格锐思生物-甘油含量测定试剂盒说明书

甘油(Glycerol)含量测定试剂盒说明书(货号:G0912W微板法96样)一、产品简介:甘油储存于脂肪细胞中是甘油三酯代谢的最终产物之一。

在生产、生活中甘油可用作溶剂,润滑剂,药剂和甜味剂。

甘油被甘油激酶(GK)的催化生成甘油-1-磷酸(G-1-P)。

G-1-P被甘油磷酸氧化酶(GPO)氧化生成过氧化氢(H2O2),H2O2与4-氨基氨替吡啉等反应生成红色醌类化合物,其在510nm处有特征吸收峰,通过检测510nm处吸光值即可得出甘油含量。

二、试剂盒的组分与配制:试剂名称规格保存要求备注提取液液体100mL×1瓶4℃保存试剂一液体μL×1支-20℃保存用前甩几下使试剂落入底部,再加4.2mL蒸馏水,充分震荡混匀溶解。

试剂二液体9mL×1瓶4℃保存试剂三液体6mL×1瓶4℃保存标准品液体mL×1支4℃保存用前用水稀释10倍即成4mM甘油标准品待检测液。

三、所需的仪器和用品:酶标仪、96孔板、可调式移液枪、离心机、研钵、蒸馏水。

四、甘油含量测定:建议正式实验前选取2个样本做预测定,了解本批样品情况,熟悉实验流程,避免实验样本和试剂浪费!1、样本制备:①组织样本:称取约0.1g组织样本加入研钵中,加入1mL提取液,在冰上进行匀浆,12000rpm,4℃或室温离心10min,取上清液待测。

【注】:若增加样本量,可按照组织质量(g):提取液(mL)为1:5~10的比例进行提取。

②细菌/细胞样本:先收集细菌或细胞到离心管内,离心后弃上清;取约500万细菌或细胞加入1mL 提取液,超声波破碎细菌或细胞(冰浴,功率200W,超声3s,间隔10s,重复30次);12000rpm4℃离心10min,取上清,置冰上待测。

【注】:若增加样本量,可按照细菌/细胞数量(104):提取液(mL)为500~1000:1的比例进行提取。

③液体样本:澄清的液体样本直接测定,若浑浊则离心后取上清检测。

都在测温度,但到底什么是温度?|贤说八道如今,全国众多公共场所都在不间断地测量过往人群的体温。

然而,什么是温度?一个体系有没有温度?温度计到底测量了什么?这些问题即便在专业的物理教科书和杂志上也是语焉不详。

-[唐]罗隐(1)温度测量的历史是科学发展史的组成部分……-Thomas D. McGee提要:感知冷热是生命必备的能力,因此冷热概念的出现远远早于热力学这门学科,也就难免纠缠不清。

热、冷、火在英文物理文献中都有多重表述,而温度也是非常不易正确理解的物理学基本概念之一。

1世事总关炎凉茫茫宇宙中一粒微不足道的尘埃——地球——上诞生了生命。

注意到地球的平均温度约为15°C,生命的物质基础之一是水而水的凝固点为0°C,以及生命作为一个远离平衡态的耗散体系需要不断获得能量而地球的能源是来自太阳的辐照等几个事实,就可以多少理解生命所选择的温度窗口(必须处于环境温区的高端)以及对冷暖的敏感(2)。

可以说一切生命最重要的感觉能力是对冷暖的感知(对许多高级动物来说,视觉或者听觉是备选项),过去中国人甚至将知冷知热看作是一个人作为好的配偶所必备的品格。

在中文语境里,温和、温暖、温柔等贴近我们体温的词汇都让人感到非常温馨,在其它文化中大概也应如此,毕竟“环球同此凉热”(3)。

冷热的概念在人体发育的早期就应该建立起来了。

给婴幼儿喂奶喂饭,一开始由大人掌握冷热,而后就要教孩子自己明白冷热。

舒适的温度,应该是体温附近不大的范围,它首先是生理的需要,其后慢慢成了心理的需求。

我们一个人一生中最需要理解的现象也许就是世态炎凉,人情冷暖!人类自愚昧中走过来,在其科学努力中自然会将很大的精力放在理解冷暖现象上。

于是,我们发展了热学、热力学,而这其中要理解和量化的一个重要概念是冷热程度,即温度。

2混乱的字面冷热是我们身体的感觉,因此我们关于冷热现象相关的词汇一定比热力学这门科学出现得早,也就容易想象存在某些概念上的夹杂不清。

校准装置采用多层结构、真空绝热、增加热阻等方法降低热量/冷量损失,设计冷量分配器减小中间层与内层的温差降低热辐射,配置辅助冷却套管降低引出电缆温度梯度,并通过在安装铜座和校准腔均匀布置标准温度计和加热器,实现温度精确控制,系统保证校准腔温度均匀性和稳定性。

校准装置性能测试实验结果表明:在45路温度传感器并行校准条件下,校准温度范围可达77 ~323 K ,有效温度场的温度均匀性和温度波动性均优于33 mK ,能够满足低温风洞温度传感器现场校准需求。

测定细胞内GSH 和GSSG含量以及GSH/GSSG比值,能够很好地反映细胞所处的氧化还原状态。

The particular design, based on a helium cooling loop and an electricaltemperature controller, gives good dynamic performances.This paper describes the experimental set-up and the data acquisition system.Results of experimental runs are also presented along with the estimated globalaccuracy for the calibration.1. INTRODUCTIONThe LHC superconducting magnets will immersed in pressurised superfluid helium at a temperature of 1.8 K. About 10,000 cryogenic thermometers will be installed along the 27 km LHC ring for its operation and control. A cryogenic thermometer calibration facility is now in operation at CERN [1]. It can handle 60 thermometers at a time which are mounted on an isothermal copper block placed in a vacuum chamber. The calibration is performed by comparison to 4 reference thermometers (working standards). The calibration cycle is performed using a controlled cryogenic process: cooling down by using gaseous helium as heat exchange gas and warming up with the help of electrical heaters. With this system any temperature between 1.6 K and 300 K can be obtained with an accuracy better than the one shown in Table 1.In this paper we present the results obtained with a small-scale model of a new calibration block. The main aim of this study was to test new ideas to improve the first calibration block, in order to increase thecalibration turnover by introducing a better control of the set points, together with a better temperature stability during the calibration procedure. This paper describes the main physical and metrological characteristics of this apparatus.2. THE CALIBRATION SYSTEM2.1 PrincipleCalibration of thermometers is made in vacuum, by measuring the thermometer resistance for a given set of temperature points. The reference temperature is given by two rhodium-iron working standards placed in the calibration copper block. These working standards thermometers have been calibrated at CERN using secondary standards [2]. The secondary standards are traceable in the range 1.6K to 30K to the Italian metrological institute (IMGC, Torino-Italy).For the LHC a thermometer block for working under vacuum conditions with built-in heat interception has been developed [3]. Apiezon grease is used to improve thermal contact. It is not necessary to solder the thermometers’ sensing wires thanks to special connectors made of RF spring contacts.The working standards are introduced inside two cylindrical holes drilled in the calibration block.The regulation copper block includes a helium cooling loop and an electrical heater whose power is adjusted by a PID temperature controller. It is possible to achieve any temperature between 1.8K and 300K. The use of a cryogenic valve and active pumping allows to get temperatures below 4.2K in the regulation block without cooling down the liquid helium bath. A brass thermal impedance has been placed in between the calibration and regulation blocks in order to reduce temperature perturbations of the former block. These perturbations observed at low temperature are due to helium flow instabilities in the heat exchanger.A copper shield thermally anchored to the regulation copper block prevents radiative thermal flux on the calibration block.In order to achieve the best operating conditions, all wires are thermally anchored to the thermalisation block before being routed to the calibration isothermal copper block. All thermometers are measured in a four wire configuration.2.2. InstrumentationA Labview program running on a PC is used for control and acquisition purposes. The data acquisition system is PC controlled through a IEEE-488 bus and measures successively each resistance using a scanner, a programmable current source and a milli-voltmeter. The scanning cycle (including the working standards and diagnostic thermometers) is performed in about 2 minutes (due to current stabilisation time and its polarity reversal). The estimate global accuracy of measures is 1.4 10-4 on the resistance which is equivalent to a 1.7 10-3 precision on the temperature for Rhodium Iron working standards for temperatures below 4 K.The same program has a Programmable Logic Controller (PLC) interface and controls the cryogenic valve. It also sends temperature set-points to the Proportional Integral regulator.2.3. Calibration sequence descriptionThe temperature calibration set-points are memorised on the acquisition PC. The program sets the valve position and the temperature controller parameters in order to obtain the required temperature. When the temperature is approached, the valve is adjusted in order to cool slightly the calibration block and a temperature controlled heater then precisely maintains a set-point. The ideal parameters for the PID controller strongly depend on the temperature set-point and are updated accordingly. The calibration scanning cycle starts when the temperature is stable within less than 1 mK/minute.3 EXPERIMENTAL RESULTS3.1 Dynamic performanceThe heat exchanger proved to be very efficient and a maximal cooling power of 100W was obtained for high temperatures using almost all the specific heat of the 2 g/min helium flow rate. Temperature cool down for the 3.5 kg copper mass from room temperature to 4.2K takes about four hours (average decrease of 1.25 K/min) without stabilisation time. The electrical heater allows to dissipate a maximum of 50W and the warm-up time is almost 8 hours. These times are essentially limited by the specific heat capacity of the calibration insert.Once the temperature set-point is reached in the calibration block, the settling time to wait before getting temperature homogeneity and stability in it is about 1h for high temperatures, about ½ hour at 50 K, and less than 15 minutes for T < 10 K.Figure 2 shows a part of a calibration sequence where 15 calibration points were collected within a three hour time period in the 1.8 K to 10 K temperature range.The full calibration time for a 60 calibration points in the whole temperature range, including two cooling down and warming up cycles, would be of two days, if running over the 24 hours.01234567891011T e m p e r a t u r e (K )Figure 2 Working standard temperature during a calibration sequence3.2 Homogeneity of the calibration blockTwo rhodium-iron working standards are mounted symmetrically in the calibration block in order to confirm the good isothermal conditions of the copper block. Figure 3 shows that both thermometers are very well correlated (better than 0.002 K) in the 1.8 K to 300 K temperature range. The difference is within the precision range of their calibration.-3-2-101231101001000Temperature (K)D T (m K )Figure 3 Temperature difference between the two working standards during a calibration run4 CONCLUSIONA calibration insert incorporating a liquid helium heat exchanger has been designed and tested for the calibration in vacuum of a small number of thermometers.The results give us confidence for building at IPN-Orsay a larger calibration facility of similar design,that should enable the simultaneous calibration of 80 thermometers.
