A change to elution buffer while maintaining upward flow will desorb the target protein in expanded bed mode. Alternatively flow is then reversed, the adsorbent particles quickly settle and target proteins are desorbed by an elution buffer as in conventional packed bed chromatography. The mode used for elution, expanded bed or settled bed, will depend on the characteristics of the feed.
