Abstract: A UPLC-MS/MS method was established to quantitatively determine the content of alliin in animal plasma to study whether alliin and alliin in garlic enteric preparations can react to produce the active ingredient allicin in the in vivo environment. Methods Reversed-phase C18 column (Waters ICQUITY UPLC BEH, 100 × 2.1 mm, 1.7μm), column temperature: 40 ℃, flow rate: 0.15 mL/min, injection volume: 2μl, Mobile phase: 0.1% formic acid (A)-acetonitrile (B), gradient elution; mass spectrometry ionization: ESI+, determination of allicin in rat plasma . Results The results of two parallel experiments of garlic enteric preparation and enzymatic garlic powder showed that in the garlic enteric preparation with allinase, the plasma concentration of alliin in the blood of rats was significantly lower. Conclusion A UPLC-MS/MS method for the quantitative determination of alliin in animal plasma has been established. 高效液相色谱法测定硫酸氢氯吡格雷片有关物质作者:李飞张黎娟孟祥松等来源:《中国医药导报》2012年第08期硫酸氢氯吡格雷是一种新型血小板聚集抑制剂,临床上用于治疗中风、心肌梗死、外周动脉疾病等[1],市场需求量较大,但国内对其制剂有关物质检测报道较少,为了更有效地控制本品质量,本文参照美国药典[2]建立测定硫酸氢氯吡格雷片有关物质的HPLC法,方法专属、灵敏、可靠,有效地控制了本品的质量。

1 仪器与试药1.1 仪器岛津高效液相色谱仪,紫外检测器(型号:SPD-20AD),泵(型号:LC-20AD),色谱工作站(LC Solution色谱工作站),电子天平(METTLER TOLOD XS105电子天平),紫外-可见分光光度仪(型号:Agilent 8453)。

1.2 试药硫酸氢氯吡格雷片(合肥医工医药有限公司,规格:75 mg/片,批号:20081226),波立维(法国赛诺菲,规格:75 mg/片,批号:2613),泰嘉(深圳信立泰,规格25 mg/片,批号:20090312),硫酸氢氯吡格雷对照品(中国药品生物制品检定所,批号:100819-200601),氯吡格雷杂质A对照品(USP,批号:G0H250),氯吡格雷杂质B对照品(USP,批号:G1H047),氯吡格雷杂质C对照品(USP,批号:F2H215),起始原料、中间体及空白辅料(合肥医工医药有限公司提供)。

1.3 试剂乙腈(色谱纯),磷酸二氢钾(国药集团化学试剂有限公司),甲醇(分析纯),水(纯化水)。

2 方法与结果2.1 色谱条件色谱柱为ULTRON ES-OVM(4.6 mm×150.0 mm,5 μm),流动相为0.01 mol/L磷酸二氢钾溶液-乙腈(75∶25),流速为1.0 mL/min,检测波长为220 nm,柱温为25℃,进样量为20 μL。

2.2 溶液的配制2.2.1 供试品溶液配制本品细粉适量(约相当于硫酸氢氯吡格雷25 mg),精密称定,置100 mL量瓶中,加甲醇适量使溶解,再加流动相稀释至刻度,摇匀,滤过,作为供试品溶液。

非洛地平纳米脂质载体在大鼠体内的药动学与药效学研究曹桑博;王敏;谢鹏【期刊名称】《西北药学杂志》【年(卷),期】2022(37)4【摘要】目的研究非洛地平(FLD)纳米脂质载体(FLD-NLCs)在大鼠体内的药动学特征以及对高血压模型大鼠的血压控制效果。

方法以山嵛酸甘油酯(Compritol 888 ATO)作为固体脂质,聚乙二醇-15羟基硬脂酸酯(Solutol HS 15)作为液体脂质,泊洛沙姆188(Poloxamer 188)作为表面活性剂,使用高压均质技术制备FLD-NLCs,并对FLD-NLCs的理化性质进行评价,考察FLD-NLCs在不同pH介质溶液中的稀释稳定性以及体外释药特性;研究FLD原料药和FLD-NLCs的细胞跨膜转运性质;比较FLD混悬剂和FLD-NLCs经大鼠口服给药后的体内药动学及药效学。

结果FLD-NLCs的平均粒径为(126.8±3.3)nm,多聚分散系数(PDI)为(0.184±0.005),Zeta电位为(-26.3±0.6)mV,在透射电镜下可观察到FLD-NLCs呈球形,均匀分散;FLD-NLCs经不同pH介质溶液稀释后稳定性良好,在不同pH介质溶液中均表现为双相释药特征,且药物释放速率无明显差异;FLD-NLCs能有效提高药物的跨膜转运能力;与FLD混悬剂相比,大鼠口服给予FLD-NLCs后显著提高了药物生物利用度,可长时间有效控制血压。

结论FLD-NLCs能显著提高药物的生物利用度,有效控制血压,对非洛地平的临床应用具有重要价值。

摘要:本发明涉及一种超声联合靶向CD133载sPD1微泡的制备方法,具体是将生物素化脂质微泡滴加PE标记链酶亲和素孵育,加入再加入生物素化的CD133抗体,经孵育后离心得到靶向CD133微泡。

将所得的靶向CD133微泡和sPD1质粒室温下孵育后,再用PBS冲洗,离心,即可得到靶向

【摘要】目的建立复方亚油酸乙酯胶丸中橙皮苷的含量测定方法.方法液相色谱法.色谱柱:Kromasil C18柱(4.6mm×250mm,5μm);流动相:甲醇-0.5%冰醋酸溶液(40:60);流速1.0mL·min-1;柱温:40℃;检测波长:283nm.结果橙皮苷在0.07505~0.3752μg范围内呈线性关系(r =0.9997,n=5),平均加样回收率为99.16%,RSD为1.7%(n=6).结论该方法操作简便,结果可靠,可用于复方亚油酸乙酯胶丸中橙皮苷的含量测定.
