无论是构建克隆载体还是表达载体,要根据载体选择合适的内切酶(当然,使用T 载就不必考虑了)。

III类限制性内切酶
能完全肯定的识别位点和切割位点,但切 割位点也是在识别位点的一侧的一定距离,通 常距特异性位点24bp-26bp。
I 型:切割位点不确定,不适用于基因工程。 Ⅱ型:基因工程的工具酶。 Ⅲ型:切割位点不在识别位点,对分子克隆操作 亦无实用意义。
I型限制性内切酶
首先由M.Meselson和R.Yuan在1968年从大 肠杆菌B株和K株分离得到。 作用原理:识别双链DNA未甲基化修饰的特异 序列,与该序列相互作用,然后延DNA分子移 动,在距离特异性识别位点约1000-1500bp处 随机切开一条单链,造成大约75个核苷酸的 切口。
4.DNA的分子结构
DNA分子的不同构型对限制性内切酶 的活性也有很大的影响。某些限制性核酸 内切酶切割超螺旋的质粒DNA所需要的酶量 要比消化线性的DNA量高出很多倍。
5. 缓冲液
是影响限制酶活性的重要因素。商品化的限 制酶一般都带有专用缓冲液。
化学组成 MgCl2、NaCl/KCl:提供Mg2+和离子强度 Tris-HCl:维持pH 二硫苏糖醇(DTT):保持酶稳定性 牛血清白蛋白BSA等:有助于酶的稳定
• 识别序列
通常识别的是4-8bp的回文序列,多为6bp,在 识别序列内或其附近水解DNA链中的磷酸二酯键。
• 酶切切口
齐平末端:在识别序列内的对称轴上切割,其 切割产物具平头末端(不易重新连接)。

限制性核酸内切酶详细资料大全限制性核酸内切酶是可以识别并附着特定的脱氧核苷酸序列,并对在每条链中特定部位的两个脱氧核糖核苷酸之间的磷酸二酯键进行切割的一类酶,简称限制酶。
根据限制酶的结构,辅因子的需求切位与作用方式,可将限制酶分为三种类型,分别是第一型(Type I)、第二型(Type II)及第三型(Type III)。
Ⅰ型限制性内切酶既能催化宿主DNA的甲基化,又催化非甲基化的DNA的水解;而Ⅱ型限制性内切酶只催化非甲基化的DNA的水解。
III型限制性内切酶同时具有修饰及认知切割的作用。

由来一般是以微生物属名的第一个字母和种名的前两个字母组成,第四个字母表示菌株(品系)。
例如,从Bacillus amylolique faciens H 中提取的限制性内切酶称为Bam H,在同一品系细菌中得到的识别不同碱基顺序的几种不同特异性的酶,可以编成不同的号,如HindⅡ、HindⅢ,HpaI、HpaⅡ,MboI、MboⅡ等。
限制性核酸内切酶别名:Endodeoxyribonuclease简称限制酶酶反应限制性内切酶能分裂DNA分子在一限定数目的专一部位上。
它能识别外源DNA并将其降解。
单位定义:在指明pH与37℃,在0.05mL反应混合物中,1小时消化1μg的λDNA的酶量为1单位。
性状制品不含非专一的核酸水解酶(由10单位内切酶与1μg λDNA,保温16小时所得的凝胶电泳图谱的稳定性表示),这类酶主要是从原核生物中分离出来的,迄今已经从近300多种不同的微生物中分离出约4000种限制酶。
