现行物理化学教材中重点阐述了仅含有体积功的热力学基本方程,而对涉及非体积功的热力学基本方程介绍很少。

对材料类专业的学生来说,学习和研究材料热力学,经常会涉及到非体积功。

因此,学习涉及非体积功的热力学基本方程非常必要。

本文重点讨论了涉及非体积功的表面系统、弹性杆、电、磁介质中的热力学基本方程的推导和应用,并总结了处理这类问题的方法。

为了提高储能系统效率,本工作提出了一种耦合光热发电储热-有机朗肯循环的先进绝热压缩空气储能系统(AA-CAES+CSP+ORC)。

该系统中光热发电储热用来解决先进绝热压缩空气储能系统压缩热有限的问题,而有机朗肯循环发电系统中的中低温余热发电来进一步提升储能效率。

本工作首先在Aspen Plus软件上搭建了该耦合系统的热力学仿真模型,随后本工作研究并对比两种聚光太阳能储热介质对系统性能的影响,研究结果表明,导热油和太阳盐相比,使用太阳盐为聚光太阳能储热介质的系统性能更好,储能效率达到了115.9%,往返效率达到了68.2%,㶲效率达到了76.8%,储电折合转化系数达到了92.8%,储能密度达到了5.53 kWh/m3。

此外,本研究还发现低环境温度、高空气汽轮机入口温度及高空气汽轮机入口压力有利于系统储能性能的提高。

CHAPTER1INTRODUCTION1.1What is thermodynamics?Thermodynamics is the science which has evolved from the original investiga-tions in the19th century into the nature of“heat.”At the time,the leading theory of heat was that it was a type offluid,which couldflow from a hot body to a colder one when they were brought into contact.We now know that what was then called“heat”is not afluid,but is actually a form of energy–it is the energy associated with the continual,random motion of the atoms which compose macroscopic matter,which we can’t see directly.This type of energy,which we will call thermal energy,can be converted (at least in part)to other forms which we can perceive directly(for example, kinetic,gravitational,or electrical energy),and which can be used to do useful things such as propel an automobile or a747.The principles of thermodynamics govern the conversion of thermal energy to other,more useful forms.For example,an automobile engine can be though of as a device whichfirst converts chemical energy stored in fuel and oxygen molecules into thermal en-ergy by combustion,and then extracts part of that thermal energy to perform the work necessary to propel the car forward,overcoming friction.Thermody-namics is critical to all steps in this process(including determining the level of pollutants emitted),and a careful thermodynamic analysis is required for the design of fuel-efficient,low-polluting automobile engines.In general,thermody-namics plays a vital role in the design of any engine or power-generating plant, and therefore a good grounding in thermodynamics is required for much work in engineering.If thermodynamics only governed the behavior of engines,it would probably be the most economically important of all sciences,but it is much more than that.Since the chemical and physical state of matter depends strongly on how much thermal energy it contains,thermodynamic principles play a central role in any description of the properties of matter.For example,thermodynamics allows us to understand why matter appears in different phases(solid,liquid, or gaseous),and under what conditions one phase will transform to another.1CHAPTER1.INTRODUCTION2The composition of a chemically-reacting mixture which is given enough time to come to“equilibrium”is also fully determined by thermodynamic principles (even though thermodynamics alone can’t tell us how fast it will get there).For these reasons,thermodynamics lies at the heart of materials science,chemistry, and biology.Thermodynamics in its original form(now known as classical thermodynam-ics)is a theory which is based on a set of postulates about how macroscopic matter behaves.This theory was developed in the19th century,before the atomic nature of matter was accepted,and it makes no reference to atoms.The postulates(the most important of which are energy conservation and the impos-sibility of complete conversion of heat to useful work)can’t be derived within the context of classical,macroscopic physics,but if one accepts them,a very powerful theory results,with predictions fully in agreement with experiment.When at the end of the19th century itfinally became clear that matter was composed of atoms,the physicist Ludwig Boltzmann showed that the postu-lates of classical thermodynamics emerged naturally from consideration of the microscopic atomic motion.The key was to give up trying to track the atoms in-dividually and instead take a statistical,probabilistic approach,averaging over the behavior of a large number of atoms.Thus,the very successful postulates of classical thermodynamics were given afirm physical foundation.The science of statistical mechanics begun by Boltzmann encompasses everything in classical thermodynamics,but can do more also.When combined with quantum me-chanics in the20th century,it became possible to explain essentially all observed properties of macroscopic matter in terms of atomic-level physics,including es-oteric states of matter found in neutron stars,superfluids,superconductors,etc. Statistical physics is also currently making important contributions in biology, for example helping to unravel some of the complexities of how proteins fold.Even though statistical mechanics(or statistical thermodynamics)is in a sense“more fundamental”than classical thermodynamics,to analyze practical problems we usually take the macroscopic approach.For example,to carry out a thermodynamic analysis of an aircraft engine,its more convenient to think of the gas passing through the engine as a continuumfluid with some specified properties rather than to consider it to be a collection of molecules.But we do use statistical thermodynamics even here to calculate what the appropriate property values(such as the heat capacity)of the gas should be.CHAPTER1.INTRODUCTION3 1.2Energy and EntropyThe two central concepts of thermodynamics are energy and entropy.Most other concepts we use in thermodynamics,for example temperature and pres-sure,may actually be defined in terms of energy and entropy.Both energy and entropy are properties of physical systems,but they have very different characteristics.Energy is conserved:it can neither be produced nor destroyed, although it is possible to change its form or move it around.Entropy has a different character:it can’t be destroyed,but it’s easy to produce more entropy (and almost everything that happens actually does).Like energy,entropy too can appear in different forms and be moved around.A clear understanding of these two properties and the transformations they undergo in physical processes is the key to mastering thermodynamics and learn-ing to use it confidently to solve practical problems.Much of this book is focused on developing a clear picture of energy and entropy,explaining their origins in the microscopic behavior of matter,and developing effective methods to analyze complicated practical processes1by carefully tracking what happens to energy and entropy.1.3Some TerminologyMostfields have their own specialized terminology,and thermodynamics is cer-tainly no exception.A few important terms are introduced here,so we can begin using them in the next chapter.1.3.1System and EnvironmentIn thermodynamics,like in most other areas of physics,we focus attention on only a small part of the world at a time.We call whatever object(s)or region(s) of space we are studying the system.Everything else surrounding the system (in principle including the entire universe)is the environment.The boundary between the system and the environment is,logically,the system boundary. a r X iv:c ond-ma t/9912402v122Dec1999Thermodynamics of Heat Shock Response Kristine Bourke Arnvig a ,Steen Pedersen a and Kim Sneppen b a Institute of Molecular Biology,Øster Farimagsgade 2A,1353Copenhagen K,Denmark.b Nordita,Blegdamsvej 17,2100Copenhagen Ø,Denmark July 26,1999Abstract:Production of heat shock proteins are induced when a living cell is ex-posed to a rise in temperature.The heat shock response of protein DnaK synthesis in E.coli for temperature shifts T →T +∆T and T →T −∆T is measured as function of the initial temperature T .We observe a reversed heat shock at low T .The magnitude of the shock increases when one increase the distance to the temperature T 0≈23o ,thereby mimicking the non monotous stability of proteins at low temperature.This suggest that stability related to hot as well as cold unfolding of proteins is directly implemented in the biological control of protein folding.PACS numbers:5.65.+b,82.60-s,87.,87.10.+e,87.14.Ee,87.15.-v,87.17.-d Chaperones direct protein folding in the living cell by binding to unfoldedor misfolded proteins.The expression level of many of these catalysts of pro-tein folding change in response to environmental changes.In particular,when a living cell is exposed to a temperature shock the production of these proteins are transiently increased.The response is seen for all organisms[1],and in fact involves related proteins across all biological kingdoms.The heat shock response (HS)in E.coli involves about 40genes that are widely dispersed over its genome [2,3].For E.coli the response is activated through the σ32protein.The σ32binds to RNA polymerase (RNAp)where it dis-places the σ70subunit and thereby changes RNAp’s affinity to a number of promoters in the E.coli genome.This induce production of the heat shock1proteins.Thus if the gene forσ32is removed from the E.coli genome the HS is suppressed[2,4]and also the cell cannot grow above20o C.The HS is fast.In some cases it can be detected by a changed synthesis rate of e.g.the chaperone protein DnaK already about a minute after the temperature shift.Given that the DnaK protein in itself takes about45 seconds to synthesize the observed fast change in DnaK production must be very close to the physical mechanism that trigger the response.In fact we will argue for a mechanism which does not demand additional synthesis ofσ32in spite of the fact that DnaK is only expressed from aσ32promoter,and thus postulate that changed synthesis ofσ32only plays a role in the later stages of the HS.To quantify the physical mechanism we measure the dependence of HS with initial temperature andfind that the magnitude of the shock is inversely proportional to the folding stability of a typical globular protein.The present work measure the expression of protein DnaK.Steady state levels at various temperature and growth conditions can be found in[5,6]. The steady state number of DnaK in an E.coli cell varies from approx.4000at T=13.5to approx.6000at37o C,thereby remaining roughly proportional to number of ribosomes.DnaK is a chaperone,and have a high affinity for hydrophobic residues[7],as these signal a possible misfold(for folded proteins the hydrophobic residues are typically in the interior).σ32control the expression of DnaK,theσ32must bind to the RNAp before this bind to the promoter for DnaK.One expect at most a few hundredσ32in the cell, a number which is dynamical adjustable because the in vivo half life ofσ32 is short(in steady state it is0.7minutes at42o C and15minutes at22o C [8,10]).The life timeσ32is known to increase transiently under the HS.The measurement was on E.coli K12strain grown on A+B medium[12] with a3H labelled amino acid added.After the temperature shift we ex-tracted0.6ml samples of the culture at subsequent times.Each sample was exposed to radioactive labelled methionine for30seconds,after which non radioactive methionine was added in huge excess(105fold).Methionine is an amino acid that the bacteria absorb very rapidly,and then use in protein synthesis.Protein DnaK was separated by2-dim gel electrophoresis as de-scribed by O’Farrell[11],and the amount of synthesis during the30seconds of labeled methionine exposure was counted by radioactive activity and nor-malizedfirst with respect to3H count and then with respect to total protein production.This result in an overall accuracy of about10%.The result is2a count of the differential rate of DnaK production(i.e.the fraction DnaK constitute of total protein synthesis relative to the same fraction before the temperature shift[12])as function of time after the temperature shift.For the shift T→T+∆T at time t=0we thus record:r(T,t)=Rate of DnaK production at time tT1−T2=0.2·K−1(i.e.α=0.03K−2).In order to interpret this result wefirst assume that the production rate of DnaK is controlled by two factors,a slowly varying factor C that depends on composition of some other molecules in the cell,and an instantaneous chemical reaction constant K.Thus at time t after a shift in temperature the production of DnaK in the cell is:d[DnaK]To lowest approximation,where we even ignore feedback from changed DnaK in the cell until DnaK production rate have reached its peak value:K(T+∆T)R=∆T(5)dTUsing the linear approximation in Fig.2αln(K)≈const+(T−T0)2(7)2Thus∆G has a maximum at T=T0=23o.To interpret the fact that HS is connected to a∆G that have a maximum at T=T0≈23o we note that many proteins exhibit a maximum stability at T between10o C and30o C[13,14].Thus∆G=G(folded)−G(unfolded) connected to the folded state of a protein is at a minimum at T0.The corre-sponding maximum of stability is in effect the result of a complicated balance between destabilization from entropy of polymer degrees of freedom at high T,and destabilization due to decreased entropic contribution to hydrophobic stabilization of proteins at low T[14,15].One should expect a similar be-haviour also for some parts of a protein[15],and thus expect a max binding for hydrophobic protein-protein associations around T0.Quantitatively the size of the∆G change inferred form the measured value ofα=0.03K−2 correspond to a changed G of about20→30kT(about15Kcal/mol),for a temperature shift of about40-50o C.This matches the change observed for typical single domain proteins[14].Thus the HS is associated to a∆G change equivalent to the destabilization of a typical protein.The above picture still leave us with the puzzle that protein binding and folding stability is at a maximum around T0,whereas the effective∆G we4observe have a minimum there.This can only be reconciled if the interaction we consider is inhibitory.An inhibitory binding that controls the feedback is indeed possible[3].To summarize our understanding we infigure3display the molecular network that we believe is controlling the transient heat shock levels of DnaK in the cell.The key inhibitory control mechanism is the association of DnaK toσ32.DnaK binds to unfolded protein residues[7],and the amount of DnaK·σ32association thereby monitor cellular consequences of a shift in temperature.Impacts of mutants:We have measured the heat shock in a strain where theσ32gene is located on a high copy number plasmid.In this strain where the synthesis rate forσ32may approach that of DnaK wefind a HS that was smaller and also remained positive down to temperature jumps from T well below T0=23.According tofig.3this reflects a situation where bothσ32and DnaK are increased.With increased DnaK level,one may have a situation where DnaK exceeds the amount of unfolded proteins,and free DnaK concentration thus becomes nearly independent of the overall state of proteins in the cell.Also the huge increase inσ32supply may decrease the possibility for the sink to act effectively.Thereby other effects as e.g. the temperature dependence of the bindingσ32·RNAp versus the binding σ70·RNAp(i.e.K32/K70fromfigure3)may govern a response that otherwise would be masked by a strongly varying inhibition fromσ32binding to DnaK.The reaction network in Figure3allow a more careful analysis of the production rate of DnaK:d[DnaK](8)1+g[DnaK]where theσ32changes when bound to DnaK due to degradation by proteases (the“sink”infigure3):d[σ32](9)1+(g[DnaK])−1 Here g=exp(−∆G/T)is an effective reaction constant.In the approxima-tion where we ignore freeσ32,freeσ70and the fraction of DnaK bound by5σ32then:g= K70[σ70]1+K DU[U f] (10) Thefirst term expresses theσ’s competition for RNAp binding whereas the second term expresses the DnaK controlled response.[U f],which denote unfolded proteins that are not bound to DnaK,decreases with increasing [DnaK].When moving away from T0,i.e.lowering g by increasing[U f],the rate for DnaK production increases.For an approximately unchanged“Supply”the extremum in production occurs when d[σ32]/dt=0and has a value that approximately is∝1/g.With the assumption that“Supply”does not have time to change before extreme response is obtained,we identify R with1/g and thereby with the free energy difference∆G that controls the HS.The early rise in r is reproduced when mostσ32are bound to RNAp reflected in the condition g[DnaK]<<1.This implies a significant increase inσ32 lifetime under a positive HS,and implies that the early HS is due to a changed depletion rate ofσter the response is modified,partly by a changed “Supply”andfinally by a changed level of the heat shock induced protease HflB that depends on and counteracts theσ32level in the cell.The largest uncertainty in our analysis is the possibility of a significant time variation in“Supply”and HflB level during the HS.As these will govern the late stages of the heat shock,in particular including its decline,the variation in“∆G”for proteins in the cell may easily be underestimated from using the peak height variation with T.Adding to the uncertainty in what ∆G precisely represents is also the fact that although we only measure DnaK, it can be the complex of the heat shock proteins DnaK,GrpE and DnaJ that sense the state of unfolded proteins in the cell,as removal of any of these display an increased life time ofσ32[9,10].Such cooperativity may amplify the heat shock.For thefinal interpretation of∆G we stress that it effectively counts the free energy difference between the complex DnaK·σ32and that of DnaK being free or being bound to unfolded proteins in the cell.Dependent on the fraction of DnaK relative to unfolded proteins[U]in the cell,i.e.whether K DU[U f]is larger or smaller than1,the HS will depend or not depend on the overall folding stability of proteins in the cell.Thus for much more unfolded proteins than DnaK in the cell,the measured∆G reflect both an increases6of the binding to unfolded residues K DU[U f]as well as a decrease of the DnaK·σ32binding K D32when moving away from T0.Our data does not discriminate between these processes.This discrimination can however be obtained from the data of ref.[16]where it was found that overexpression of DnaK through aσ32dependent pathway in fact repress HS.As DnaK·σ32 binding still play a crucial role in this setup,the vanishing HS of[16]support a scenario where too much DnaK imply K DU[U f]<<1.Then g in eq.10 and thereby theσ32response becomes insensitive to the amount of unfolded proteins in the cell.We conclude that the HS is induced through the changed folding stability of proteins throughout the cell,sensed by a changed need of chaperones.We believe this reflect primarily an increased amount of proteins that are on the way to become folded(nascent proteins),and not an increased denaturation of already folded proteins,because spontaneous denaturation of proteins is extremely unlikely at these temperatures.Thus the deduced sensitivity to the thermodynamic stability of proteins may primarily reflect a correspondingly sensitivity to change in folding times.We now discuss related proposals of“cellular thermometers”for the HS.
