【摘要】目的观察大黄虫丸对单侧输尿管梗阻(UUO)致大鼠肾间质纤维化血液流变学及微循环变化的影响,从"络病学说"探讨其抗肾间质纤维化的中医病机.方法采用单侧输尿管结扎法复制单侧输尿管梗阻致大鼠肾间质纤维化模型,将其随机均分为模型组和大黄虫丸组,并设假手术组;观察大黄虫丸对该模型动物全血黏度150/s 切变率、30/s切变率、5/s切变率、1/s切变率、红细胞压积、舌下及耳廓脉络变化的影响.结果大黄虫丸能显著降低肾间质纤维化大鼠的全血黏度150/s切变率、30/s切变率、5/s切变率、1/s切变率、红细胞压积、透明质酸(HA)、层粘连蛋白(LN)、Ⅲ型前胶原(PCⅢ)、Ⅳ型胶原(CⅣ)、血清肌酐(SCr)、尿素氮(BUN)升高的指数,改善纤维化大鼠舌下、耳廓脉络的循环形态.结论大黄虫丸对肾间质纤维化大鼠血液流变学和微循环变化的改善,提示"活血通络"可能是其抗肾间质纤维化的主要病机之一.%Objective To observe Effects of Dahuangzhechongwan on Hemorheology and microcirculation in renal interstitial fibrosis rat caused by unilateral ureteral obstruction (UUO) and attempt to use "luo bing xue shuo" explain the mechanism of Dahuangzhechongwan' s resistive effects on renal interstitial fibrosis. Methods Rat fibrosis models were established by unilateral ureteral obstruction, then rats were randomly assigned into the renal interstitial fibrosis model group , Dahuangzhechongwan group and the sham operation group . Hematocrit and Blood viscosity at shear
rate in 150/s, 30/s, 5/s, 1/s were detected respectively among those groups; At the same time, use microcirculation microscope observe the thread of hypoglottis and auricle in rats. Results Dahuangzhechongwan could reduce the indexes of Hematocrit, Blood viscosity at different shear rate, Hyaluronic Acid (HA), Laminin (LJN), Procolagen Type HI (PCM), Collagen TypeIV (CIV), serum creatinine (SCr) and blood urea nitrogen (BUN) in renal interstitial fibrosis, and improve the microcirculation of hypoglottis and auricle in model rats. Conclusion The improvement of blood rheology and microcirculation suggest the mechanism of Dahuangzhechongwan' s resistive effects on renal interstitial fibrosis lie in "promoting blood circulation to remove meridian obstruction".
