GENERAL DESCRIPTION
The OP07 has very low input offset voltage (75 μV maximum for OP07E) that is obtained by trimming at the wafer stage. These low offset voltages generally eliminate any need for external nulling. The OP07 also features low input bias current (±4 nA for the OP07E) and high open-loop gain (200 V/mV for the OP07E). The low offset and high open-loop gain make the OP07 particularly useful for high gain instrumentation applications.

Due to reducing the number of external component, the board space can be saved easily. The external shutdown function can be controlled by logic level and then come into standby mode. The internal compensation makes feedback control have good line and load regulation without external design. Regarding protected function, thermal shutdown is to prevent over temperature operating from damage, and current limit is against over current operating of the output switch. The AP1501 series operates at a switching frequency of 150Khz thus allowing smaller sized filter components than what would be needed with lower frequency switching regulators. Other features include a guaranteed +4% tolerance on output voltage under specified input voltage and output load conditions, and +15% on the oscillator frequency. The output version included fixed 3.3V, 5V, 12V, and an adjustable type. Thermal ConsiderationsThe AP1501 is available in two packages, a 5-pin TO-220 and a 5-pin surface mount TO-263.The TO-220 package needs a heat sink under most conditions. The size of the heatsink depends on the input voltage, the output voltage, the load current and the ambient temperature. The AP1501 junction temperature rises above ambient temperature for a 3A load and different input and output voltages. The data for these curves was taken with the AP1501 (TO-220 package) operating as a buck switching regulator in an ambient temperature of 25o C (still air). These temperature rise numbers are all approximate and there are many factors that can affect these temperatures. Higher ambient temperatures require more heat sinking.The TO-263 surface mount package tab is designed to be soldered to the copper on a printed circuit board. This curve can be used for a quick check for the approximate junction temperature for various conditions, but be aware that there are many factors that can affect the junction temperature. When load currents higher than 2A are used, double sided or multilayer PC boards with large copper areas and/or airflow might be needed, especially for high ambient temperatures and high output voltages.For the best thermal performance, wide copper traces and generous amounts of printed circuit board copper should be used in the board layout. (Once exception to this is the output (switch) pin, which should not have large areas of copper.) Large areas of copper provide the best transfer of heat (lower thermal resistance) to the surrounding air, and moving air lowers the thermal resistance even further.Package thermal resistance and junction temperature rise numbers are all approximate, and there are many factors that will affect these numbers. Some of these factors include board size, shape, thickness, position, location, and even board temperature. The effectivenessof the PC board to dissipate heat also depends on the size, quantity and spacing of other components on the board, as well as whether the surrounding air is still or moving. Furthermore, some of these components such as the catch diode will add heatto the PC board and the heat can vary as the input voltage changes. 方法:对利巴韦林软胶囊制备工艺以正交设计法进行确定,利巴韦林软胶囊质量用高效液相色谱法检测,进而确定利巴韦林软胶囊的制备工艺及质量。
结果:利巴韦林精密度良好,在人工胃液中能维持8h 药效稳定性,平均回收率计算为100%。
利巴韦林软胶囊含量测定率为99.5%、101.5%、99.6%。
利巴韦林软胶囊保质期为18个月。
结论:利巴韦林软胶囊制备工艺合理安全,操作简单方便,质量可靠。
【关键词】利巴韦林软胶囊;制备工艺;质量利巴韦林是临床常用抗病毒药物,对呼吸道合胞病毒、甲型肝炎病毒、流感病毒等多种病毒起到抑制作用[1]。
目前利巴韦林临床应用多为利巴韦林注射液注射,软胶囊是胶囊剂一种,具有高度生物利用度,密封性良好等优势,逐渐受到临床重点关注。
本文就分析利巴韦林软胶囊的制备工艺,探究其质量,其结果分析如下。
1 仪器与试剂1.1 仪器武汉朗玛恒瑞科技有限公司生产的LM-C2000超声波清洗器;VERTEX VI500高效液相色谱仪;Ascentis Express C18HPLC 色谱柱;上海天平仪器厂生产的TG328B 型电光分析天平;UV-2201型紫外扫描仪;上海光谱仪器有限公司生产的SP-756PC 紫外-可见分光光度计;北京联合科仪科技有限公司生产的FE20K 梅特勒FE20K 酸度计。
1.2 试剂广东汕头市西陇化工厂生产的硫酸铵;辛集市嘉泽化工有限公司生产的盐酸;天津市化学试剂公司分公司生产的明胶;石家庄市泰鑫化工有限公司生产的甘油。
利巴韦林(生产批号:20020324,中国药品生物制品检定所)。
甲醇、乙腈均属于色谱纯,纯化水。

2 利巴韦林软胶囊制备方法2.1 辅料选择利巴韦林溶解度较差,多为混悬液,为了稳定利巴韦林混悬液,在固定主药含量前提进行正交实验,通过L4(23)表确定处方。

IntroductionThe Thermo Scientific Pierce DNA Coating Solution is a quick, simple and cost-effective method for immobilizing DNA and oligonucleotides onto plastic surfaces. Because DNA has a low binding affinity for unmodified plastic surfaces, many researchers use binding proteins, such as avidin, to capture biotinylated DNA or chemical binding of DNA through thephospho groups to specially manufactured plates containing amino groups. These procedures are costly and time-consuming. The Pierce DNA Coating Solution, however, uses a simple mix-and-coat procedure that results in chemical bonding of DNA to most general laboratory plastic surfaces such as polystyrene microplates and polypropylene microcentrifuge tubes.Important Product Information•If the DNA Coating Solution is exposed to cold, a precipitate may form. To dissolve particulate, gently heat the solution in a water bath (25-37°C) with swirling. Do not use the solution if precipitate does not dissolve when heated. •Empirically establish if the DNA coated to surfaces using the DNA Coating Solution is suited for a specific downstream experiment. Hybridization procedures are likely to be more successful than complex enzymatic manipulations. •Although the DNA Coating Solution is suitable for oligonucleotides, a lower limit of oligo size has not been determined.Example Protocol1. In a glass test tube, combine up to 1 volume of DNA at 1-10µg/mL in water or TE buffer with 1 volume of Pierce DNACoating Solution. Mix for 10 minutes.2. Bradford法蛋白定量试剂盒(Catalog : DBI-1039,DBI-1040)原理这一方法基于考马斯亮蓝G-250有红、蓝两种不同颜色的形式。
在一定浓度的乙醇及酸性条件下,可配成淡红色的溶液,当与蛋白质结合后,产生蓝色化合物,反应迅速而稳定。
反应化合物在465~595nm处有最大的光吸收值,化合物颜色的深浅与蛋白浓度的高低成正比关系,因此可检测595nm的光吸收值的大小计算蛋白的含量。
描述1 所需时间:10min。
2 优点:快速(反应时间仅需2min);敏感,几乎没有蛋白质损失。
3 缺点:对各种纯化蛋白质反应不同;用这种测定方法对蛋白质引起不可逆的变性。
4 灵敏度范围:蛋白质溶液25μg/ml~200μg/ml,最小测量体积0.1ml,最小测量蛋白质2.5μg。

制作标准曲线1)做标准曲线所用的已知蛋白质应和未知浓度蛋白质溶液一起准备,这对于定量测定蛋白质浓度是十分必要的。
另外,每一样品应当做两到三次重复,如果用同一个比色杯来测定蛋白质的浓度,则应当先测量低蛋白浓度的样品,以避免因Bradford染料吸附在比色杯上不能彻底冲洗干净而带来误差。
