2. 用75%酒精擦拭细胞培养瓶表面，显微镜下观察细胞状态。

先不要打开培养瓶盖，将细胞置于细胞培养箱内静置培养2-4小时，以便稳定细胞状态。

3. 仔细阅读细胞说明书，了解细胞相关信息，如贴壁特性（贴壁/悬浮）、细胞形态、所用基础培养基、血清比例、所需细胞因子、传代比例、换液频率等。

4. 静置完成后，取出细胞培养瓶，镜检、拍照，记录细胞状态（所拍照片将作为后续服务依据）；建议细胞传代培养后，定期拍照、记录细胞生长状态。

5. 若细胞生长密度超过80%1:2~1:3（按实际收货细胞密度决，移除细胞培养瓶内培养基，预留6mL 左右原瓶培养基继续培养，直至细胞密度达左右再进行传代操作，注意拧松瓶盖或更换透气瓶盖。

Biacore X100简易操作指南一、Biacore X100设备介绍分子相互作用分析系统Biacore是由仪器主机和电脑两部分组成。

仪器开关在主机右后方，仪器正面左侧为缓冲液入口，可放置运行缓冲液，中上部分为状态面板，以指示灯显示仪器状态，状态面板右边即为芯片的放置位置，芯片会与SPR光学检测器、微流控系统IFC嵌合，开展结合分析过程。

右侧为样品舱，可放置15个1.5ml的EP管，下方放有废液瓶。

如下图所示。

二、实验准备实验前需根据实验需求选择芯片，GE提供羧基葡聚糖表面的CM系列芯片、适用于HIS标签蛋白的NTA芯片、应用于生物素标记分子的SA芯片（通常用于核酸、多肽）等，蛋白研究最常用CM5芯片，开展实验时会固定一个分子（配体）在芯片表面，另一个分子（分析物）以流动相流过芯片，通过表面等离子共振SPR的原理研究相互作用的过程。

根据实验目的，需要确定配体的偶联量，可通过以下公式进行计算，其中R max代表芯片的最大结合容量，为分析物在表面的最大响应值，Sm为化学计量比，LigandMW为配体的分子量，AnalyteMW为分析物的分子量，R L代表配体的偶联水平，也即需要求得的值，实际固定量一般设为1.5R L。

动力学分析中，R max需低于100RU。

Biacore对分子的互作检测是基于功能性的，所以实验过程必须要新鲜有活性的样品，0.22um膜过滤或者仔细离心，配体纯度要求在90%以上，分析物的纯度依实验类型而定，进行动力学/亲和力分析时需要90%以上的纯度，进行浓度/特异性测定时可以进混合样品。

缓冲体系选择：大多数缓冲液均适用，可根据样品活性确定最适缓冲液，须新鲜配制，经0.22um膜过滤及脱气。

氨基偶联方法固定配体时，缓冲液中不能含有Tris、甘氨酸等带有伯胺基的物质。

分析物溶液中，不能含甘油、蔗糖、咪唑等高折光率物质，如果样品中含有该物质，可通过缓冲液置换、透析的方法除去。

三、实验操作在该部分中，选用CM5芯片，以配体mouse-anti-human β2-microglobulin antibody和分析物human β2-microglobulin的动力学分析为例进行操作介绍（样品均来自 Biacore getting started kit），实验流程为配体固定-表面测试-再生条件筛选-分析物进样-芯片再生-数据分析。
