乳腺癌中 caspase-3 和 caspase-9 的表达及其意义

摘要:目的 探讨 caspase-3 和 caspase-9 在乳腺癌发生、发展中的作用。方法 采用免疫组化 SP 法和原位杂交( ISH) 技术检 测 80 例乳腺浸润性导管癌( invasive duct carcinoma,IDC) 、23 例导管内癌( ductal carcinoma in situ,DCIS) 、37 例增生症和 9 例 癌旁组织中 caspase-3、caspase-9 mRNA 和 蛋 白 的 表 达。结 果 caspase-3 mRNA 在 各 组 中 的 表 达 差 异 无 统 计 学 意 义 ( P > 0. 05) 。caspase-3 蛋白在 IDC、DCIS、增生症中的表达均高于癌旁组织( P < 0. 05) ,在 IDC、DCIS 中的表达均高于增生症组( P < 0. 05) 。caspase-9 mRNA 和蛋白在 IDC、DCIS、增生症中的表达均低于癌旁组织( P < 0. 05) ,IDC、DCIS 中的表达均低于增生症 组( P < 0. 05) 。caspase-3、caspase-9 蛋白在 IDC 与 DCIS 中的表达差异均无统计学意义( P > 0. 05) 。caspase-3 蛋白与 mRNA 在 DCIS 和增生症中的表达呈正相关( r = 0. 429,r = 0. 563,P < 0. 05) 。caspase-9 mRNA 与蛋白在各组中的表达均存在相关性 ( r = 0. 94,r = 0. 414,r = 0. 391,r = 0. 827,P < 0. 05) 。caspase-3、caspase-9 mRNA 和蛋白的表达与肿瘤大小等临床病理参数 无相关性( P > 0. 05) 。结论 caspase-3 和 caspase-9 均参与乳腺导管癌的发生、发展。caspase-9 蛋白表达降低,而 caspase-3 表 达增高在乳腺癌的发生、发展过程中较常见; caspase-9 表达降低可能与乳腺组织恶性转化有关。 关键词:乳腺肿瘤; 凋亡; caspase-3; caspase-9 中图分类号: R 737. 9 文献标识码:A 文章编号: 1001 - 7399( 2012) 04 - 0378 - 05
Expression of caspase-3 and caspase-9 and their significance in breast cancer

Methods Eighty cases of breast invasive duct carcinoma ( IDC) ,23 cases of ductal carcinoma in situ ( DCIS) ,37 cases of adenosis and 9 cases of tumor adjacent tissues were collected. The expression of mRNA and protein of caspase-3 and caspase-9 was investigated using immunohistochemical staining and in situ hybridization. Results The expression of caspase-3 protein in IDC,DCIS and adenosis was higher than that in tumor adjacent tissues ( P < 0. 05) ,and that in IDC and DCIS was higher than that in adenosis ( P < 0. 05) . No significant differences of the expression of caspase-3 mRNA were found among IDC,DCIS and源自文库tumor adjacent tissues ( P > 0. 05) . The caspase-9 protein and mRNA expression in IDC,DCIS and adenosis was lower than that in tumor adjacent tissues ( P < 0. 05) ,and that in IDC and DCIS was lower than that in adenosis ( P < 0. 05) . Positive correlation was found between the expression of caspase-3 protein and mRNA in DCIS and hyperplasia ( r = 0. 429,r = 0. 563,P < 0. 05) ,and between the expression of caspase-9 protein and mRNA in all groups ( r = 0. 94,r = 0. 414,r = 0. 391,r = 0. 827,P < 0. 05) . The expression of caspase-3 and9 was unrelated to clinicopathological parameter,such as tumor size( P > 0. 05) . Conclusions Both caspase-3 and caspase-9 are involved in carcinogenesis and progression of breast cancer,which suggests that increased expression of caspase-3 and decreased expression of caspase-9 are common events in breast cancer,and further work is needed to elucidate the exact mechanism.
