磁力架说明书_Magnetic Stand Manual_MCE
1包装清单产品概述MCE 磁力架是磁珠产品专用配套设备,支持 MCE 全线磁珠类产品,内含强磁磁芯,可实现快速高效的分离。
MCE 磁力架 (200 μL / 2 mL / 15 mL ) 采用独特的三明治槽设计,磁条可抽出,可容纳 200 μL PCR 管,1.5 mL EP 管,2 mL EP 管,15 mL 离心管。
本产品适用于抗体纯化、免疫沉淀 (IP )、免疫共沉淀 (Co-IP )、细胞分选和核酸分离等实验。
2操作说明31. 将装有磁珠悬液的 EP 管/离心管置于磁力架对应的样品孔中,静置数分钟后磁珠被吸附聚集于管壁,溶液恢复澄清。
2. 用移液器或吸管从管底将溶液吸出,或小心倾倒出液体。
3. 抽去磁力条,加入复溶液体,轻缓震荡即可混合均匀,进行下一步操作。
注:磁性分离的时间与磁珠粒径有关,磁珠粒径越小,磁性分离时间越长。
此外,溶液的黏稠程度以及溶液的成分也会对磁性分离时间产生影响。
5注意事项1. 根据实验参数和样品体积不同,可调整磁芯位置或选用不同样式的磁力架。
2. 为减少操作过程中磁珠的损失,请将样品管底端插入磁力架底部的凹槽内。
当磁珠吸附在管壁上后,缓慢倾去上清,或用移液枪吸尽。
3. 由于磁力架有强大的磁场,请远离手机、电脑、手表、起博器、磁铁等易被磁力干扰的物体,尤其是刀具,以免对操作人员造成伤害。
4. 如需同时使用多个磁力架 (≥2 个) ,应分开放置,避免磁场之间产生干扰。
不要把多个磁棒放在一起,以防止夹伤。
5. 请勿与强酸、强碱等腐蚀性溶剂直接接触。
6. 请勿拆卸磁块。
7. 为保护外壳,请勿长时间暴露在阳光和紫外线下。
8. 为保持磁力架磁性,请勿置于高温和强外界磁场环境中。
9. 使用后请及时清洁,妥善放置在干燥环境中。

溶解于 DMSO。为了获得更高的浓度,可以将离心管在 37℃加热 10 分钟和/或在超声波浴中震荡一段时间。原液可以在-20℃以下 储存几个月。
35 nM,3、12、24、48 和 72 小时
用 35 nM 的 UM171 处理 3、12、24、48 和 72 小时,在每个时间 点裂解细胞,提取 RNA 并测序。UM171 显著抑制与红系和巨核 细胞分化相关的转录物。在 UM171 处理的细胞中,只有 6-7 个 基因是普遍上调或下调的,上调幅度最大的基因编码表面分子。 这些基因包括 PROCR(也称为 EPCR 或 CD201),是小鼠 LT-HSCs 的标志物。

将 UM171 或 DMSO 处理的 CD34+ CB 细胞注射到 NSG 小鼠中,在 约 300 只小鼠中评估人细胞的植入水平,并以热图的形式表示。 数据分析表明人类重组的两个新兴模式,一个来自主要的 lymphomyeloid LT-HSCs,在高细胞剂量时出现,另一个来自 LT-HSCs,具有淋巴缺陷分化的表型,主要在 UM171 处理中观察 到。B 淋巴细胞的增殖与 lymphomyeloid LT-HSCs 的频率或数量均 不受 UM171 影响。UM171 对 LT-HSC 的影响在移植后 30 周仍然 存在,在高细胞剂量时其多向贡献明显。

10074-G5抑制Daudi Burkitt淋巴瘤细胞的生长并破坏c-Myc / Max二聚化。针对Daudi和HL-60细胞的IC50值分别为15.6和13.5μM[1]。10074-G5在区域Arg363-Ile381中结合Myc肽Myc353-437,Kd值为2.8μM。10074-G5结合在由诱导螺旋结构域(Leu370-Arg378)的N末端的扭结(Asp379-Ile381)产生的空腔中[3]。

【摘要】目的建立人血清中左氧氟沙星(LOX)残留量的快速测定方法。
方法采用十二烷基硫酸钠(SDS)增敏荧光光度法。
结果用无水乙醇沉淀蛋白,对血清进行预处理,利用LOX-SDS体系测定,人血清中LOX在0.1~5.0μg.ml-1时,荧光强度与浓度呈线性关系,其回归方程为:F=5.552+40.019C(r=0.9990),平均回收率为
99.72%,RSD=0.68%。
结论所建方法简便、准确、快速,可用于人血清中LOX的测定。

说起WB凝胶配置,小编可有一肚子苦水要倒,第一个让人头疼的一点就是多种溶液都需要计算量取,每次调移液器就要调半天。
还有各种试剂的保存条件也不一样,配胶之前还需要从冰箱,室温柜子里分别拿出不同的溶液。
这还不算,最容易出问题的促凝剂APS,需要新鲜配置,4度保存不能超过一周,关键是粉末还特容易吸水,打开后需要密封保存。
小编之前就踩过一次坑,APS粉末吸水结块,称量配置后的APS含量不够,凝胶凝结不好,整个WB实验条带跑的七扭八歪。

如果没有酶标仪,也可以使用普通的分光光度计测定,但是测定蛋白浓度时,需根据测定吸光度的杯子的体积,按比例调整A液,B液和样品的体积。
使用分光光度计测定蛋白浓度时,每个试剂盒可以测定的样品数量可能会显著减少。
O 如发现样品稀释液或裂解液本身就有较高背景,请试用Bradford蛋白浓度测定试剂盒。
O 为了加快BCA法测定蛋白浓度的速度可以适当用微波炉加热,但是切勿过热。

【摘要】目的:建立测定痕量铬(Ⅵ)的双波长双指示剂催化动力学光度分析的新方法。
方法:在稀硫酸介质中,利用痕量铬(Ⅵ)催化溴酸钾氧化甲基橙和结晶紫褪色的指示反应,通过测量510nm和600nm下,催化体系和非催化体系吸光度的变化,用双波长双指示剂催化动力学光度测定痕量铬。
结果:测定铬的线性范围为0.0010—0.20μg/ml,检出限为1.1×10μg/ml。
结论:该法简单、灵敏度高、选择性好,可用于废水中痕量铬(Ⅵ)的测定。

【摘要】建立一种悬浮固化分散液液微萃取-液相色谱荧光法测定血清中阿齐沙坦含量的新方法。
在超声辅助下,乳化后的萃取剂1-十一醇高效快速萃取溶液中的阿齐沙坦,将萃取液离心后冰水浴中冷却,萃取剂凝固漂浮在水面而与溶液分离,分离后的1-十一醇用乙醇溶解后利用液相色谱荧光法测定其中阿齐沙坦的含量。
实验中对液相色谱测定条件、萃取条件进行了优化,包括激发与发射波长、流动相组成、萃取剂用量、萃取溶液pH、超声时间、离心时间、冷却时间等。
在优化条件下,工作曲线的线性范围为0.02~4.0μg/mL,最低检出限为6.0 ng/mL,回收率为
93.1%~100.4%。
专属性实验及血清样品测定结果表明该方法可以用于人血清中阿齐沙坦含量的测定。
