Hospital, China Academy of Chinese Medical Sciences, Beijing 100091, China Abstract: Objective To establish an HPLC method for the simultaneous determination of nine hydrophilic compounds and three lipophilic compounds in Salviae Miltiorrhizae Radix et Rhizoma; To discuss the effects of solvent polarity difference on the dissolution of active components in Salviae Miltiorrhizae Radix et Rhizoma represented by different concentrations of ethanol. Methods The HPLC separation was performed on an Agilent Extend-C18 column (4.6 mm × 250 mm, 5 μm), eluted with the mobile phase of acetonitrile-0.1% phosphoric acid solution. The flow rate was 0.8 mL/min; the column temperature was 25 ℃; the detection wavelength was 280 nm; the injection volume was 10 μL. The contents of 12 components in Salviae Miltiorrhizae Radix et Rhizoma extracted from different concentrations of ethanol (0%, 20%, 40%, 60%, 80%, 95%) were investigated. Results The complete separation was obtained within 95 minutes for the 12 components. There was a good linear relationship between concentration and peak area in the linear range (r>0.998). The average recoveries were 95.28%–104.97% and RSD were less than 4.58%. Different concentrations of ethanol had different influence on the contents of 12 components. The total amount of water-soluble components increased first and then decreased with the decrease of the polarity of the solvent, and the fat-soluble component continued to rise. Conclusion The method has good specificity, high sensitivity and accurate quantitative, which can be used for simultaneous determination of nine hydrophilic compounds and three lipophilic compounds in Salviae Miltiorrhizae Radix et Rhizoma, and different concentrations of ethanol have great influence on the dissolution of the 12 components.
