基于网络药理学探讨当归四逆汤治疗2型糖尿病的作用机制

(1.广州中医药大学第一临床医学院,广东广州510405;2.广州中医药大学第一附属医院,广东广州510405)摘要:目的通过网络药理学方法探讨当归四逆汤治疗2型糖尿病(T2DM )的作用机制。方法借助中药系统药理学技术平台(TCMSP )获取当归四逆汤主要化学成分、对应靶点及靶标基因,借助人类基因数据库(Gene-Cards )获取2型糖尿病相关靶标基因,将药物靶标基因与疾病靶标基因映射得到当归四逆汤作用于2型糖尿病的预测靶点。利用Cytoscape 3.7.1软件构建中药化合物-靶点网络及蛋白与蛋白相互作用网络(PPI ),利用DA-VID6.8在线工具进行潜在基因的基因本体(Gene Ontology ,GO )分析及基因组百科全书(Kyoto Encyclopedia of Genes and Genomes ,KEGG )通路富集分析。结果当归四逆汤作用于2型糖尿病的有效成分有112个,相关靶点65个,关键靶点16个。GO 分析结果显示,当归四逆汤治疗2型糖尿病潜在基因的生物功能主要涉及细胞凋亡、氧化应激、蛋白结合等;KEGG 通路富集结果显示,当归四逆汤治疗2型糖尿病潜在基因的通路主要涉及缺氧诱导因子(HIF -1)信号通路,胰岛素信号通路,肿瘤坏死因子(TNF )信号通路,胰岛素抵抗,叉头转录因子(FoxO )等信号通路。结论当归四逆汤治疗2型糖尿病是多成分、多靶点、多通路的复杂过程,主要通过参与细胞凋亡、氧化应激、蛋白结合等发挥治疗2型糖尿病的作用。

Abstract :Objective To explore the mechanism of Danggui Sini Decoction in the treatment of type 2diabetes mellitus (T2DM )by means of network pharmacology.Methods Major chemical constituents ,corresponding targets and target genes of Danggui Sini Decoction were obtained by TCMSP ,and target genes related to T2DM were obtained by Gene-Cards.The target genes of drug and disease were mapped to predict the effect of Danggui Sini Decoction on T2DM.Cyto-scape 3.7.1software was used to construct the Chinese medicine compound -target network and protein -protein interaction network (PPI ).Gene Ontology (GO )analysis of potential genes and enrichment analysis of KEGG pathway were carried out using DAVID 6.8online tool.Results There were 112effective components ,65related targets and 16key targets of Danggui Sini Decoction on T2DM.GO analysis showed that the biological functions of Danggui Sini Decoction in the treatment of T2DM were mainly related to apoptosis ,oxidative stress ,protein binding ,etc.KEGG pathway enrichment re-sults showed that the pathway of Danggui Sini Decoction in the treatment of T2DM potential genes mainly involved in hy-poxia inducible factor (HIF -1)signaling pathway ,insulin signaling pathway ,tumor necrosis factor (TNF )signaling path-way ,insulin resistance ,fork transcription factor (FoxO )and other signaling pathways.Conclusion Danggui Sini Decoction is a complex process of multi -component ,multi -target and multi -pathway in the treatment of T2DM.It plays an impor-tant role in the treatment of T2DM by participating in apoptosis ,oxidative stress and protein binding.
