三、培养条件1. 培养温度:37℃2. CO2浓度:5%3. 外源性添加物:Sciencell内皮细胞培养基中已经包含了必要的营养因子、生长因子和抗生素/抗菌素,不需要额外添加外源性添加物。
4. 培养密度:内皮细胞的培养密度应当根据不同细胞类型和研究需要适当调整,一般为1-3x10^4 cells/cm2。
5. 培养时间:培养时间应当根据不同细胞类型和实验需要适当调整,一般为4-7天。
四、保存条件Sciencell内皮细胞培养基应当在4℃低温保存,避免阳光直射和冻结。
保存期长达6个月,但是在保质期内使用会更好。
打开后,建议在一个月内使用完毕。
五、使用说明1. 移液操作:使用Sciencell内皮细胞培养基时应当采用无菌条件下的移液操作,并避免出现气泡和异物。
2. 细胞接种:将内皮细胞接种到预先涂覆的细胞培养底物上,并根据需要进行细胞划分和培养状态监测。
3. 细胞增殖:使用Sciencell内皮细胞培养基可促进内皮细胞的增殖和生长,建议每3-4天更换新的培养基。
4. 细胞检测:在细胞培养期间需监测细胞的培养状态、生长状态和增殖情况。

内质网钙的检测包括沉淀法、EDTA鳌合滴定法、火焰光度法(flame photometry)、原子吸收分光光度法等技术,但容易受到钠、钾、磷酸盐、硫酸盐的干扰,或者受限于仪器的特殊的要求,以及需要内质网纯化。
Mag-Fluo--AM染料是一种钙离子低亲和性的荧光标记染料,特异性地由内质网捕获,可以检测内质网内的游离钙离子浓度的变化。
在荧光分光光度仪下,激发波长490nm,散发波长525nm,来定量测定内质网的钙浓度。

点击显示屏的全血模式→运行,执行完毕仪器自动打印结果。
2、预稀释模式:推开紫色小门,将空子弹头杯放入带小适配器的绿色适配器,然后将样品适配器放入样品台内,关上样品台门,点击显示屏预稀释模式→分装→执行,仪器自动放入500UL的稀释液,执行完毕后,取出子弹头杯手工加入20UL的末梢血(也可以手工稀释500UL+20UL),再放到小适配器上,关上样品台门,主界面上选择预稀释模式→运行,执行完毕仪器自动打印结果。
注意:在输入样本编号时,可按ALPH进行数字,大写字母,小写字母循环切换。

1、空白确认:取新杯用稀释液测量空白:WBC≦0.3,RBC≦0.03,HGB≦1G/L,PLT≦10.空白确认后,并重复性好,即可做较正。

收到常温细胞后如何处理?(细胞培养详细操作步骤请参照《普诺赛细胞培养操作指南》)1. 收到常温细胞后,及时拍照记录有无漏液/瓶身破损现象。
2. 用75%酒精擦拭细胞培养瓶表面,显微镜下观察细胞状态。
先不要打开培养瓶盖,将细胞置于细胞培养箱内静置培养2-4小时,以便稳定细胞状态。
3. 仔细阅读细胞说明书,了解细胞相关信息,如贴壁特性(贴壁/悬浮)、细胞形态、所用基础培养基、血清比例、所需细胞因子、传代比例、换液频率等。
4. 静置完成后,取出细胞培养瓶,镜检、拍照,记录细胞状态(所拍照片将作为后续服务依据);建议细胞传代培养后,定期拍照、记录细胞生长状态。
5. 若细胞生长密度超过80%1:2~1:3(按实际收货细胞密度决,移除细胞培养瓶内培养基,预留6mL 左右原瓶培养基继续培养,直至细胞密度达左右再进行传代操作,注意拧松瓶盖或更换透气瓶盖。

Biacore X100简易操作指南一、Biacore X100设备介绍分子相互作用分析系统Biacore是由仪器主机和电脑两部分组成。
仪器开关在主机右后方,仪器正面左侧为缓冲液入口,可放置运行缓冲液,中上部分为状态面板,以指示灯显示仪器状态,状态面板右边即为芯片的放置位置,芯片会与SPR光学检测器、微流控系统IFC嵌合,开展结合分析过程。
右侧为样品舱,可放置15个1.5ml的EP管,下方放有废液瓶。
如下图所示。
二、实验准备实验前需根据实验需求选择芯片,GE提供羧基葡聚糖表面的CM系列芯片、适用于HIS标签蛋白的NTA芯片、应用于生物素标记分子的SA芯片(通常用于核酸、多肽)等,蛋白研究最常用CM5芯片,开展实验时会固定一个分子(配体)在芯片表面,另一个分子(分析物)以流动相流过芯片,通过表面等离子共振SPR的原理研究相互作用的过程。
根据实验目的,需要确定配体的偶联量,可通过以下公式进行计算,其中R max代表芯片的最大结合容量,为分析物在表面的最大响应值,Sm为化学计量比,LigandMW为配体的分子量,AnalyteMW为分析物的分子量,R L代表配体的偶联水平,也即需要求得的值,实际固定量一般设为1.5R L。
动力学分析中,R max需低于100RU。
Biacore对分子的互作检测是基于功能性的,所以实验过程必须要新鲜有活性的样品,0.22um膜过滤或者仔细离心,配体纯度要求在90%以上,分析物的纯度依实验类型而定,进行动力学/亲和力分析时需要90%以上的纯度,进行浓度/特异性测定时可以进混合样品。
缓冲体系选择:大多数缓冲液均适用,可根据样品活性确定最适缓冲液,须新鲜配制,经0.22um膜过滤及脱气。
氨基偶联方法固定配体时,缓冲液中不能含有Tris、甘氨酸等带有伯胺基的物质。
分析物溶液中,不能含甘油、蔗糖、咪唑等高折光率物质,如果样品中含有该物质,可通过缓冲液置换、透析的方法除去。
三、实验操作在该部分中,选用CM5芯片,以配体mouse-anti-human β2-microglobulin antibody和分析物human β2-microglobulin的动力学分析为例进行操作介绍(样品均来自 Biacore getting started kit),实验流程为配体固定-表面测试-再生条件筛选-分析物进样-芯片再生-数据分析。

(ECM)
Catalog #1001 Product Description
Endothelial Cell Medium (ECM), when used with Endothelial Cell Growth Supplement (ECGS, Cat #1052) and 25 ml of fetal bovine serum (FBS) is a complete medium designed for optimal growth of normal endothelial cells in vitro. It is a sterile, liquid medium which contains essential and non-essential amino acids, vitamins, organic and inorganic compounds, hormones, growth factors, trace minerals and a low concentration of fetal bovine serum (5%). The medium is HEPES and bicarbonate buffered and has a pH of 7.4 when equilibrated in an incubator with an atmosphere of 5% CO2/95% air. The medium is formulated (quantitatively and qualitatively) to provide an optimally balanced nutritional environment that selectively supports the growth of normal endothelial cells in vitro.
Components
ECM consists of 500 ml of basal medium, 25 ml of FBS (Cat. #0025), 5 ml of Endothelial Cell Growth Supplement (ECGS, Cat. #1052) and 5 ml of penicillin/streptomycin solution (P/S, Cat. #0503). Note: FBS, ECGS and P/S are not pre-mixed in ECM; they must be added separately to make the complete ECM.
Product Use
ECM is for research use only. It is not approved for human or animal use, or for application in in vitro diagnostic procedures.
Storage
Store the basal medium at 4o C and the ECGS, FBS and P/S solution at -20o C. Protect from light. Shipping
Basal medium: room temperature. Supplements: dry ice.
Instructions for Use
Thaw ECGS, FBS and P/S solution at 37o C. Gently tilt the tubes several times to ensure the contents are completely mixed before adding to the medium. Spray the medium bottle and tubes with 70% ethanol, and wipe to remove excess liquid. In a sterile field, remove the caps without touching the interior threads with fingers. Add ECGS, FBS and P/S solution to the medium and mix well. Since several components are light-labile, the medium should not be exposed to light for extended periods. We do not recommend warming medium in a 37o C water bath prior to use. When stored in the dark at 4o C, the reconstituted medium is stable for one month.
Caution: If handled improperly, some components of the medium may present a health hazard. Take appropriate precautions when handling it, including the wearing of protective clothing and eyewear. Dispose of properly.。
