Example of a separation using CD45 (TIL) MicroBeadsWarningsReagents contain sodium azide. Under acidic conditions sodium azide yields hydrazoic acid, which is extremely toxic. Azide compounds should be diluted with running water before discarding. These precautions are recommended to avoid deposits in plumbing where explosive conditions may develop.1. DescriptionThis product is for research use ponents1 mL CD45 (TIL) MicroBeads, mouse:MicroBeads conjugated to monoclonal anti-mouse CD45 antibodies (isotype: rat IgG2b).CapacityFor 10⁹ total cells, up to 100 separations.Product format CD45 (TIL) MicroBeads are supplied in buffercontaining stabilizer and 0.05% sodium azide.StorageStore protected from light at 2−8 °C. Do not freeze. The expiration date is indicated on the vial label.1.1 Principle of the MACS® SeparationFirst, the CD45+cells are magnetically labeled with CD45 (TIL) MicroBeads. Then, the cell suspension is loaded onto a MACS® Column, which is placed in the magnetic field of a MACS Separator. The magnetically labeled CD45+ cells are retained within the column. The unlabeled cells run through; this cell fraction is thus depleted of CD45+ cells. After removing the column from the magnetic field, the magnetically retained CD45+ cells can be eluted as the positively selected cell fraction. To increase the purity, the positively selected cell fraction containing the CD45+ cells must be separated over a second column.1.2 Background informationCD45 (TIL) MicroBeads, mouse have been developed for theisolation of tumor-infiltrating leukocytes (TILs) from single-cell suspensions of solid mouse tumors. The CD45 antigen is expressed on all cells of hematopoietic origin except erythrocytes and platelets.1.3 Applications●Positive selection of CD45+ leukocytes from solid mouse tumors, e.g., B16F10, 4T1, or CT26.WT.1.4 Reagent and instrument requirements●Buffer: Prepare a solution containing phosphate-buffered saline (PBS), pH 7.2, 0.5% bovine serum albumin (BSA), and 2 mM EDTA by diluting MACS® BSA Stock Solution (# 130-091-376) 1:20 with autoMACS® Rinsing Solution (# 130-091-222). Keep buffer cold (2−8 °C). Degas buffer before use, as air bubbles could block the column. Always use freshly prepared buffer. ▲ Note: EDTA can be replaced by other supplements such as anticoagulant citrate dextrose formula-A (ACD-A) or citrate phosphate dextrose (CPD). BSA can be replaced by other proteins such as mouse serum albumin, mouse serum, or fetal bovine serum (FBS). Buffers or media containing Ca2+ or Mg2+ are not recommended for use.●MACS Columns and MACS Separators: CD45+ cells can be enriched by using MS or LS Columns. Protocol2.1 Sample preparationFor preparation of a single-cell suspension from solid mouse tumors use the Tumor Dissociation Kit, mouse (# 130-096-730) in combination with the gentleMACS™ Dissociators. For details refer to /protocols.▲ Dead cells may bind non-specifically to MACS® MicroBeads. To remove dead cells, we recommend using the Dead Cell Removal Kit (# 130-090-101).2.2 Magnetic labeling▲ Cells can be labeled with MACS MicroBeads using the autolabeling function of the autoMACS® Pro Separator. For more information refer to section 2.4.▲ Work fast, keep cells cold, and use pre-cooled solutions. This will prevent capping of antibodies on the cell surface and non-specific cell labeling.▲ Volumes for magnetic labeling given below are for up to 10⁷ total cells. When working with fewer than 10⁷ cells, use the same volumes as indicated. When working with higher cell numbers, scale up all reagent volumes and total volumes accordingly (e.g. for 2×10⁷ total cells, use twice the volume of all indicated reagent volumes and total volumes).▲ For optimal performance it is important to obtain a single-cell suspension before magnetic labeling. Pass cells through 30 µm nylon mesh (MACS SmartStrainers (30 µm), # 130-098-458). Moisten filter with buffer before use.▲ The recommended incubation temperature is 2–8 °C. Higher temperatures and/or longer incubation times may lead to non-specific cell labeling. Working on ice may require increased incubation times.1. Determine cell number.2. Centrifuge cell suspension at 300×g for 5 minutes. Aspiratesupernatant completely.3. Resuspend cell pellet in 90 µL of buffer per 10⁷ total cells.▲ Note: Always use freshly prepared buffer.4. Add 10 µL of CD45 (TIL) MicroBeads per 10⁷ total cells.5. Mix well and incubate for 15 minutes in the dark in therefrigerator (2−8 °C).6. (Optional) Add staining antibodies according tomanufacturer’s recommendations.7. Add buffer to a final volume of 500 μL for up to 5×10⁷ cells.▲Note: If more cells were used, split the sample onto multiple columns duringmagnetic separation.▲Note: For higher cell numbers, scale up buffer volume accordingly.8.Proceed to magnetic separation (2.3).2.3 Magnetic separation▲ Choose an appropriate MACS Column and MACS Separator according to the number of total cells and the number of CD45+ cells. For details refer to the table in section 1.4.▲ Note: MS Columns are recommended for highest purity of CD45+ cells. LSColumns are recommended for highest recovery of CD45+ cells.▲ For optimal performance it is important to obtain a single-cell suspension before magnetic separation. Pass cells through 30 µm nylon mesh (Pre-Separation Filters (30 µm), # 130-041-407) to remove cell clumps which may clog the column. Moisten filter with buffer before use.▲Always wait until the column reservoir is empty before proceeding to the next step.Magnetic separation with MS or LS Columns1. Place column in the magnetic field of a suitable MACSSeparator. For details refer to the respective MACS Columndata sheet.2. Prepare column by rinsing with the appropriate amount ofbuffer:MS: 500 µL LS: 3 mL3. Apply cell suspension onto the column. Collect flow-throughcontaining unlabeled cells.4. Wash column with the appropriate amount of buffer. Collectunlabeled cells that pass through and combine with theflow-through from step 3.MS: 3×500 µL LS: 2×1 mL▲ Note:Perform washing steps by adding buffer aliquots as soon as the columnreservoir is empty.5. Remove column from the separator and place it on a suitablecollection tube.6. Pipette the appropriate amount of buffer onto the column.Immediately flush out the magnetically labeled cells by firmly pushing the plunger into the column.MS: 1 mL LS: 3 mL7. Flexitallic Thermiculite 870产品数据表说明书

Flexitallic Thermiculite 870产品数据表说明书

PRODUCT DATASHEETThermiculite ®870 is a high temperature sealing material designed for low stress sealing in solid oxide fuel cells. It is based on the mineral vermiculite and contains no organic components.870Service:Thermiculite ®870 is specifically designed for use in solid oxide fuel cells (SOFC) and solid oxide electrolysers (SOE) and has a high degree of compression combined with good sealing under low applied surface stresses. Thermiculite ®870 contains only two minerals, vermiculite and steatite. There are no organic materials or other elements (such as phosphorous or sulphur) that would contaminate SOFC or SOE cell chemistry.Gasket Test Data:For some SOFC designs a softer, more compressible sealing material is required.TH870 is significantly more compressible under low loads, e.g. 1MPa. TH870 is around ten times more compressible than TH866.Figure 1(below) shows load vs defection for TH870 compared to TH866.At a stress of 1 MPa TH870 compresses 0.22mm whereas TH866 gasket onlycompresses 0.02mm under the same load.Availability:Thermiculite ®870 is available in sheet form:Standard sheet size: 450mm x 350mm Standard thicknesses:0.5mm, 0.7mm and 1.0mm**Please note that these nominal thicknesses are provided as a guide. As the material is very soft the measured thicknesses may differ slightly due to compression during measurement.Special thicknesses between 0.3mm and 1.0mm are available on request.Flexitallic can provide a gasket cutting service, ideally a .dxf file of the gasket shape would be provided by the customer.Flexitallic will review the dimensions and design and advise if cutting is feasible and provide a quote. Sample material available on request. This Data Sheet refers to the material as supplied. The information contained herein is given in good faith, but no liability will be accepted by the Company in relation to same.We reserve the right to change the details given on this Data Sheet as additionalinformation is acquired. Customers requiring the latest version of this Data Sheet should contact our Applications Engineering Department. The information given and, in particular, any parameters, should be used for guidance purposes only. The Company does not give any warranty that the product will be suitable for the use intended by the customer.Health & SafetyFor further Health and Safety information please see the relevant Material SafetyPRODUCT DATASHEETThermiculite®870 also seals at least aseffectively as TH866, but at much lowerapplied loads. This is the case at ambientand elevated temperatures. Figure 2(below) shows an 80 hour leakage testat ambient temperature.Table 1 (right) shows the effect ofgasket thickness on leak rate for TH870gaskets. It is recommended that the thinnest material is chosen after taking account of other requirements e.g. flange surface imperfections, spacing requirements between cells or for the gasket to compensate for tolerances throughout the stack.TH870 Thickness Leak rate after 100 hours (mm)(ml/min/m)0.4 2.0360.7 2.6211.0 3.525TABLE 1.Leak Rate Variation with Gasket Thickness. S T U DI OC ON DE NS ER MI CR OP HO N EB -2P R OVersion 1.0 September User s ManualSTUDIO CONDENSER MICROPHONE B-2 PRO1.SAFETY PRECAUTIONSDetailed safety precautions:Please read all safety precautions and operating instructions before you attempt to operate the unit. Keep all safety precautions and operating instructions for future reference.Water and moisture:Condenser microphones are extremely moisture-sensitive. Never use your microphone in close proximity to water (e.g. bath-tubs, wash-basins, sinks, washing machines, pools, etc.). When doing voice recordings, always use the enclosed popscreen to safely protect the capsule from moisture.Power supply:Always use the power supply specified in the operating instructions.Damage:Take care not to drop your microphone as this can lead to severe damage. BEHRINGER assumes no liability for any damage caused by the user.Maintenance and care:When the microphone is not in use, please take it out from its elastic suspension, clean it with a soft cloth, and then place it in its case. Place the bag containing moisture-absorbing crystals close to the microphone head.Caution:Never dismount the screen holding the capsule, as this can damage the microphone! Never try to modify the printed circuit board inside your B-2 PRO! This can lead to microphone damage that cannot be repaired. Any such attempt will void the warranty.2.POWER SUPPLYYour condenser mic B-2 PRO needs a +48 V phantom power supply. BEHRINGER assumes no liability for any damage caused by a defective phantom power supply. Always mute the sound reinforcement system before you switch on the phantom power supply. After power-up, the B-2 PRO needs about 6 seconds to stabilize.3.DIRECTIVITYWith the switch on the front, you can set your B-2 PRO to provide a cardioid, omnidirectional or figure eight directivity pattern. If the microphone is used to pick up both the signal source and ambient signals, we recommend to use an omnidirectional directivity (switch position: left). However, to pick up specific instruments or voices, please set the switch to the cardioid position (right). The figure eight directivity pattern (center) is recommended, for example, for choir miking. Thanks to the figure eight directivity pattern, your B-2 PRO picks up more ambient signals than when it is set to the omnidirectional directivity.4.LOW-CUT FILTER AND LEVEL ATTENUATIONThe low-cut filter can be activated with the left switch on the rear, in order to filter low-frequency interference such as pop sounds, etc. With the low-cut filter on, the B-2 PRO provides an almost linear frequency response with signals picked up at close proximity to the source. The built-in shielding minimizes the microphone s sensitivity to high-frequency interference.Use the right switch on the rear to activate the -10 dB level attenuator, which should be used with pulse-type signal sources producing high sound pressure levels (e.g. kick drum).21.SAFETY PRECAUTIONS3STUDIO CONDENSER MICROPHONE B-2 PRO5.MOUNTING MICROPHONE AND ELASTIC SUSPENSION 5.MOUNTING MICROPHONE AND ELASTIC SUSPENSIONAttach the enclosed elastic suspension to the microphone stand. Fasten the screw, once the suspension has been positioned properly. Open the elastic suspension by pressing the two circular levers, then insert the B-2 PRO from above.6.AUDIO CONNECTIONUse a balanced XLR microphone cable with the following pin assignment: pin 1 = shielding;pin 2 = +; pin 3 = -. Since your B-2 PRO features gold-plated contact points throughout, we recommend that you use only microphone cables with gold-plated connectors.7.LEVEL SETTING / ADJUSTING THE BASIC SOUNDAdjust the gain control in the microphone channel of your mixing console so that the peak LED lights up only occasionally or never at all. The EQ controls in the microphone channel should be set to mid-travel position to start with; low-cut filter and level attenuator should be off. Try to achieve the desired sound by experimenting with the microphone position. Use the omnidirectional, cardioid and figure eight directivity patterns (see chapter 3). Often, it will be useful to set up acoustic barriers ( gobos ) at various angles towards the signal source. Only when the desired basic sound has been achieved, should you start to use equalizers and signal processors, if any at all (Remember: less is often more!)The B-2 PRO provides a level peak around 12 kHz producing some kind of presence in this range;so, there is no need for high-frequency EQing which could deteriorate the signal and raise the overall noise floor. On the contrary, the B-2 PRO provides that much-desired transparency which often gets lost during recording and mixing.8.WARRANTY +Please find our current warranty terms on our web site under orsendusarequestbye-mail(********************),fax(+49(0)2154920665),or telephone (+49 (0) 2154 920666).9.SPECIFICATIONSTransducer type:condenser, 1" dual-diaphragm Operating principle:pressure gradient Polar pattern:cardioid, omnidirectional or figure eight Connection:gold-plated balanced XLR connector Open circuit sensitivity (at 1 kHz):cardioid: -36 dBV (0 dBV = 1 V/Pa), 16 mV/Pa omnidirectional: -37 dBV (0 dBV = 1 V/Pa), 14 mV/Pa figure eight: -35 dBV (0 dBV = 1 V/Pa), 18 mV/Pa Frequency response:20 Hz - 20 kHz Level attenuation:-10 dB (switchable)Low-Cut filter: 6 dB/oct. at 150 Hz (switchable)Max. You have the choice of either the fine directional fixture for specimen clamps with zero indicators and click stops every 2° or the directional fixture for specimen clamps, with 8° XY orientation. Both orientation systems can be equipped with the quick clamping system, allowing rapid specimen clamp exchange or clamp removal for cleaning.• I f you prefer a fixed and stable object head, the rigid fixture (with the integrated quick clamping system) is the right choice for you.• A re you going to section specimens embedded in standard or super size cassettes or paraffin blocks? Do you want to cool your standard cassettes while sectioning to keep the block colder longer, especially when sectioning serial sections? Find the appropriate clamp for your specimen in the cassettes or blocks section.• D epending on whether you prefer disposable blades or reusable knives, select the blade or knife holder. The 2-in-1 Blade Holder (to be used with the blade holder base) can be used with either high or low profile blades.• K nife holder E with a trough is used with the knife holder base and low profile blades. The trough can be filled with water and the sections will stretch on the water surface and can be collected with an object slide.• W hen using reusable c or d profile knives, Knife Holder N is the best choice.• T he Knife Holder Base (for Knife Holder N and E with a trough) can be equipped with a backlighting system, which helps you to better align the specimen to the knife.• A large variety of disposable blades or reusable knives can be found in the consumable section of the Website.Raw data from a E15,5 stage mouse embryo COURTESY OFCONFIGURE YOUR OWN MICROTOME FOR SEMI-THIN SECTIONING AND 3D APPLICATIONSSELECT YOURMICROTOME Motorized14 0524 58261ORIENTING ORNON-ORIENTING FIXTURE FOR SPECIMEN CLAMPS Rigid14 0502 38160Fine Directional14 0502 37717Directional14 0502 38949Quick ClampingSystem14 0502 37718APPLICATIONSegment Arc(direct assembly)14 0502 40314Segment Arc(indirect assembly)14 0502 299693D SEMI-THIN SECTIONINGRound SpecimenHolder with 3 in-serts: 6, 15 and25 mm ø14 0502 38002KnifeHolder Base14 0502 37962EM SpecimenHolder Fixture14 0502 29968EM Flat SpecimenHolder14 0355 10405EM UniversalSpecimen Holder14 0356 10868Special Wrench14 0356 10869Backlighting14 0502 38719Knife Holder GD14 0502 39052Diamond KnivesTriangular GlassKnivesMicroscopeCarrier with LEDHi Power Spotsand Microscope14 0502 38463CombinedCarrierwith Magnifierand Illumination14 0502 29971120/60Hz14 0502 29972230V/50HzLED 1000Hi-PowerSpots, 2 Arms14 6000 04826LED 1000Control Unit14 6000 04825Foot Switch14 0502 38257OTHER ACCESSORIES Knife HolderE-TC14 0502 37997DisposableTungsten CarbideBlades TC-6514 0216 26379NANOCUT R8#RecommendationOrder No.NANOCUT R Basic Instrument 14 0521 582611Fine-Directional Fixture for Specimen Clamps 14 0502 377172Quick Clamping System 14 0502 377183EM specimen holder fixture 14 0502 299684EM holder flat or 14 0355 104055EM holder universal 14 0356 108686Special wrench 14 0356 108697Knife holder GD 14 0502 390528Diamond knifeN/A9Microscope carrier w/LED Hi-Power spots 14 0502 3846310LED 1000 control unit 14 6000 0482511Foot switch14 0502 38257#RecommendationOrder No.NANOCUT R Basic Instrument 14 0521 582611Fine-Directional Fixture for Specimen Clamps 14 0502 377172Quick Clamping System 14 0502 377183Round specimen holder 14 0502 380024Knife holder base 14 0502 379625Backlighting14 0502 387196Knife holder E-TC for TC-65 disposable blades 14 0502 379977TC-65 disposable blades 14 0216 263798Foot switch14 0502 38257RECOMMENDED SOLUTION FOR 3D RECONSTRUCTIONRECOMMENDED SOLUTION FOR SEMI-THIN SECTIONING123456123561147898103D APPLICATION – FOLLOW THE MERLOT DECISION TREE• T he NANOCUT R is the instrument of choice for 3D reconstruction with its precise stop in the object head’s optimal position for photo-capture of the cut sample surface when running 3-dimensional reconstruction programs.•C hoose whether you would like to use a specimen orientation (fine directional or directional) or prefer a fixed rigid object head for extra stability.• T he quick clamping system will allow you to quickly remove the specimen clamp.• A s most specimens are embedded using molds with round specimen clamp adapters, the use of a round specimen clamp is recommended. • F or 3-dimensional reconstruction programs, high qualiy surfaces of the cut block surface are required. This can be obtained by using disposable tungsten carbide TC-65 blades, which are clamped in the E-TC blade holder.SEMI-THIN SECTIONING – FOLLOW THE GREEN DECISION TREE• T he NANOCUT R is the instrument of choice for semi-thin sectioning with a minimum section thickness of 250 nm. The use of a footswitch is recommended allowing you to have both hands free for the section preparation.• T he specimen size for semi-thin sectioning is limited. Specimens are embedded in resin and will be clamped in either an EM flat or universal specimen holder. The specimens are clamped in the holders using the special wrench.• O ften a segment arc is used to clamp the EM holders – you have the choice to either select the segment arc, which is directly clamped onto the cylinder of the microtome, which gives you the most stability, or the segment arc with indirect assembly, which is clamped on either the fine directional or directional fixture for specimen clamps.• S emi-thin sections can be produced using either glass or diamond knives, which are clamped in the Knife holder GD. Backlighting is recommended to help better align the specimen to the knife. • L ED illumination and the microscope will help you to prepare and collect the section ribbons.SELECT YOURMICROTOMEORIENTING OR NON-ORIENTING FIXTURE FOR SPECIMEN CLAMPSSPECIMEN SIZE AND FORMATSPECIMEN HARDNESS SECTION OR SURFACEDESIRED MINIMAL SECTION THICKNESSOTHER ACCESSORIESMotorized14 0523 58241Motorized14 0254 58261Rigid14 0502 38160Fine Directional14 0502 37717Directional14 0502 38949Quick ClampingSystem14 0502 37718Standard SpecimenClamp 40x40 mm14 0502 37998Standard SpecimenClamp 50x55 mm14 0502 38005Round SpecimenHolder with 3inserts: 6 mm,15 mm and25 mm diameter14 0502 38002EM FlatSpecimen Holder14 0355 10405EM SpecimenHolder Fixture14 0502 29968EM UniversalSpecimen Holder14 0356 10868Foil Clamp Type 114 0402 09307V-Insert to clampround specimens14 0502 38000Special Wrench14 0356 10869Knife Holder GD14 0502 39052Knife Holder NZ14 0502 37994Knife Holder E-TC14 0502 379972-in-1 BladeHolder for Highand Low ProfileBlades14 0502 54497Knife Holder Base14 0502 37962Backlighting14 0502 38719Blade Holder Base14 0502 55546Reusable TungstenCarbide KnivesProfile c, 16 cm:14 0216 04206Profile d, 16 cm:14 0216 04813DisposableTungsten CarbideBlades TC-6514 0216 26379for complete disposableblade portfolioUniversalMicroscopeCarrier14 0502 40580Foot Switch14 0502 38257DiamondKnives TriangularGlass KnivesMicroscopeCarrier with LEDHi Power Spotsand Microscope14 0502 38463Magnifier 2 fold14 0502 42790CombinedCarrier withMagnifier andIllumination14 0502 29971120/60Hz14 0502 29972230V/50HzLED 1000Control Unit14 6000 04825LED 1000Hi-Power Spots,2 Arms14 6000 04826AUTOCUT R NANOCUT RSTANDARDSPECIMEN CLAMPSROUNDSPECIMENS1 μm2 μm 5 μm Max. 5 μmto 250 nm HARD E.G.
