突发性聋免疫损伤证据:MMP-9表达下调及意义

【摘要】目的已有研究显示基质金属蛋白酶9(MMP-9)参与了系统性红斑狼疮的免疫损伤过程。本研究拟检测突发性聋患者血清MMP-9表达水平,探究突发性聋是否也存在类似损伤机制。方法采用ELISA方法检测自

2015年11月至2016年1月在重庆医科大学附属第一医院耳鼻咽喉科住院的43例突发性聋患者以及同期23例健康对照组志愿者血清中MMP-9的含量。结果对照组血清MMP-9平均浓度310.85±218.27ng/ml,患者组MMP-9平均浓度73.45±51.59ng/ml,其中低频型56.28±39.22ng/ml,高频型86.30±49.91ng/ml,平坦型61.01±47.77ng/ml,全聋型85.51±58.69ng/ml。突发性聋患者血清MMP-9表达水平均明显低于对照组(H=32.526,P<0.01),不同分型间血清MMP-9浓度差异无统计学意义(P>0.05)。结论MMP-9在突发性聋患者血清低表达,可能与其参与了内耳免疫损伤而大量消耗有关。推测MMP-9可能通过介导免疫损伤的方式参与了该病的发生发展。

Corresponding author:KANG Houyong Email:kanghouyong@ 【Abstract】Objective Matrix metalloproteinase9(MMP-9)has been shown to be associated with immune injury in systemic lupus erythematosus(SLE).This study aims to test the concentration of serum MMP-9in patients with sud-den sensorineural hearing loss(SSNHL)and to investigate whether there is a similar MMP-9associated injury mecha-nism in this disease.Methods Forty-three patients with SSNHL and23healthy volunteers were studied from November 2015to January2016.Serum MMP-9was tested by ELISA.Results Average serum MMP-9concentration was310.85±218.27ng/ml in the control group,compared with the much lower73.45±51.59ng/ml in the experiment group,(56.28±

39.22ng/ml in those with low frequency SSNHL,86.30±49.91ng/ml in those with high frequency SSNHL,61.01±

47.77ng/ml in those with flat type SSNHL,and85.51±58.69ng/ml in those with total deafness)(H=32.526,P<0.01).

Serum MMP-9levels were not significantly different among different SSNHL types(P>0.05).Conclusions Decrease of serum MMP-9in patients with SSNHL may be related to excessive consumption in the process of auto-immune dam-age in the inner ear.It is speculated that MMP-9may participate in the occurrence and development of the disease via immune injury.
