STAT3 在乳腺癌中的表达及临床意义

摘要 目的: 探讨细胞信号传导和转录激活因子 3 (STAT3) 在乳腺癌及癌旁组织中的差异性表达及 与乳腺癌其他临床病理指标的相关性, 并分析 STAT3 表达与乳腺癌者生存数据的关系。方法: 收集我 院行乳腺癌根治术的存档蜡块 130 例, 癌旁组织标本 61 例。采用免疫组化法检测组织中 STAT3 的表达 情况, 比较表达率的差异, 并分析乳腺癌患者 STAT3 表达与其他临床病理指标间的相关性。结果: 乳腺 癌组织中 STAT3 的阳性表达率显著高于癌旁组织。 STAT3 的阳性表达与临床分期、 肿瘤大小、 淋巴结 转移、 ER 及 PR 表达情况密切相关; 但与年龄、 组织学分级及 Her⁃2 表达无显著相关性。生存分析结果 显示, STAT3 高表达的患者较低表达患者预后差。 Cox 回归模型分析则显示, PR 及 STAT3 均为乳腺癌 患者独立的预后因子。结论:STAT3 在乳腺癌的发生、 发展中发挥重要作用, 是乳腺癌患者独立的预 后因子。 关键词 STAT3 ; 免疫组化; 乳腺癌 Expression level and clinical features of STAT3 in breast cancer WANG Qingtao* ,HUANG Haiyang ,ZOU Qingfeng , DENG Xinghui. *Jiangmen Central Hospital , Jiangmen 529000 , China 【Abstract】 Objective To detect the expression of STAT3 in breast cancer and adjacent paracancerous tissues ,then analyze the correlationship between its expression level and some other clinical features of breast cancer ,and to evaluate its significance in prognosis of patients with breast cancer. Methods 130 breast cancer patients were enrolled in the Jiangmen Central Hospital ,and 61 cases of adjacent normal tissues. The expression of STAT3 was detected by IHC. We compared the expression of STAT3 between the two groups and analyzed the relationship between STAT3 and other clinical features. Results The expression of STAT3 in breast cancer group was much higher than the adjacent paracancerous tissues. STAT3 expression showed obvious correlationship with clinical stage ,tumor size ,lymph node metastasis ,the expression of ER and PR ,but no statistical correla⁃ tion with age ,histological grade and Her⁃2 expression. Survival analysis showed that patients with high STAT3 expression level had a significantly poor prognosis. The Cox regression analysis showed that PR and STAT3 expres⁃ sion were independent prognostic factors for patients with breast cancer. Conclusion These results may implicate that STAT3 will promote the development of breast cancer and higher expression of STAT3 indicates the poor survival rate. 【Key words】 STAT3 ; Immuneohistochemisty ; Breast cancer
