miR ̄613在肝细胞癌中的表达变化及其对癌细胞增殖㊁凋亡及侵袭㊁迁移能力的影响

㊀㊀摘要:目的㊀观察miR ̄613在肝细胞癌(HCC)组织和细胞中的表达情况ꎬ并探讨miR ̄613对癌细胞增殖㊁凋亡及侵袭㊁迁移能力的影响ꎮ方法㊀选取60例HCC患者ꎬ手术切除肿瘤组织和癌旁组织ꎬ采用实时荧光定量PCR技术检测癌组织和癌旁组织中的miR ̄613ꎬ并分析miR ̄613与HCC患者临床病理特征的关系ꎮ采用实时荧光定量PCR技术检测人HCC细胞系MHCC97H㊁SMMC ̄7721和人正常肝细胞系L02中的miR ̄613ꎮ将miR ̄613模拟物和模拟物阴性对照转染入SMMC ̄7721细胞ꎬ设为miR ̄613mimics组和mimics ̄NC组ꎬ采用MTT实验检测细胞增殖能力ꎬ流式细胞术测算细胞凋亡率ꎬTranswell小室检测细胞迁移和侵袭能力ꎮ结果㊀miR ̄613在HCC组织的表达低于癌旁组织(P<0.05)ꎮTNM分期越晚ꎬmiR ̄613表达越低(P<0.05)ꎻEdmondson分级越高ꎬmiR ̄613表达越低(P<0.05)ꎮmiR ̄613在人HCC细胞系中的表达低于人正常肝细胞系(P<0.05)ꎮ与mimics ̄NC组相比ꎬmiR ̄613
mimics组miR ̄613表达增高(P<0.05)ꎬ细胞增殖活性降低(P<0.05)ꎬ凋亡率上升(P<0.05)ꎬ迁移和侵袭穿膜细胞数均减少(P均<0.05)ꎮ结论㊀miR ̄613在HCC中呈低表达ꎬ过表达miR ̄613能够抑制HCC细胞增殖㊁
迁移和侵袭ꎬ同时促进细胞凋亡ꎮ

SixtypatientswithHCCwereselectedꎬandthemiR ̄613expressioninthetumortissuesandadjacenttissuesweredetec ̄tedbyreal ̄timequantitativePCRꎬandtherelationshipbetweenmiR ̄613andclinicopathologicalcharacteristicsofHCCpa ̄tientswasanalyzed.TheexpressionofmiR ̄613inthehumanHCCcelllinesMHCC97HꎬSMMC ̄7721andhumannormallivercelllineL02wasdetectedbyreal ̄timefluorescentquantitativePCR.ThemiR ̄613mimicsandmimicsnegativecontrolweretransfectedintoSMMC ̄7721cellsꎬwhichweretakenasthemiR ̄613mimicsgroupandmimics ̄NCgroup.MTTassaywasusedtodetectcellproliferationꎬflowcytometrywasusedtomeasureapoptosisrateꎬandTranswellchamberwasusedtodetectthecellmigrationandinvasion.Results㊀
TheexpressionofmiR ̄613inHCCtissueswaslowerthanthatintheadja ̄
centtissues(P<0.05).ThelatertheTNMstageꎬthelowerthemiR ̄613expressionwouldbe(P<0.05)ꎻthehighertheEdmondsongradeꎬthelowerthemiR ̄613expressionwouldbe(P<0.05).TheexpressionofmiR ̄613inthehumanHCC
celllineswaslowerthanthatinhumannormalhepatocyteline(P<0.05).Comparedwithmimics ̄NCgroupꎬtheexpres ̄sionofmiR ̄613increased(P<0.05)ꎬcellproliferationactivitydecreased(P<0.05)ꎬapoptosisrateincreased(P<0.05)ꎬandmigrationandinvasionoftransmembranecellsdecreasedinthemiR ̄613mimicsgroup(allP<0.05).Con ̄clusion㊀
TheexpressionofmiR ̄613islowexpressedinHCCꎬover ̄expressionofmiR ̄613caninhibittheproliferationꎬ
migrationandinvasionꎬandpromoteapoptosisofHCCcells.
