the TIME DELAY and HIGH PRESSURE will clear.Hard error:DRAIN CLOG: The drain clog sensor, located in the chassis, underneath the rear drain pan, will detect the presence of water just below the top edge of the pan. If water does not properly flow out of the drain pan it will overflow into the chassis and rise to the sensor (especially during a self-flushing purge cycle). Pressing the reset button will restart the ice machine.Relay output indication:Each relay on the board has an indicator light associated with its output. For example, when the relay for the water feed solenoid is energized, the adjacent indicator light glows green.Comp/Sol output:The output for the liquid line solenoid valve is labeled COMP/SOL.Flushing logicFlush on fly: For every one (1) hour of ice making time, the machine will open the drain valve for a duration of 60 seconds. While the drain valve is open, the machine will continue to make ice and the water feed valve will cycle to maintain water level.Off cycle: At the completion of off-cycle time delay, the machine checks for a cumulative one (1) hour of ice making time since the last off-cycle flush. If the cumulative ice making time exceeds one (1) hour, the machine will open the drain valve for 60 seconds to drain the evaporator in its entirety. It will then refill with water and begin making ice. If the ice making time is less than 1 hour, the machine will start and begin making ice without draining the evaporator.Mechanical System Fig. 125. 2.机器正面两个警示灯代表机器状态。

待警示灯 (右) 由闪烁橘色灯转变成稳定橘色灯,警示灯 (左) 由橘色转变成绿色 (约三分半钟),表示机器已经启动完成。

3.按压机器正面上唯一的按钮,放入已经封膜完成之 96 或 384 孔盘。

放置完成后,左右两警示灯都呈现绿色。

•启动软件1.开启计算机,并以正确的使用者名称与密码登入 Windows。

User name: OPERATORPassword: LC4802.开启 LightCycler 480 软件,并输入正确的使用者与密码。

二.软件设定-- 开启新实验•设定新实验1.软件开启后,会进入主画面。

点选“New Experiment”。

2.出现实验设定窗口。

任何时候需要回主画面的话,点选画面右边的按钮。

•设定实验❖若要套用设定好的实验步骤 (Template),请跳至 6.。

1.在实验设定画面上方,先选择适当的 Detection Forma t。

选项包含:另外可再点选“Customize” 勾选所需的滤片组合。

2.输入反应体积: 96 well plate 范围为 10-100 ul,而 384 well plate 范围为 5-20 ul。

3.设定实验步骤 (Program)•设定 program,包含有 Pre-incubation, Amplification (PCR), Melting curve (optional) 与 Cooling。

利用『+』与『-』增加或删除步骤。

•设定循环数目 (Cycle) 与分析模式。

LightCycler480荧光定量PCR仪操作规程1.每次反应最后一步都必须做降温,降至40度,维持至少30秒。

2.在操作过程中,切忌戴有粉末的乳胶手套接触光学透性封板膜。

3.试剂加完样,封膜完毕后,在板式离心机中1500×g(3000rpm)离心两分钟,以排除气泡。

4.先开电脑在开仪器,先关仪器再关电脑。

打开总电源,打开连接LightCycler480荧光定量PCR仪的电脑,并打开LightCycler480 Software软件,然后打开LightCycler480仪器后方的电源开关,此时仪器会在与电脑软件接通后进行自检,仪器正面右边两盏桔黄色的灯会闪烁。

自检完毕后,左边的灯会变绿,右边的灯仍为桔黄色。

这时按一下灯右边带双箭头的开关,仪器的载板器会自动伸出,将准备好的反应板放入载板器中(反应板的切角方向应与载板器的切角方向相应才能将反应板正确放入,切勿用裸手接触反应板的光学透性封板膜)。

再按一下带双箭头的开关,仪器的载板器会自动缩入仪器内,此时仪器能自动检测到反应板,原来呈桔黄色的灯会变绿,LightCycler480 Software中Start Run按钮也同时变为激活状态。

5.设定完实验程序并编辑完样本后,点击软件中Start Run按钮,运行实验。

6.软件界面上出现Run Complete….标识时,说明实验运行完毕,按一下仪器上带双箭头的按钮,使载板器伸出,取下反应板,再按一下带双箭头的按钮,使载板器缩回,关闭LightCycler480仪器后方的电源开关,在电脑上分析实验数据。

7.分析完实验数据后保存结果,打印报告,并关闭电脑,关闭总电源。

实验员在离开PCR扩增区时带走用完的反应板。

维护SOPLightCycler 480是一台免维护的仪器。

做任何维护工作前,须确认主机电源已被关闭。

预防性维护:1.检查LightCycler 480主机两侧与背后空间,确保无任何阻碍空气流通的物品,包括书本、纸张或其他任何物品;2.若主机表面和桌面有灰尘,用不起毛的软布蘸少量清水擦拭干净。

清洁性维护:若有需要,可用性质温和的商品化清洁剂或70%的乙醇溶液对LightCycler 480主机表面、热循环96孔模块和热盖进行清洁。

清洁96孔热循环模块:1. 用移液器移取125μl 70%乙醇溶液或异丙醇至所有孔内;2. 15分钟后,使用移液器对孔内溶液进行多次吹洗;3. 移除孔内的液体,并使96孔模块孔内自然风干;注意操作时不要让清洗液腐蚀热循环模块的涂层。

各图标的功能在不同状态下以颜色来区分是否具备。

软件操作一.打开软件打开电脑,Windows操作系统的用户名为operator,密码为LC480(区分大小写),电脑操作系统启动完毕,检查电脑右下角是否有图标。

如果没有,点击桌面“Exor4 for XDMS_R”快捷方式文件。

然后右键点击图标,左键点击“Show”一栏,如最后一句话显示“Exor 4 server is running”表示数据库加载成功。

如果显示“Exor4 failed to start”则表示数据库没有加载上,关闭该窗口,右键点击图标,选“Shutdown”一栏,退出数据库,重新点击桌面“Exor4 for XDMS_T”文件,加载数据库。

然后点击桌面“LightCycler480 Software”快捷方式文件,运行该软件。

输入用户名和密码。

用户名默认为admin(管理者权限),密码为LightCycler480(区分大小写),如用户已经建立新用户名,则可以按照新用户名登录。

二.用户管理为了实验结果的有效管理,可以建立不同级别的用户。

如: 标准用户(Standard User)、专业用户(Expert User)、管理者(Local Administrator)。

一般实验室建议建立专业用户(Expert User)用户名,然后以该用户名登录软件进行实验操作。

建立新用户操作:打开LightCycler480 操作软件,点击右侧按钮打开“Tools”工具栏,选择“User Access/Users and Groups”选项。

在右边窗口中点击“New User”键,在右边“Enter the user’s full name”一栏中填写用户的全名,在“”Enter the name the user wants to log in as”一栏填写用户登录名,在“Enter the user’s password”一栏中输入密码(密码须含有6个以上字母,1个以上数字,其中有至少一个字母大写),在“Confirm the password”一栏中重新输入一次密码,加以确认,在“Select the user’s role”一栏中选择用户级别,然后点击右下方“Close”按钮加以确认。

在“Tools”工具栏中的“System Settings”界面中可以设置各用户级的权限,以及密码的时效。

三.仪器自检每次开机时,仪器会自动进行初始化自检程序。

自检通过,仪器左侧指示灯变绿。

如放置了样本,仪器右侧指示灯变绿。

在自检时,不能放置反应板,否则自检无法通过。

四.建立实验程序打开LightCycler480 操作软件,成功登录后点击按钮,进入New Experiment/Run Protocol程序设置界面。

在“Detection Format”一栏中选择所使用的荧光检测技术,可选择各类探针和染料,如常见的SYBR Green I染料、Mono Color Hydrolysis Probe单色TaqMan水解探针、Multi Color Hydrolysis Probe多色TaqMan水解探针。

在进行多色荧光PCR实验时,须点击按钮选择用户所采用的荧光检测通道,点击“√”确认。

在一栏中填写反应体积。

在“Programs”界面中设置实验程序:在“Program Name”下方命名程序,“Cycles”一栏内填写该程序的循环数,在“Analysis Mode”一栏选择该程序的分析模式。

定量分析选择“Quantification”,溶解曲线分析选择“Melting Curves”,颜色补偿实验选择“Color Compensation”。

而后在“Temperature Targets”界面内设置所指定程序的内容,在“Target(℃)”处填写目标温度,“Acquisition Mode”处选择信号采集方式,“Hold”一栏填写该温度所持续的时间,“Ramp Rate(℃/s)”处填写升降温速率(注:如目标温度为50℃以下时,为了减少硬件的损耗,必须将降温速率调至1.5℃/s以下)。

如一个程序有多个步骤,则在Temperature Targets界面的左侧处点击,增加步骤;也可点击上下箭头调整各步骤的先后顺序。

如一个实验中有多个程序,也可在“Programs”界面左侧点击,增加程序,也可点击上下箭头调整各程序的先后顺序。

实验程度设置完成后,在下方“Overview”界面内会自动生成温度随时间变化的温控曲线,以及估计的运行时间,绿色点表示采集荧光信号的温度和时间点。

可以依照该图检查一下程度设置是否正确。

设置好的实验程序可以保存成模板文件,下次使用同样的实验程序时可以调用。

具体操作如下:点击按钮右侧的下拉箭头,选中“Save As Template”选项,系统默认将实验程序模板文件保存在所登录的用户名下“Templates”目录中,用户可将其存放在其下的子目录“Run Templates”下,填写该程序模板文件的名称,点击“√”按钮即完成保存。

调用模板程序时可点击“Apply Template”按钮,从目录树中选定所要调用的模板,点击“√”按钮确认。

五.样本设置1、样本亚组编辑:LightCycler480软件拥有一个独特的样本编辑管理功能——亚组编辑(Subset Editor),即将反应板上所有的样本根据用户需要分成亚组,对亚组进行独立地分析定量。

具体操作如下:在“New Experiment”界面中,点击左侧按钮,进入Subset设置界面,点击按钮新建亚组,并给其命名。

点击按钮可复制所选定的亚组,点击按钮可更改所选定亚组的名称,点击“Delete”按钮可删除所选定的亚组。

设置亚组:在Subsets界面中选定要设置的亚组名,在右边反应板孔位处选定该亚组所包含的孔位,使所选定的孔位中都出现深兰色凹陷的,点击下方按钮加以确认。

设置亚组可以在一块反应板上实现多个检测项目的实验,前提是这些检测项目的实验运行程序都一致(这在使用TaqMan水解探针时比较容易实现),由于各检测项目均有各自的质控对照,因而,设定亚组可以单独对各检测项目进行分析。

2.样本属性编辑点击New Experiment界面左侧按钮设置样本信息。

在“Workflow”一栏在选择实验方案:Abs Quant:绝对定量或定性分析;Rel Quant:相对定量;Scanning:基因扫描;Color Comp:颜色补偿;Tm:熔解温度检测;Melt Geno:熔解曲线法基因分型;Endpt Geno:终点法基因分型。
