仪器操作程序
1600、1800 仪器的安装非常简单,请您按照一下步骤操作: 1、打开仪器包装箱,取出仪器,平稳放在工作台上。 2、连接仪器电源线,打开电源开关开启主机,如图 6.1 所示,点击分光光度计应用软件,仪器进入初 始化状态,如图 6.2 所示。

mat封闭MSA1平板,上下轻微颠倒平板十次;9.将MSA1平板放置在4℃下30min;10.如果即将进入重悬MSA1步骤,现在可以取出RA1放在室温下解冻(RA1不易解冻,解冻后含有少量沉淀,需要摇匀溶解);11.4℃下3000g离心MSA1平板20min(离心机只能达到2200g,将离心时间延长至30min),迅速取出MSA1平板,可见样品孔中有蓝色沉淀;迅速进入下一步骤,避免沉淀重悬;12.弃掉cap mat13.迅速弃掉样品孔中溶液,可以用手瞬间甩出样品孔里的溶液,再将MSA1平板连续多次倒扣于吸水纸上,直至样品孔中没有溶液流出为止;14.将没有封口的MSA1平板保持倒置,放于试管架上,在室温下干燥1hr;可见样品孔底部蓝色沉淀;15.进入下一步实验,或者用cap mat封闭平板,保存在-20℃;重悬MSA1需要RA1(-20℃保存,illumina)Heat sealer准备将Hybridization Oven温度调至48℃预热;打开heat sealer预热,大约需要20min;室温下融解RA1,颠倒使沉淀溶解;实验1.每个样品孔加入46μl RA1;2.用foil heat seal封闭MSA1平板,将foil heat seal压在MSA1平板上,置于heat sealer中,用sealing block紧紧压在foil heat seal上持续5sec,再将MSA1平板水平旋转180°,再次用sealing block紧紧压住持续5sec,迅速进入下一步骤;3.取出MSA1平板,快速用工具压过整个MSA1平板表面,使每一个样品孔表面都有少许凹陷,确定样品孔完全密封;若平板表面温度在密封好之前已经下降,可以用sealing block 再一次加热;4.将密封好的MSA1平板放在Hybridization Oven里48℃反应1hr;5.用vortexer 1800rpm振荡MSA1平板1min;6.280g短暂离心;如果MSA1平板之前保存在-20℃,可以重复振荡和离心步骤直至沉淀完全悬浮;7.进入下一步实验(可在室温下短暂保存)或者在-20℃保存(超过24hr保存需放置在-80℃);芯片杂交需要PB2(室温,illumina)BeadChipsHyb ChambersHyb Chamber lidHyb Chamber gasketsHyb Chamber inserts准备将Heat block调至95℃预热;将Hybridization Oven温度调至48℃预热速度调至5;实验1.将Hyb Chamber gaskets放入Hyb Chamber;2.在Hyb Chamber中,给每一个需要用的保湿槽加入400μl PB2;3.用Hyb Chamber lid盖住Hyb Chamber防止PB2蒸发;4.将重悬后的MSA1平板放在95℃的heat block上20min变性;5.从4℃中取出BeadChips但暂时不用拆封;6.从heat block取下MSA1平板,放置在室温下30min;7.280g短暂离心MSA1平板;8.小心去除BeadChips外包装;小心:避免触碰芯片中心条纹区域和边沿上样孔;9.将BeadChips放入Hyb Chamber insert,使芯片的条形码与Hyb Chamber insert上的条形码区域方向一致;10.从MSA1平板中取42μl DNA样品,沿芯片上样孔缓慢加入芯片,此过程保持移液器垂直于芯片,并在上样孔保留一个小液滴;可以额外增加上样的样品以保证在上样孔形成液滴;11.按照图示顺序完成所有样品上样;上样完成后等待样品溶液扩散并浸没所有芯片表面;12.确定Hybridization Oven温度升至48℃;13.将Hyb Chamber insert放入Hyb Chamber,使芯片条形码方向与Hyb Chamber中条形码区域方向一致;14.盖上Hyb Chamber lid,用夹锁固定;15.将Hyb Chamber平稳的移至48℃的Hybridization Oven中,全程避免振荡和摇晃,并且保证Hyb Chamber在Hybridization Oven中为横向放置;16.开启Hybridization Oven的晃动开关,在48℃中温育至少16hr,但不超过24hr;可以丢弃MSA1平板;为下一步准备XC4溶液1.XC4试剂瓶中加入330ml 100%乙醇;2.剧烈振荡15sec(可手动);3.室温下放置过夜;若没有放置过夜,加完乙醇后可放置在振荡器上剧烈振荡,等待使用;4.使用前再次剧烈振荡,确保没有可见图层出现;清洗芯片需要PB1(illumina)Multi-sample BeadChip Alignment Fixture and Alignment barTe-Flow Flow-Through Chambers(black frames, spacers, glass back plates, clamps)Wash Dish(glass)Wash Rack准备从Hybridization Oven中取出Hyb Chamber,室温下冷却25min;给两个Wash Dish中分别加入200ml PB1,并标记为PB1;给BeadChip Alignment Fixture中加入150ml PB1;去掉spacers的白色贴纸;清洗glass back plates;实验1.安装好Wash rack的把手,并把Wash rack浸泡入第一个Wash dish中的PB1溶液里;2.取出Hyb Chamber insert,并取出BeadChips;3.揭开粘在BeadChips表面的IntelliHyb Seal;注意:IntelliHyb Seal粘性较强,一手捏紧BeadChips,另一手从条形码一边揭起膜,一次性、缓慢、连续揭开IntelliHyb Seal,一定避免停顿和动作的反复,会给芯片表面留下印迹;seal的胶会在BeadChip边沿留下残余,可以用无粉纸巾吸少许乙醇小心擦拭,一定避免接触BeadChip中心区域;4.快速将BeadChips放入浸泡在PB1中的Wash rack里,确保整张BeadChips都浸没PB1;5.上下缓慢提放Wash rack 1min,每次都使BeadChips完全离开PB1液面或者完全浸没液面;6.将Wash rack移入另一个Wash dish中,确保BeadChips完全浸没PB1;7.重复步骤5;安装Flow-through Chamber1.对于每一个BeadChip,都需要一个black frame放入事先乘好150ml PB1溶液的BeadChipAlignment Fixture;2.将BeadChip从Wash rack中取出,放入black frame,条形码一头与BeadChip AlignmentFixture中有条纹一侧一致;每一个BeadChip都确保完全浸没PB1;3.仔细检查确保BeadChip表面没有杂物污染;4.取一个干净的spacer(透明)放置于BeadChip之上;5.将Alignment bar装在Alignment Fixture远离BeadChip的条形码一侧;6.取一个干净的glass back plate(避免与BeadChip接触面有任何尘埃污染),放置于BeadChip和spacer之上,使glass back plate的缺口在条形码一侧,面朝BeadChip形成一个漏斗型上样槽,并使glass back plate的另一端紧贴Alignment bar;7.用两个clamps固定每一个Flow-Through Chamber;8.取出Flow-Through Chamber,用剪刀剪去spacer的边沿,需要保留条形码区域;9.在可以进入下一步实验之前用蒸馏水清洗Flow-Through Chamber的点样槽以确保不再有PB2溶液残留;注意:需要去除PB2溶液对后续反应的影响;荧光反应需要RA1(-20℃,illumina) 1~8张BeadChips需10mlXC1(-20℃,illumina)XC2(-20℃,illumina)TEM(-20℃,illumina)XC3(Room temperature,illumina) 1~8张BeadChips需50mlSTM(-20℃,illumina)PB1(Room temperature,illumina) 1~8张BeadChips需300mlXC4(Room temperature,illumina) 1~8张BeadChips需300mlEtOH(Room temperature)95% formamide/1 mM EDTA(14.25ml formamide+0.75H2O+30μl 0.5M EDTA)准备尽早取出RA1融解;室温下融解所有试剂;尽早设置water circulator温度为44℃;注意:水浴精确温度很难稳定,需尽早开启等待其平衡;使用前摇晃Chamber Rack使里面的气泡排出,能够帮助水浴温度准确稳定;使BeadChip的Flow-Through Chamber与检测温度的temperature probe相邻放置在Chamber Rack里并靠近Chamber Rack的进水口,可以使测量的温度更准确稳定;实验注意:下面的步骤不允许中断单碱基延伸1.当Chamber Rack温度达到44℃,将Flow-Through Chamber装入Chamber Rack,使上样槽向上;2.剧烈摇晃XC4使溶液完全溶解;3.在每一个Flow-Through Chamber中,沿上样槽加入:a)150μl RA1,静置30sec,再重复5次;b)450μl XC1,静置10min;c)450μl XC2,静置10min;d)200μl TEM,静置15min;e)450μl95% formamide/1 mM EDTA,静置1min,再重复一次;f)静置5min;g)将Chamber Rack的温度调节至STM管子所写温度,若没有写明则调节至37℃;h)450μl XC3,静置1min,再重复一次;4.等待Chamber Rack的温度达到正确温度;荧光标记1.如果计划实验后立即上机扫描BeadChips,现在需要打开HiScan;2.在每一个Flow-Through Chamber中,加入:a)250μl STM,静置10min;b)450μl XC3,静置1min;再重复一次,静置5min;c)250μl ATM,静置10min;注意:ATM和STM都比较粘稠,如果溶液流过Flow-ThroughChamber的速度比较慢可以多等待几分钟时间;d)450μl XC3,静置1min;再重复一次,静置5min;e)250μl STM,静置10min;f)450μl XC3,静置1min;再重复一次,静置5min;g)250μl ATM,静置10min;h)450μl XC3,静置1min;再重复一次,静置5min;i)250μl STM,静置10min;j)450μl XC3,静置1min;再重复一次,静置5min;3.取出Flow-Through Chamber,静置于试验台;清洗和包埋1.给Wash dish加入310ml PB1(300ml即可);2.将staining rack放入PB1溶液中,使locking arm面对自己;3.用dismantling tool打开Flow-Through Chamber上的clamps,去掉glass back plate和spacer,取出BeadChip,并将BeadChip放入PB1里的staining rack上;使BeadChip的条形码面对远离自己的方向;4.上下缓慢提放staining rack10次,每次都使BeadChips完全离开液面或者完全浸没液面;5.浸没5min;注意:不要让BeadChips在PB1中浸泡超过30min;6.剧烈摇晃XC4使完全溶解;7.给另一个Wash dish中加入300ml XC4;注意:不要让XC4静置超过10min;8.将staining rack放入XC4中;9.上下缓慢提放staining rack10次,每次都使BeadChips完全离开液面或者完全浸没液面;10.浸没5min;11.取出staining rack,放置于试管架或者试验台;注意:确保staining rack的locking arm连接稳固;12.用镊子小心夹住每一片BeadChips的条形码区域取出,放置于一个与真空干燥器大小匹配的试管架上,使条形码朝上且指向自己;13.将所有BeadChips放在vacuum desiccator里抽真空干燥1hr,气压为508mm Hg(0.68bar);14.用酒精和无尘吸水纸清理BeadChips背面;注意:BeadChips背面的污染物会使BeadChips放入HiScan时无法保持水平,从而使HiScan无法扫描到BeadChips荧光信号;15.清洗所有实验用具;上机扫描荧光信号需要DMAP fileManifest fileClustal file准备用DMAP Download Client Utility下载DMAP file实验。

HTRT-1600本系统主要由高精度高稳定度的小电流源、高精度AD采样芯片以及嵌入式芯片,上位机智能管理分析软件、真空多段温控加热设备组成的超高温导电电阻率智能存储分析仪器。

测试导电金属样品根据测试工艺要求任意设定加热、,通过过程中电阻率的精密测量,完成对金属内部结构变化如相变、碳化物、固溶度等特性的分析。

设备即可直接通电加热应用于石墨自发热,又可以外加高温环境气氛进行测试,是目前研究导电材料重要设备,也是高等院校和生产检测单位的重要科研设备。

一、主要性能:导电材料的电特性主要用电阻率表征。

影响电阻率的因素有温度、杂质含量、冷变形、热处理等。

温度的影响常以导电材料电阻率的温度系数表示。

除接近熔点和超低温以外,在一般温度范围,电阻率随温度变化呈线性关系。

二、复合型高分子导电材料,由通用的高分子材料与各种导电性物质通过填充复合、表面复合或层积复合等方式而制得。

主要品种有导电塑料、导电橡胶、导电纤维织物、导电涂料、导电胶粘剂以及透明导电薄膜等。

石墨能够作为导电材料,其根本原因在于其独特的结构和电子性质。

石墨是碳的一种同素异形体,它由一层层的碳原子平面组成,每层被称为石墨烯。

在这些平面内,碳原子以sp^2杂化轨道形成六角形蜂窝状结构,每个碳原子与其三个最近邻的碳原子通过σ键相连,形成一个非常稳定的二维平面。
