Chk family checkpoint kinases include Chk1 and Chk2. They are activated in response to DNA damage and phosphorylate CDC25A, CDC25C protein phosphatases, which delay cell cycle progression. Therefore, Chk activation initiates cell cycle checkpoint, causes cell cycle arrest, and allows DNA repair.AZD7762 is a potent selective inhibitor of Chk1. It binds to the ATP binding pocket and compete ATP binding in a reversible manner. AZD7762 inhibits Chk1 phosphorylation of CDC25C peptide with an IC50 of 5 nM. The Ki is 3.6 nM. It is equally potent against Chk2 but less potent against CAM, Yes, Fyn, Lyn, Hck and Lck. [1]AZD7762 prevents cell cycle arrest and DNA repair in DNA damaged tumor cells, causing tumor cell apoptosis. 近年来,药品中残存的有机溶剂的毒性和致癌作用日益引起各方面的重视[4-8]。
由于在那格列奈原药合成过程中使用了有机溶剂二氯甲烷和吡啶,因此在成品中可能会有一定量的残留。
按《中华人民共和国药典2005年版》(附录Ⅷ)规定,该2种有机溶剂均有残留限量,因此对那格列奈原药中二氯甲烷和吡啶的残留量检测具有一定意义。

1. 1-[(4-氟苯基)甲基]-苯并咪唑-2-(3H)-酮的制备
在反应瓶中加入2-羟基苯并咪唑5.0g(37.3mmol)和NaH 1.6g(53mmol)(NaH含量大约为80%,浸入矿物油中) 的DMF 100ml的悬浮液.加毕.在60ºC.(最好有N2保护)搅拌反应1h.再加入4-氟苄基氯(FBC)5.4g(37mmol),加热 ( 6 0 ºC ) 搅 拌 反 应 5 . 5 h . 冷 却 至 室 温 后 加 入 冰 水 7 0 0 m l , 用 二 氯 甲 烷 ( 5 0 0 m l × 2 ) 提 取 . 有 机 层 用 食 盐 水 洗 . 无 水 N a 2 S O 4 干燥.过滤.滤液减压浓缩.剩余物用石油醚析晶.得1-[(4-氟苯基)甲基]-苯并咪唑-2-(3H)-酮固体8.0g,为无色 结 晶 m p 1 7 8 ~ 1 7 9 ºC , 收 率 8 8 % .

阿司咪唑中毒的治疗要点为: 1.大量摄入者予洗胃,后灌服活性炭和导泻。 2.对心肌抑制和Q-T间期延长者予5%碳酸氢钠250ml静注可能有效。 3.对症、支持治疗。

方法 以廉价的乙酰乙酸乙酯为起始原料,经亚硝化、溴化、环合、甲基化一锅反应,水解后得氨噻肟酸,与N-羟基邻苯二甲酰亚胺反应合成邻苯二甲酰亚胺氨噻肟活性酯,然后与7-氨基-3-去甲基-3-头孢烷酸(72ANCA)缩合制得头孢唑肟酸,再与成盐剂反应得头孢唑肟。
结果 该合成工艺切实可行,其产率达到93.5%。
结论 该合成工艺操作简单,原料易得,总收率高,副反应少,同时降低了成本,具有良好的应用前景。
