植物查尔酮合酶(CHS) 酶联免疫分析使用说明书使用目的:本试剂盒用于测定植物组织,细胞及其它相关样本中查尔酮合酶(CHS) 活性。
实验原理本试剂盒应用双抗体夹心法测定标本中植物查尔酮合酶(CHS) 水平。
用纯化的植物查尔酮合酶(CHS) 抗体包被微孔板,制成固相抗体,往包被单抗的微孔中依次加入植物查尔酮合酶(CHS) ,再与HRP 标记的查尔酮合酶(CHS) 抗体结合,形成抗体-抗原-酶标抗体复合物,经过彻底洗涤后加底物TMB 显色。
TMB 在HRP 酶的催化下转化成蓝色,并在酸的作用下转化成最终的黄色。
颜色的深浅和样品中的植物查尔酮合酶(CHS) 呈正相关。
用酶标仪在450nm 波长下测定吸光度(OD 值),通过标准曲线计算样品中植物查尔酮合酶(CHS) 活性浓度。
标本要求1.标本采集后尽早进行提取,提取按相关文献进行,提取后应尽快进行实验。
若不能马上进行试验,可将标本放于-20℃保存,但应避免反复冻融2.不能检测含NaN3 的样品,因NaN3 抑制辣根过氧化物酶的(HRP)活性。
操作步骤1. 标准品的稀释:本试剂盒提供原倍标准品一支,用户可按照下列图表在小试管中进行稀释。

产品说明:辅酶ⅡNADP(H)广泛存在于动物、植物、微生物和培养细胞中,NADP +和NADPH 含量测定可以计算NADP (NADPH +NADP +)含量和NADPH/NADP+比值,其变化与磷酸戊糖途径和生物合成以及抗氧化反应密切相关。
NADPH/NADP +比值不仅是细胞氧化还原态的主要标志之一,而且在PPP 途径、生物合成和抗氧化代谢中具有重要调控作用。
分别用酸性和碱性提取液提取样品中NADP +和NADPH 。
NADPH 通过PMS 的递氢作用,使氧化型噻唑蓝(MTT )还原为甲瓒,570nm 下检测吸光值,从而测定NADPH 含量。

复合磷酸酯酶拼音名:Fuhe Linsuanzhimei英文名:Phosphoesterase Complex书页号:D9-210 标准编号:WS-10001-(HD-0858)-2002本品系自大麦芽中提取的具有磷酸酯酶活性的物质。
按干燥品计算,每1mg含磷酸酯酶的效价不得少于3200单位。
【性状】本品为浅褐黄色粉末,微臭,味微酸。
【鉴别】取本品0.5g,置研钵中,加三羟甲基氨基甲烷缓冲液(pH7.0)(取三羟甲基氨基甲烷3.0g,加水400ml溶解,用0.1mol/L盐酸溶液调节pH值至7.0,用水稀释至500ml)10ml,充分研磨,匀浆液以7000转/分的转速离心10分钟,取上清液用定性滤纸过滤,取滤液作为供试品溶液:取供试品溶液1ml,加核糖核酸溶液(取核糖核酸2g,加水80ml,摇匀,用氢氧化钠溶液调节pH值至7.0~7.2使溶解,加水稀释至100ml,现配现用或-20℃冷冻保存)1ml与0.01mol/L氯化锌溶液0.5ml,混匀,置65.0℃±0.5℃水浴保温15分钟;以上述三羟甲基氨基甲烷缓冲液1ml代替供试品溶液同法操作作为对照。
保温结束后,各加0.005ml溴乙锭溶液(取溴乙锭1g,加水100ml,置磁力搅拌器搅拌数小时使其完全溶解,避光保存),摇匀,于暗处在紫外灯(波长300nm)下观察,对照应显明亮的橙色荧光,供试品则不显荧光。
【检查】干燥失重取本品,在105℃干燥4小时,减失重量不得过8.0%(中国药典2000年版二部附录ⅧL)。
【效价测定】供试品溶液的制备取本品250mg,精密称定,置100ml量瓶中,加水溶解并稀释至刻度,摇匀,呈浑浊的溶液。
测定法取试管2支,分别精密加入核糖核酸溶液[取核糖核酸(含量80%以上)1.0g,加水30ml,摇匀,用氢氧化钠试液调节pH值至7.0~7.2,再加水稀释至50ml]3ml、三羟甲基氨基甲烷缓冲液[取三羟甲基氨基甲烷1.2g,加水50ml溶解后,用0.1mol/L的盐酸溶液(约85ml)调节pH值至7.0~7.2,加水稀释至200ml] 1.5ml、0.01mol/L的氯化锌溶液0.5ml,在70℃水浴中保温5分钟,分别精密加入供试品溶液1ml,摇匀,置70℃水浴中准确反应30分钟,再精密加入高氯酸钼酸铵试液(取钼酸铵0.25g,加70%~72%高氯酸3.5ml及水适量使溶解,用水稀释至100ml)5ml,摇匀,立即置冰浴中冷却15分钟,滤过,取续滤液2ml置100ml量瓶中,加水稀释至刻度,照分光光度法(中国药典2000年版二部附录ⅣA),在260nm的波长处以水为参比测定吸收度A,以水1ml代替供试品溶液,同法操作,测定吸收度A。

【摘要】目的:建立复方苦参注射液中5-羟甲基糠醛(5-HMF)含量的测定方法。
方法:采用高效液相色谱法,色谱柱为Waters XSelect CSH TM C 18(250 mm×4.6 mm,5μm),流动相为甲醇-0.05%甲酸水溶液梯度洗脱,流速为0.8 mL·min^(-1),检测波长为284 nm,柱温20℃,进样量10μL。
结果:5-HMF质量浓度在
0.022~1.11μg(r=0.9998)范围内质量呈良好线性关系。
5-HMF平均回收率为95.6%(RSD=6.70%)(n=9)。
结论:本研究方法重复性好,准确度高,可用于复方苦参
注射液中5-HMF的含量测定。
