终止子:转录终止子按照是否依赖和不依赖ρ因子的作用分为两类,这两类终止子均在终止点前含有一段7-20bp的回文序列。终止子可以保护mRNA在核外不被降解,显著延长mRNA的寿命,由此提高重组蛋白的表达量。但是对于T7系统来说,由于T7 RNA聚合酶效率极高,宿主中随时都有充足的mRNA以供翻译,因此大部分在T7系统中表达的重组蛋白并不在意质粒上是否有终止子,只有一些自身带有翻译起始信号的外源基因需要终止子。启动子受细胞类型的限制,在不同的细胞系中有很大不同,因此需根据宿主细胞(尤其是真核宿主)的类型选择不同的启动子以便于目的基因的高效表达。

1.3 优化表达条件
重组蛋白的表达流程很少有一次成形的,为了提高蛋白表达量、改善蛋白质量,表达条件和白不表达时:

如果重组蛋白不表达(包含体和可溶蛋白都没有),首先检查cDNA和质粒是否正确,蛋白对宿主菌是否有很大毒性,然后尝试更换菌株、质粒载体和融合标签。原核蛋白在大肠杆菌中不能表达的情况很少见,通常是真核蛋白不能表达。不能表达的重组蛋白,即使在更换了宿主、载体后可以表达,表达量也不会很高,如果需要大规模生产,最好尝试酵母和昆虫细胞表达系统。
融合标签:融合标签是与目的蛋白共表达的一段多肽,方便重组蛋白的纯化、固定和检测,表3给出了常用的重组标签。如果不需要对重组蛋白进行纯化,尽量不要引入融合标签,以免影响蛋白性质;如果重组蛋白本身能够结合某种亲和柱,如某些金属结合蛋白可以结合Ni-NTA,某些糖结合蛋白能够特异识别糖类,也不必引入标签。融合标签的引入能够大大简化重组蛋白的纯化流程,并提高蛋白溶解度。商业化表达质粒,如pET、pGEX等提供了各种纯化标签和融合蛋白供选,应根据蛋白具体情况进行选择。His-tag是最常用的纯化标签,它具有很多优点:标签较短(10-20个氨基酸残基),不带电(pH8.0),免疫原性差,通常不影响重组蛋白的结构和功能,Ni2+亲和力高,能够通过一步纯化达到60%-90%的纯度。如果蛋白质溶解度不高,导致折叠困难、表达量低,可以选择较大的融合标签(GST、MBP、Trx等)帮助重组蛋白表达和折叠,提高重组蛋白溶解度,从而提高表达量。较大的融合标签有时也会导致翻译困难甚至提前中止,纯化后发现大部分都是标签蛋白也是常见现象。翻译的提前中止会大大影响重组蛋白产率和后续纯化,所以在短标签能够达到目的的时候,尽量不要选择大的融合标签。标签位置的选择也很重要:N端标签(短的或长的)自身带有启动子和适应宿主偏好的密码子,可以帮助目的蛋白表达,提高表达量,但是提前中止翻译的蛋白片段也会被一并纯化出来,降低重组蛋白纯度,对蛋白酶敏感的、自身容易降解的以及一级序列中有集中的疏水残基区的蛋白尤其要避免使用N端标签;C端标签则可以保证只有完整蛋白得到纯化。另外,如果蛋白的近N端或近C端有重要功能区,如酶活中心、配体结合位点、二硫键、多聚体稳定界面、相互作用界面等,则要避免纯化标签位于该末端,以免影响重组蛋白的结构和功能。如果融合标签对蛋白性质有较大影响,但又是纯化所必须的,就可以考虑在纯化过程中去除标签。主要有三种方法:化学裂解,如溴化氰(CNBr)、羟胺(NH2OH)等,能够简单有效地去除标签,但反应条件苛刻(羟胺需要在pH9.0下反应),特异性较差,而且会引入不必要的修饰,除包含体蛋白的处理外已经很少使用了;酶解,如PPase等,其底物一般是一段比较长的肽链,特异性强,是目前比较常用的方法,缺点是酶切反应需要较长的时间,也增加了蛋白纯化的步骤,使纯化变得繁琐;IMPACT质粒,该质粒在纯化标签和目的蛋白之间插入了一个蛋白质内含子(intein),intein具有可诱导的自切割活性,使用IMPACT质粒表达的重组蛋白,只需要改变缓冲液的pH和温度,即可切掉融合标签。

(1)稳定的遗传复制和传代能力;

重组蛋白质的分离纯化 (1)

重组蛋白质的分离纯化摘要:90年代以来基因重组技术得到很大的发展,基因工程产品的分离纯化的成本约占其全部成本的60%~80%,因此重组蛋白的分离纯化技术越来越重要。

本文主要介绍了沉淀、液液萃取、层析等常用分离重组蛋白方法的原理及应用,旨在为开展蛋白质的制备及其应用研究提供理论依据。

关键词:重组蛋白质;分离;纯化;沉淀;液液萃取;层析;包涵体随着基因重组技术的发展,出现了很多基因工程产品,而作为基因工程技术的下游工程中的基因重组蛋白的分离纯化技术越来越显示其重要性。

据有人统计,基因工程产品的分离纯化成本约占到其全部成本的60%~80%[1]。

由此可见产品的分离纯化是获得目的产物的关键一步,也是比较困难的一步,它标志着生物产业的高低。

纯化重组蛋白质和普通蛋白质的不同就在于要选择合适的表达系统,因为表达系统决定了细胞培养过程中产物的性质以及可能产生的杂蛋白,而纯化重组蛋白质的主要目的是去除杂蛋白质,通常对一种重组蛋白质的纯化会采用多个系统[2]。

但是重组蛋白有几种不同的表达形式,如细胞外的分泌表达;细胞内可溶性表达以及包涵体形式的存在,因此对于重组蛋白的纯化要依据其表达形式的不同,采取不同的纯化工艺。

与传统方式相似,重组蛋白的分离纯化也是利用其物理和化学性质的差异,即以分子的大小、形状、溶解度、等电点、亲疏水性以及与其它分子的亲和性等性质建立起来的。

目前主要的纯化方法有浓缩沉淀法,层析和电泳技术。

重组蛋白质在分离纯化的过程中,必须维持一定的浓度和生物活性形式,以及防止被降解。

因此从生物体中有效分离纯化重组蛋白质一直是个难题。

90 年代以来,国内外许多科学工作者在蛋白质分离纯化技术和工艺上进行了大量的研制和开发,将原有的纯化技术水平提高到一个新的高度。

本文将简单介绍一些传统的分离纯化方法,并介绍近10 年来重组蛋白分离纯化中的新进展和一些新出现的技术。

1 沉淀分离技术1.1 盐析法其原理是蛋白质在高浓度盐溶液中,随着盐浓度的逐渐增加,由于蛋白质水化膜被破坏、溶解度下降而从溶液中沉淀出来。

三、凝胶滞留试验(EMSA)凝胶迁移滞留试验(electrophoretic mobility shift assays,EMSA)是研究核酸与蛋白质相互作用简单、快速、敏感的方法,目前已经成为转录因子研究的经典方法,并且用于研究RNA结合蛋白和特定的RNA序列的相互作用。

其基本原理是蛋白质可以与末端标记的核酸探针结合,电泳时这种复合物比无蛋白结合的自由探针在凝胶中泳动的速度慢,即表现为相对滞后。

(一)转录因子-标签融合蛋白和标签蛋白的原核表达纯化以GST标签为例。

裂解液:25mM pH7.8Tris-HCl、100mM NaCl、2 mM NaEDTA、1mM DTT和21×Complete Protease InhibitorCocktail。

洗脱液:50 mMpH7.8Tris-HCl、200mM NaCl、1mM DTT、0.02%(v/v)Triton X-100和10 mM还原性谷胱甘肽。

EDTA、1mM DTT和1×Complete 透析液:25mM pH8.0 Tris-HCl、1mM Na2Protease InhibitorCocktail。

1将转化了pGEX-4T-2载体或者pGEX-4T-2-目的基因重组载体的BL21大肠杆菌在5 mL含100 µg mL-1氨苄青霉素的LB培养液中37℃、200 rpm震荡培养12 h。

2 取1 mL菌液加入100 mL 含100 µg mL-1氨苄青霉素的LB培养液中,37℃、200 rpm震荡培养至OD600为0.5左右。

3 加入IPTG至终浓度为0.5 mM,28℃、200 rpm培养6 h。

4 冰上放置10 min,4℃、3000×g离心15 min,收集菌体并称重。

5 每克菌体加入3 mL裂解液,重悬菌体,加入溶菌酶至终浓度为0.5 mgmL-1,在冰上温和地搅动菌液30 min。

蛋白体外表达与纯化

蛋白体外表达与纯化随着后基因组时代的到来,蛋白质组成为科学研究的热点。

蛋白质作为生命机体的主要活动的承担者,其体外表达与纯化在研究相应基因的功能上有重要意义。

蛋白体外表达系统按其表达宿主可分为原核表达系统,真核表达系统和哺乳动物细胞表达系统。

一:原核表达系统原核表达系统的宿主菌主要以大肠杆菌为代表,大肠杆菌表达体系是目前应用最广泛的外源基因表达体系,这也是外源基因表达的首选体系。

该表达体系的优点:遗传学和生理学背景清楚;容易培养;外源基因经常可以高效表达及操作简单、周期短、成本低等。

其不足之处是不能进行典型真核细胞所具有的复杂的翻译后修饰;广泛的二硫键的形成及外源蛋白组装成蛋白复合体的能力也受到限制;另外外源基因产物在大肠杆菌中易形成不溶的包涵体;有时由于真核mRNA的结构特性及密码子使用频率与大肠杆菌的差异,而的不到足够的产物。

二:真核表达系统真核表达系统的宿主菌主要以酵母表达系统为代表,酵母基因表达系统的载体通常既能在酵母中进行复制也能在大肠杆菌中进行复制,形成所谓酵母菌――大肠杆菌穿梭载体。

因以大肠制备质粒DNA较方便,通常利用大肠杆菌系统构建酵母载体以简化手续,缩短时间。

作为基因表达系统的宿主应该具备以下条件:安全无毒,不致病;遗传背景较清楚,容易进行遗传操作;容易进行载体DNA的导入;培养条件简单;有良好的蛋白分泌能力;有类似高等真核生物的蛋白翻译后修饰功能。

三:哺乳动物细胞表达系统由于本专业不涉及哺乳动物细胞表达系统的应用,故此不赘述。

表达载体的种类及相应的分离纯化方法作为表达载体必须具备以下特征:稳定的遗传复制、传代能力,无选择压力下能存在于宿主细胞内;具有显性的筛选标记;启动子的转录是可调控的;启动子的转录的mRNA能够在适当的位置终止;具有外源基因插入的多克隆位点。

在原核表达系统中常用的表达载体有:PET-载体系列,用这类载体表达出的外源蛋白在N 端或C端或两端均具有his tag。

大肠杆菌基因表达系统

必须用cDNA或者是化学合成的基因。 外源基因不能带有内含子。
