壳聚糖羟丁基衍生物的制备及性质

【摘要】以1,2-环氧丁烷在碱性条件下的开环产物为醚化剂,对壳聚糖(CS)分子进行改性,制备了水溶性羟丁基壳聚糖(HBCS),并采用红外光谱、固体13C核磁共振对产物的分子结构进行表征.结果表明:羟丁基成功连接到壳聚糖分子链上;元素分析法测定产物取代度为1.2;羟丁基壳聚糖可溶于水并可在较广的pH值范围内溶解;由于改性反应破坏了壳聚糖的结晶区结构,因此羟丁基壳聚糖的热稳定性比原料壳聚糖差.对壳聚糖和羟丁基壳聚糖的抑菌性研究结果表明:羟丁基壳聚糖能够抑制大肠杆菌和金黄色葡萄球菌的生长,并随着质量分数的增加抑菌效果明显增强;相同质量分数条件下羟丁基壳聚糖的抑菌活性比壳聚糖弱.%Water-soluble hydroxybutyl chitosan ( HBCS ) was prepared by the reaction of chitosan ( CS ) with 1,2-butane oxidation in an alkali solution. The structure of HBCS was characterized by FT-IR and solid state 13C NMR, and the results prove that the hydroxybutyl has been successfully connected to chitosan molecular chain. The degree of hydroxybutyl substitution estimated by means of elemental ananlysis is 1.2. Besides, HBCS exhibited good water-solubility and could dissolve in aqueous solutions in a broader pH range. In comparison with CS, HBCS shows a poorer thermostability. This is because the modified reaction destroys the crystalline region of chitosan. Antibacterial activities of CS and the modified derivative were explored by
turbidity method. The results show that HBCS exhibits antibacterial activity against E. Coli and S. Aureus, and with the increase of concentration, the antibacterial effect enhances obviously. It was found that the antimicrobial activity of HBCS is weaker than that of CS under the same mass concentrations.
