【摘要】Objective To observe the effects of Shenqi Bufei Decoction on the expression of HDAC 2 and histone acetylation H4 in airway smooth muscle cells (ASMCs) proliferation of COPD rat model.Methods The 4 ~6 cells of cultured primary rat ASMCs were divid -ed into blank,model,low,high dose Chinese medicine and aminophylline groups .ASMCs of the blank control group were normally cultured airway smooth muscle cells without treatment .ASMCs of the model group,low dose Chinese medicine
group ,high dose Chinese medicine group and aminophylline group respectively were the airway smooth muscle cells of the COPD rat .The model group was stimulated with ciga-rette smoke extract (CSE) and serum of the low,high dose Chinese medicine and aminophylline were added with low ,high dose Chinese med-icine and aminophylline respectively.The expression of proliferation cell nuclear antigen (PCNA) and histone deacetylase 2(HDAC2),ace-tyl histone H4 cell proliferation were tested by immunohistochemical staining .Results Compared with blank group,the airway smooth muscle cells proliferation and expression of cetyl histone H 4 protein were significantly increased (P<0.05),the expression of HDAC2 protein was significantly decreased of model group
(P<0.05).Compared with the model group,there was not significant difference between low -dose Chi-nese medicine group and model group (P>0.05),the airway smooth muscle cells proliferation and expression of acetyl histone H 4 protein were significantly decreased (P<0.05),the expression of HDAC2 protein was significantly increased of the high dose Chinese medicine group (P<0.05),the airway smooth muscle cells proliferation was significantly decreased (P<0.05),the expression of HDAC2 protein was signifi-cantly increased(P<0.05) and expression of acetyl histone H4 protein was unchanged of the aminophylline group
(P>0.05).Conclusions Shenqi Bufei decoction can restrain the proliferation
of ASMCs of COPD by increasing the expression of HDAC 2,inhibiting the expression of acetyl histone H4 in airway smooth muscle cell.%目的探讨参
芪补肺汤对慢阻肺(COPD)大鼠气道平滑肌细胞(ASMCs)增殖中组蛋白去乙
酰基酶2(HADC2)、乙酰化组蛋白 H4表达的影响。
方法将 SD 大鼠随机分为
正常组和 COPD 模型组。
原代培养大鼠 ASMCs 取4~6代细胞,分为空白组、
模型组、中药低剂量组、中药高剂量组和氨茶碱组。
其中空白组为正常培养ASMCs,不予处理。
模型组、中药低、高剂量组和氨茶碱组为 COPD 模型组培养ASMCs,模型组加入香烟提取物(CSE)及正常组大鼠血清;中药低剂量组加入CSE 及中药低剂量组大鼠血清;中药高剂量组加入CSE 及中药高剂量组大鼠血清;氨茶碱组加入CSE 及氨茶碱。
细胞免疫组织化学检测大鼠 ASMCs 增殖细胞核抗
原(PCNA)及各组 ASMCs 中 HDAC2、H4蛋白的表达。
结果与空白组比较,
模型组大鼠 ASMCs 明显增殖,ASMCs 中的 HDAC2表达明显降低,乙酰化组蛋白 H4表达明显增高(P<0.05)。
与模型组比较,中药低剂量组 ASMCs 增殖
和 HDAC2、乙酰化组蛋白 H4表达无统计学差异(P>0.05);中药高剂量组
ASMCs 平滑肌细胞增殖明显下降,ASMCs 中的 HDAC2表达明显增高,乙酰化组蛋白 H4的表达明显降低(P<0.05);氨茶碱组 ASMCs 增殖降低、ASMCs 中的 HDAC2表达明显增高(P<0.05),乙酰化组蛋白 H4表达无统计学差异(P>0.05)。
结论参芪补肺汤可抑制 ASMCs 增殖,其机制可能通过提高ASMCs 中 HDAC2的表达、抑制组蛋白 H4的表达,影响其平衡状态发挥作用。
