摘要:目的 探讨靶向磁性纳米粒子用于肿瘤磁共振分子成像的可行性。方法 将靶向部分重组人促性腺激素释放激素类似物与成像部分超顺磁性氧化铁纳米颗粒连接形成靶向探针,无靶向物质的成像部分作为对照材料。利用体外细胞实验和荷瘤裸鼠体内实验探索探针的靶向结合作用。体外细胞实验采用靶向探针和敏感肿瘤细胞株A549共同孵育,非靶向材料和同种细胞孵育作为对照,细胞洗脱后裂解,磁共振成像测量细胞裂解液的T2值。体内实验将靶向探针和非靶向材料分别注入实验组和对照组荷瘤裸鼠体内,磁共振成像动态检测肿瘤局部组织的信号和T2值变化。结果 体外细胞实验和肿瘤体内实验均证实靶向探针与非靶向颗粒溶液相比,与体外细胞和体内肿瘤组织有更强的结合能力。结论 肿瘤磁共振分子成像可使用靶向磁性纳米粒子作为探针,实现肿瘤的靶向成像,具有良好的发展前景。
