In cell adhesion studies assessing the human melanoma M21 or UCLA–P3 human lung carcinoma cell lines, Cilengitide inhibits integrin–mediated binding to vitronectin with IC 50s of 0.4 and 0.4 μM [1]. In vitro treatment of Cilengitide, at a concentration greater than 1 μM, shows concentration– andtime–dependent cytotoxic effects. However, lower doses of Cilengitide monotherapy (0.1 and 0.5 μM) does not elicit the effective death of the both U87MG and U251MG cells. Significant cytotoxic effects are observed in the U87MG cells with the addition of 1μM Cilengitide in combination with Belotecan monotherapy at concentration of 6.25 nM. BIOLOGICAL ACTIVITY:
Maslinic acid(Crategolic acid) is a pentacyclic triterpene found in a variety of natural sources; exerts a wide range of biological activities, i.e. antitumor, antidiabetic, antioxidant, cardioprotective, neuroprotective, antiparasitic and growth–stimulating.

in vitro: MA was able to suppress the viability of cardiac carcinoma cells in both a time– and dose–dependent manner. MA induced the activation of p38 MAPK in cardiac carcinoma cells and, in turn, changed their mitochondrial membrane potential (MMP) [1]. MA exerts anti–diabetic effects by increasing glycogen content and inhibiting glycogen phosphorylase activity in HepG2 cells.
Furthermore, MA was shown to induce the phosphorylation level of IRβ–subunit, Akt, and GSK3β. The MA–induced activation of Akt appeared to be specific, since it could be blocked by wortmannin [2]. the combination of MA and GEM inhibited constitutive NF–κB activation and NF–κB–regulated gene products, including cyclin D1, Bcl–2, Bax, MMP–2 and MMP–9, to a greater extent [4].
in vivo: Maslinic acid exhibits the growth–suppressing activity of MA on T24 and 253J xenograft tumor in mouse models [3]. he group that was treated with MA + GEM showed significant reductions in tumor volume and a decreased expression of NF–κB–regulated gene products [4].
