RealtimePCR检测L1—CAM在大肠癌中的表达及其临床意义

[摘要] 目的探讨L1细胞黏附分子(L1-CAM)在大肠癌中的表达情况及其与临床病理的相关性。
方法应用实时荧光定量PCR检测浙江省人民医院58例大肠癌组织及其癌旁正常组
织中L1-CAM的表达,分析L1-CAM在大肠癌中的表达及其与临床病理学特征和预后的关系。
结果癌组织和癌旁正常组织L1-CAM mRNA表达差异有统计学意义(P < 0.05)。
癌组
织L1-CAM mRNA表达与肿瘤分化程度、静脉侵犯、淋巴管侵犯、淋巴结转移、Dukes分期密切相关(均P < 0.05),而与性别、年龄、肿瘤大小、肿瘤类型无明显相关(均P > 0.05)。
结论L1-CAM在大肠癌组织中高表达,并且与大肠癌的浸润、转移有关。

[Abstract] Objective To investigate the L1-CAM mRNA expression in cancerous tissue of patients with colorectal carcinoma and to evaluate the relationship between L1-CAM mRNA expression and clinicopathological significance. Methods Cancerous tissue and para-carcinoma tissue samples were collected from 58 patients with colorectal carcinoma from Zhejiang Provincial People's Hospital. Real time PCR was used to detect the L1-CAM mRNA expression in cancerous tissue and para-carcinoma tissue, and the relationships between L1-CAM
mRNA expression and pathologic parameters were observed. Results The expression of L1-CAM mRNA was significantly higher in cancerous tissue than in para-carcinoma tissue (P < 0.05). L1-CAM mRNA expression was found to be significantly related with the pathologic parameters, such as depth of differentiation,venous invasion, lymphatic invasion, lymph node metastasis,and clinicopathological stage (Dukes),with statistically significant differences (P < 0.05);however,no significant association between L1-CAM mRNA expression and age, sex,tumor size, or the type of tumor were found (all P > 0.05). Conclusion The high expression of L1-CAM may be related with the devolopment and progression of colorectal cancer.
[Key words] Colorectal carcinoma; L1-CAM; Real time PCR L1细胞黏附分子(L1 cell adhesion molecules,L1-CAM)是分子量为200~220 kD的免疫球蛋白超家族Ⅰ型跨膜糖蛋白,是主要介导细胞间及细胞与胞外基质间黏附作用的细胞表面糖蛋白,其胞外域由6个细胞外免疫球蛋白区域、5个纤维连接蛋白区域以及由85~147个氨基酸组成的疏水跨膜细胞质区域组成[1]。
研究发现,L1-CAM对中枢神经系统的发育起着关键的作用:最初它是在神经元细胞特别是神经鞘细胞中被发现,促进L1-CAM阳性的神经细胞突。
