转化 建议所使用的感受态细胞效率≥1×108cfu/μg. 1)冰上融化一管100 μl的DH5a感受态细胞,加入5μl已稀释的 的反应液到感受态细胞中,轻轻混匀,冰上孵育30分钟。 2)42℃水浴中热激90秒后快速放入冰上3分钟。 3)加入500μl SOC液体培养基,37℃复苏45-60分钟。 4)9,000rpm离心3分钟收集菌体,根据需要将一定量的菌体均 匀的涂布在含抗生素平板(Amp. 50-100 ug/mL)上。 5) 37 ℃培养18-24h后,平板置于4 ℃保存。

⽣物素标记EMSA探针-AP1⽣物素标记EMSA探针-AP1产品简介:⽣物素标记EMSA探针-AP1是⽤于EMSA(也称gel shift)研究的并经⽣物素(Biotin)标记的AP1 consensus oligonucleotide。

这个⽣物素标记的双链寡核苷酸含有公认的AP1结合位点,可以⽤作EMSA研究时的探针。

AP1 consensus oligo的序列如下:5’-CGC TTG ATG ACT CAG CCG GAA-3’3’-GCG AAC TAC TGA GTC GGC CTT-5’本⽣物素标记EMSA探针已经过纯化,可以直接⽤于EMSA结合反应。

本⽣物素标记EMSA探针可以和碧云天的化学发光法EMSA试剂盒(GS009)配套使⽤。

⼀个包装的⽣物素标记探针可以进⾏约200-400个样品的EMSA检测。

保存条件:-20℃保存,⼀年有效。

注意事项:避免加热到40℃以上,温度过⾼会导致双链DNA探针解聚成单链。

⽽单链⽆法⽤于EMSA研究。

对于基于⽣物素标记的EMSA检测的详细操作可以参考碧云天的化学发光法EMSA试剂盒(GS009)的使⽤说明。

荧光纳米级显微镜揭示HIV-1病毒成熟过程中表面蛋白结构重排泛瑞翻译人类免疫缺陷病毒I型(Human immunodeficiency virus type 1,HIV-1)从宿主细胞出芽后依然是未成熟的粒子,其主要结构蛋白Gag经过后续的酶解后才能形成成熟的病毒粒子且具有侵染能力。

10月26日的科学杂志刊发了德国海德堡大学一个团队的研究文章,他们利用超高倍成像技术直观观察了HIV病毒粒子成熟的关键步骤。

Chojnacki等通过利用受激发射湮灭超高分辨率荧光显微镜观察HIV-1病毒外层包膜糖蛋白在病毒颗粒上的分布,发现成熟过程诱发的包膜蛋白聚集依赖于包膜蛋白尾端和Gag蛋白互作。

他们把糖蛋白在表面的聚集程度和病毒的侵染效率关联分析后发现,病毒内部和外部有着巧妙的耦合作用:内部蛋白结构的酶解后结构重排促使病毒表面结构改变,为侵染做准备。

Chojnacki等认为Gag酶解诱发的稀少的包膜蛋白二聚体在病毒表面聚集可能是HIV-1成熟的一个基本方面。

这一运用直观的高倍成像技术揭示的HIV成熟的过程和机制,有利于改善人们研发临床HIV疫苗的操作和技术,具有重要的理论和指导意义。

Visualization of the viral envelope (Env) glycoprotein distribution on the surface of individual HIV-1 particles with stimulated emission depletion (STED) superresolution fluorescence microscopy revealed maturation-induced clustering of Env proteins that depended on the Gag-interacting Env tail. Correlation of Env surface clustering with the viral entry efficiency revealed coupling between the viral interior and exterior: Rearrangements of the inner protein lattice facilitated the alteration of the virus surface in preparation for productive entry. We propose that Gag proteolysis-dependent clustering of the sparse Env trimers on the viral surface may be an essential aspect of HIV-1 maturation.。

组织内质网内钙离子浓度荧光检测试剂盒产品说明书(中文版)主要用途组织内质网内钙离子浓度荧光检测试剂是一种旨在通过内质网钙离子特异性荧光探针Mag-Fluo--AM,与钙离子结合后,其荧光强度显著增强,在荧光分光光度仪下观察相对荧光峰值的变化,来测定内质网中总钙离子浓度的权威而经典的技术方法。

该技术经过精心研制、成功实验证明的。

其适用于各种组织(动物、人体等)内内质网钙离子的浓度检测。

产品严格无菌,即到即用,操作简捷,性能稳定,检测敏感。

技术背景钙是人体中的一种重要电介质和矿物质,占1150克。

其中99%的钙以氟磷灰石钙(calcium fluorophosphate apatite)形式存在于骨组织中。

非骨组织钙成分主要存在于细胞内。

钙具有两种主要形式:一种是非弥散型蛋白结合钙,其构成约40%至50%的血清中的总钙含量;另一种为弥散型游离钙,其进一步分成具有活性的复合钙(complexed calcium)和离子钙(ionized calcium)。

钙对神经肌肉兴奋性(neuromuscular excitability)、血液凝固、酶的激活、离子跨膜运输、释放神经传导介质、信使传导等方面起着重要作用。

在细胞内,钙离子主要储存在线粒体和内质网等细胞器中。

其中内质网钙离子储存在控制细胞活化、调控信号通路、蛋白分子转录后修饰,维持细胞内环境钙离子平衡方面起着重要作用。

内质网钙的检测包括沉淀法、EDTA鳌合滴定法、火焰光度法(flame photometry)、原子吸收分光光度法等技术,但容易受到钠、钾、磷酸盐、硫酸盐的干扰,或者受限于仪器的特殊的要求,以及需要内质网纯化。

Mag-Fluo--AM染料是一种钙离子低亲和性的荧光标记染料,特异性地由内质网捕获,可以检测内质网内的游离钙离子浓度的变化。

在荧光分光光度仪下,激发波长490nm,散发波长525nm,来定量测定内质网的钙浓度。

MMP-1基因位于人类染色体11q22.3-q23,由10个外显子编码出一种含585个氨基酸的酶。
