摘要:Disclosed herein are antibody-coupled T cell receptor (ACTR) polypeptides comprising: a CD16A extracellular domain, a transmembrane domain, one or more co-stimulatory signaling domains, at least one of which is a CD28 co-stimulatory signaling domain, and a CD3z cytoplasmic signaling domain. Also disclosed herein are genetically engineered immune cells, expressing: a first polypeptide which is an antibody-coupled T cell receptor (ACTR); and a second polypeptide that elicits a co-stimulatory signal as well as methods of enhancing antibody-dependent cell cytotoxicity (ADCC) in a subject comprising administering to a subject in need thereof a therapeutically effective amount of a therapeutic antibody and an effective amount of immune cells (e.g., T lymphocytes and/or NK cells) expressing an antibody-coupled T-cell receptor (ACTR) polypeptide.
