一氧化氮(NO)含量检测试剂盒说明书(货号:NM-W-0132 微量法100T/96S)一、产品简介:一氧化氮(Nitric Oxide,NO)是一种气态分子自由基,在生物体内作为重要的信使分子和效应分子,参与调节血管形成、神经传递、免疫调控及肿瘤生长等多种生理及病理过程。

此外,NO是植物的主要生物活性分子,在植物的生长发育和对胁迫的反应过程中参与了多种生理活动,如种子萌发、叶扩展、根生长、花器官发生、气孔运动的调节以及植物的抗逆反应等。

NO在体内或水溶液中极易氧化生成NO2-。

在酸性条件下,NO2-与重氮盐磺酸胺生成重氮化合物,进一步与萘基乙烯基二胺偶合,产物在550nm处有特征吸收峰,测定其吸光值,可以计算NO含量。

方法:液相采用ACQUITY UPLC®BEH C18(2.1 mm×100 mm,1.7 μm)柱,以超纯水(含0.1%甲酸)-乙腈(0.1%甲酸)为流动相,进行梯度洗脱,进样量2 μL,流速0.3 mL·min-1,柱温30 ℃。

质谱采用ESI,正、负离子多反应监测模式,进行定性分析和定量分析。

结果:盐酸普萘洛尔、酒石酸美托洛尔、艾司洛尔、比索洛尔、坎地沙坦、厄贝沙坦、替米沙坦、氯沙坦、缬沙坦和吲达帕胺的分离度良好,线性范围内相关性均较好,R2均大于0.996 3;平均回收率为93%~118%;RSD为1.4%~4.9%(n=6);方法检出限为0.10~5.00 μg·kg-1;方法定量限为0.30~15.00 μg·kg-1;单次分析仅需12 min。

结论:该方法操作便捷、检测时间短、灵敏度高、结果准确,适用于保健食品中10种降压类非法添加化学药的快速筛查及定量检测。

Method: ACQUITY UPLC®BEH C18( 2.1 mm×100 mm, 1.7 μm) column was used in the liquid phase with ultrapure water (containing 0.1% formic acid)-acetonitrile (0.1% formic acid) as the mobile phase for gradient elution, the injection volume was 2 μL, flow rate was 0.3 mL·m in-1, and column temperature was 30 ℃. The mass spectrometry was performed by ESI, positive/negative ion multiple reaction monitoring mode for qualitative and quantitative analysis. Result: The separation degree of propranolol, metoprolol, amlodipine, bisoprolol, candesartan, irbesartan, telmisartan, losartan, valsartan, and indapamide was good, and the linear range correlations were good, all R2 values were greater than 0.996 3. The average recovery rate was 93%~118%. RSD was 1.4%~4.9% (n=6). The LOD was between 0.10~5.00 μg·kg-1, and the LOQ was between 0.30~15.00 μg·kg-1. A single analysis only requires 12 min. The lowest temperature at which self-accelerating decomposition may occur with a substance in the packaging as used for transport is the Self-Accelerating Decomposition Temperature (SADT). The SADT is determined on the basis of the Heat Accumulation Storage Test.Method The Heat Accumulation Storage Test is a recognized test method for thedetermination of the SADT of organic peroxides (see Recommendations on theTransport of Dangerous Goods, Manual of Tests and Criteria - United Nations, NewYork and Geneva).StorageDue to the relatively unstable nature of organic peroxides, a loss of quality will occur over a period of time. To minimize the loss of quality, Nouryon recommends a maximum storage temperature (Ts max. ) for each organic peroxide product.Ts max.40°CNote When stored according to these recommended storage conditions, Trigonox® B-C30 will remain within the Nouryon specifications for a period of at least 3 monthsafter delivery.Packaging and transportThe standard packaging is a 1000 l composite Intermediate Bulk Container (IBC). Delivery in a 20 m3 Nouryon tank container is also possible in a number of countries. Both packaging and transport meet the international regulations. 总一氧化氮检测试剂盒产品简介:总一氧化氮检测试剂盒(Total Nitric Oxide Assay Kit, 即Nitrate/Nitrite Assay Kit)采用了硝酸盐还原酶(Nitrate reductase)还原硝酸盐(nitrate)为亚硝酸盐(nitrite),然后通过经典的Griess reagent检测亚硝酸盐,从而测定出总一氧化氮。

一氧化氮本身极不稳定,在细胞内很快代谢为硝酸盐(Nitrate)和亚硝酸盐(Nitrite),通过上述方法检测出硝酸盐和亚硝酸盐的总量,就可以推算出总的一氧化氮的量。

本试剂盒采用了NADPH依赖性硝酸盐还原酶(NADPH dependent nitrate reductase)。

高浓度的NADPH会干扰后续的检测,消除NADPH的一种常用方法就是使用Lactate dehydrogenase (LDH)清除NADPH。

本试剂盒采用了LDH清除NADPH的方法,使检测结果更加准确。

对亚硝酸盐的检测下限达到2微摩尔/升,在2-80微摩尔/升的范围内有很好的线性关系。

浓度过高的样品可以适当稀释后再进行检测。

样品范围广,可以检测细胞裂解液、组织裂解液、细胞或组织的培养液、血清、血浆或尿液等中一氧化氮的含量。

酚红和10%血清对测定无明显干扰。

样品需要量少。

根据样品中一氧化氮的浓度不同,仅需0-60微升样品。

检测速度快,仅需约80分钟即可完成检测。

本试剂盒采用了间接的一氧化氮检测方法,如需检测细胞内实际的一氧化氮水平,可以采用碧云天生产的DAF-FM DA (NO荧光探针)(S0019)。

保存条件:-20℃保存,一年有效。

NADPH,Nitrate Reductase,NaNO2 (1M),Griess Reagent I 和Griess Reagent II需避光保存。

NADPH配制成溶液后必须分装并-70℃保存。

注意事项:RPMI 1640等含有较高浓度硝酸盐的培养液容易对本试剂盒的检测产生干扰,请尽量避免。
