摘要:【目的】Mutator(Mu)转座子是玉米基因组内产生诱变的内源转座因子,其在基因组内的拷贝数和诱变效率远远优于其它转座子。利用Mu转座子介导的玉米株高性状的突变体来研究其遗传特性。通过计算后代株高的分离情况,来验证株高的突变是否与插入有关,以及扩增Mu因子插入位点的侧翼序列,为找到影响株高的基因奠定基础,从而扩展株高突变的分子遗传学研究。此外,还为高产玉米育种提供了种质资源和理论指导。【方法】使用原始Mu群体(uniformMu)与玉米优良自交系综31杂交获得M1代,对其自交后代的M5群体的植株表型进行逐株全生育期的田间调查,根据极端性状的遗传分析和数量性状的统计分析筛选和鉴定Mu转座因子插入诱变的突变群体。【结果】研究结果表明,筛选了M5群体的的75个家系共543株,11BD753、613、728、1748、1779家系出现株高性状的分离,极有可能是Mu插入引起的,其中,11BD753、1779为矮化突变,11BD613、728、1748为高杆突变。【结论】目前经过分析的这几个家系的胚大小性状出现了显著分离,需要进一步对其研究,尤其是要找出Mu插入位点的侧翼序列,再经行基因功能和遗传方面的研究。

Screening Mutator–mediated Plant Height mutation advantage

Abstract: 【Objective】Mutator (Mu) transposons is endogenous transposons producing mutation groups, i n maize genomic .It’s copies and mutation frequency are superior to other transposons. By Mutator–mediated maize plant height to study their genetic characteristics. By calculating the separation of mutation plant height, to verify whether the plant height is related with the insertion of Mutator, and find Mutator flanking sequences, then to lay foundations for finding genes that affect the height of the plant. Moreover, to extend the molecular genetic studies of plant height mutation development, in addition ,to provide germplasm resources and theoretical guidance for breeding high yeild maize. 【Method】Using uniformMu and elite inbred line Z31 crossing got M1and to it’s Self-crossed Progeny M4 do the field survey of the plant phenotype during the whole growth stage, and screening and identifying Mutator–mediated inspectional mutations basis on genetic analysis of the extreme characters and statistical analysis of the quantitative characters. 【Result】The study results show that the plant height mutation, from M5group’s 75 families, total over 543

plant ,11BD753,613,728,1748,1779 family lines appear separation of plant height, most likely caused by Mu insertion, and 11BD753,1779 family lines appear dwarf mutation,11BD613,728,1748 family lines appear high pole mutation.【Conclusion】At present there are remarkable separation of this lines after the analysis the character of these maize embryo size, needing more further research on this lines, particularly needing discover the flanking sequence of the Mu insertion and develop aspect research on gene function and the heredity character .
