Easy for use 套装二使用说明书一、SCSP number: SCSP- 112二、主要用途:Easy for use套装二专为小鼠胚胎干细胞(mES)、小鼠iPS细胞、人胚胎干细胞(hES)及人iPS细胞研究者设计,尤其适合对以上细胞有少量多次传代需求的研究者。

本套装包含三支CF-1小鼠MEF细胞、一管MEF培养液及一管0.2%明胶,无需准备其它试剂,方便即用。

三、产品组成:备注:每支CF-1-r P3 106/支可铺6孔板的两个孔;两个3.5 cm dish;一个6cm dish 或其它底面积为20cm2的细胞培养器皿。

四、保存温度:MEF培养液:-20℃可长期保存6个月,4℃保存请于两周内使用完。

0.2%明胶:4℃可保存6个月。

CF-1-r P3:液氮可长期保存。

五、使用方法:(一)用于培养mES、小鼠iPS细胞的使用方法:1.在细胞培养皿内加入0.2%明胶,摇匀后覆盖底面即可,于37℃细胞培养箱至少放置15 min以上。

2.取出细胞培养皿,吸除0.2%明胶,加入事先水浴加热至37℃的MEF培养液,6孔板加入2 ml/孔,3.5cm dish加入2 ml/dish,6cm dish加入4 ml/dish,置于37℃细胞培养箱。

3.按实验需要复苏CF-1-r P3若干支,将冻存管从液氮中取出,置于37℃水浴中使之迅速融解,取出后拿到超净台内用75%酒精擦拭冻存管旋口处及外壁,防止污染。

4.将冻存管内细胞悬液转移至含2 ml MEF培养液的15 ml离心管内,以1000 rpm离心5 min,离心后将上清液吸除,另加入新鲜的MEF培养液1 ml,重悬后平均加入到含MEF培养液的6孔板内,将6孔板轻轻摇匀后置于37℃细胞培养箱。

24 h以后可以传入mES或小鼠iPS细胞。

(二)用于培养hES、人iPS细胞的使用方法1.在细胞培养皿内加入5% Matrigel,摇匀,覆盖底面即可,于37℃细胞培养箱至少放置30 min以上。

En caso de la adquisición de más de un filtro yestén apilados, deben separarse desatornillando la unión en los apoyos superiores.2.Colocar los colectores de entrada y salida fijando las bridas con sus correspondientesjuntas.3.Conectar las tuberías de drenaje y dirigir a la zona deseada.4.Llenado del filtro. Primero llenar el filtro de agua hasta la mitad. Abrir la tapa superior yverter el material filtrante despacio para evitar daños en la estructura interna. La cantidad de arena necesaria se indica en la ficha técnica (Pág.2).5.Colocar los manómetros de presión, uno en el colector de entrada del agua y otro en elcolector de salida del agua filtrada.probar que las uniones están bien ajustadas, abrir el suministro de agua y verificarque no hay ninguna fuga.7.Realizar una primera limpieza para eliminar las posibles impurezas de la arena.8.Filtrado. Abrir el paso del agua al caudal recomendado (según la calidad del agua) y auna presión de trabajo de 8kg/cm².9.Cuando los manómetros indiquen una diferencia de presión igual o superior a 0,5Barproceder a la limpieza del filtro invirtiendo la dirección del agua.4.FuncionamientoEl filtrado mediante arena es un método de filtración en profundidad y con una gran eficiencia. Consiste en la separación física entre el agua y las sustancias que se encuentren suspendidas en ella. El agua desciende atravesando el lecho filtrante de arena donde quedan atrapadas las impurezas y el sistema de brazos permite la salida del agua filtrada.Cuando el filtro de arena se colmata produce una diferencia de presión entre la entrada y la salida de agua. El ciclo de limpieza comienza al alcanzar una diferencia de presión de 0.5bar. El ciclo de limpieza consiste en invertir el sentido de circulación: el agua asciende y arrastra todas las partículas atrapadas en el lecho filtrante de arena y son expulsadas.5.Mantenimiento y recambios5.1Mantenimiento1.Evitar dejar el filtro sin agua para un correcto mantenimiento del material filtrante. Si laarena se seca y se apelmaza, abrir la boca inferior del filtro, vaciar su contenido y reemplazarla. En caso de que la arena no evacue adecuadamente, conectar el flujo de agua para facilitar su salida.2.Un correcto mantenimiento del filtro incluye comprobar el estado de la pintura. 以下对各种软件的功能进行大致说明。

1.1 HiDiscovery软件说明利用HiDiscovery软件可以对交换机的IP地址进行设置。

注意:输入密码时请注意大小写之分,并确保JAVA软件的版本足够新。

1.2 Hirschmann Industrial HiVision软件说明软件Hirschmann Industrial HiVision可以扫描网络,生成网络拓扑结构图,同时能够监控网络状态。

Hirschmann Industrial HiVision界面如下图所示:在‘选项’->‘发现设备’->‘新建’中设置搜索设备的IP地址范围,并勾选‘激活’选项,如下图所示。

1.3 HiView软件说明HiView软件是一款绿色版的交换机Web配置软件,内部集成JAVA,输入交换机的IP地址,即可访问。

第二章交换机配置主要内容:1.配置环网协议Hiper-ring2.VLAN划分3.路由功能⏹在每一项的配置页面中,通常都会有“Set”和“Reload”两个按钮。

其中“Set”按钮的作用是使所修改的配置即时生效;“Reload”按钮的作用是刷新该页的数据。

因此,在对交换机进行配置修改后,都需要点击该页面中的“Set”按钮,以确保配置修改可以即时生效。

⏹注意:点击“Set”按钮后,只是确保修改生效,并没有保存进交换机内部的配置文件。

For aiming the cabinet downward greater than a small angle, the use of the thumbscrew insert on the rear bottom of the cabinet, in the pole mount recess, can allow an M8 x 1.25 mm eyebolt to pull the cabinet back. Maximum angle is 45 degrees.EYEBOLT SPECIFICATIONSEyebolts used in the Group A should be M6 metric thread. Eyebolts and bracket bolts must conform to certain minimum strength criteria for safety reasons. Unspecified eyebolts found at local hardware stores are not strong enough to maintain safety for overhead flying or rigging. Use only forged steel shoulder machinery eyebolts de-signed for rigging use, which adhere to the stated standards as outlined below.Forged shoulder machinery eyebolts used for Group A inserts should be an M6 size, 1.0 mm per thread, and must conform to DIN 580 or ASTM A489 standards, for Germany, DGUV 17 (formerly BGV-C1) is the relevant standard. Minimum shank length should be 12 mm. Maximum recommended shank length is 26 mm (to avoid fouling the sound absorbent liner inside). 百奥赛图基因⽣物技术有限公司1. CRISPR-Based EGE技术简介CRISPR(clustered,regularly interspaced,short palindromic repeats)是⼀种来⾃细菌降解⼊侵的病毒DNA或其他外源DNA的免疫机制。

在细菌及古细菌中,CRISPR系统共分成3类,其中Ⅰ类和Ⅲ类需要多种CRISPR相关蛋⽩(Cas蛋⽩)共同发挥作⽤,⽽Ⅱ类系统只需要⼀种Cas蛋⽩即可,这为其能够⼴泛应⽤提供了便利条件。

⽬前,来⾃Streptococcus pyogenes的CRISPR/Cas9系统应⽤最为⼴泛。

Cas9蛋⽩(含有两个核酸酶结构域,可以分别切割DNA两条单链。

Cas9⾸先与crRNA及tracrRNA结合成复合物,然后通过PAM序列结合并侵⼊DNA,形成RNA-DNA复合结构,进⽽对⽬的DNA双链进⾏切割,使DNA双链断裂。

由于PAM序列结构简单(5’-NGG-3’),⼏乎可以在所有的基因中都能找到⼤量靶点,因此得到⼴泛的应⽤。

CRISPR/Cas9技术已经成功应⽤于植物、细菌、酵母、鱼类及哺乳动物细胞,是⽬前最⾼效的基因组编辑技术。

通过基因⼯程⼿段对crRNA和tracrRNA进⾏改造,将其连接在⼀起得到sgRNA(single guide RNA)。

融合后的RNA具有与野⽣型RNA类似的活⼒,但因为结构得到了简化,更⽅便研究者使⽤。

通过将表达sgRNA的元件与表达Cas9的元件组装到⼀个载体中,得到可以同时表达两者的质粒,将其转染细胞,就能够对⽬的基因进⾏操作。

⽬前的CRISPR/Cas9技术应⽤于基因敲⼊时,外源DNA同源重组效率⼀直很低。

为提⾼其重组效率,百奥赛图在CRISPR/Cas9技术基础上开发和优化出⼀套⾼效的基因敲除/敲⼊技术(Biocytogen Extreme Genome Editing System,简称EGE技术)。

该技术⽐普通的CRISPR/Cas9技术介导的同源重组效率提⾼了10-20倍,从⽽使得基因改造更加快速和⽅便。
