The is a very important property when mixing the material with ceramic or polymer matrixes when trying to improve their electrical and mechanical properties. However, on the other hand, the electrical conductivity of GO is reduced and sometimes described as an electrical insulator, due to the bonding of the oxygen molecules (Source: ).There are many ways in which GO can be functionalized, depending on the desired application. For optoelectronics, biodevices or as a drug-delivery material, for example, it is possible to substitute amines for the organic covalent functionalization of graphene to increase the dispersability of chemically modified graphenes in organic solvents. It has also been proved that porphyrin-functionalized primary amines and fullerene-functionalized secondary amines could be attached to graphene oxide platelets, ultimately increasing nonlinear optical performance (Source: ).In order for GO to be usable as an intermediary in the creation of monolayer or few-layer graphene sheets, it is important to develop an oxidization and reduction process that is able to separate individual carbon layers and then isolate them without modifying their structure.GRAPHENE OXIDE APPLICATIONSThere are many applications which use GO and the list continues to grow. There are many obstacles needed to be overcome before GO can be used commercially in most areas but advancements are being made every day and some of the key areas of development for GO include bioengineering, composite materials, energy technology and nanotechnology.Energy StorageNanocomposites of reduced GO have been used for high capacity energy storage in lithium ion batteries. In these studies, electrically insulating metal oxide nanoparticles were adsorbed onto reduced GO to increase the performance of these materials in batteries. High surface area of reduced GO has been made using microwaves for exfoliation and reduction of GO. The high surface area reduced GO formed is useful as an energy storage material in super capacitors (Source: ).Graphene based micro-supercapacitors will likely be developed for use in low energy applications such as smart phones and portable computing devices. Graphene-enhanced lithium ion batteries could be used in much higher energy usage applications such as electrically powered vehicles, or they can be used as lithium ion batteries are now, in smartphones, laptops and tablet PCs but at significantly lower levels of size and weight (Source: ).Photovoltaic CellsOffering very low levels of light absorption (at around 2.7% of white light) whilst also offering high electron mobility means that graphene can be used as an alternative to silicon or ITO in the manufacture of photovoltaic cells. Silicon is currently widely used in the production of photovoltaic cells, but while silicon cells are very expensive to produce, graphene based cells are potentially much less so. When materials such as silicon turn light into electricity it produces a photon for every electron produced, meaning that a lot of potential energy is lost as heat. Other Electronic devices using GO as a starting material for at least one of the key components, including the field effect transistor (FET) that uses reduced GO as a chemical sensor and biosensor to detect hormonal catecholamine molecules, avidin and DNA. Whilst, another study had GO functionalised with glucose oxidase and deposited on an electrode was used as an electrochemical glucose sensor. (Source: ).UltrafiltrationAnother standout property of graphene is that while it allows water to pass through it, it is almost completely impervious to liquids and gases (even relatively small helium molecules). This means that graphene could be used as an ultrafiltration medium to act as a barrier between two substances. The benefit of using graphene is that it is only 1 single atom thick and can also be developed as a barrier that electronically measures strain and pressures between the 2 substances.This could mean that graphene is developed to be used in water filtration systems, desalination systems and efficient and economically more viable biofuel creation (Source: ).Composite MaterialsGraphene is strong, stiff and very light. Currently, aerospace engineers are incorporating carbon fibre into the production of aircraft as it is also very strong and light. However, graphene is much stronger whilst being also much lighter. Ultimately it is expected that graphene is utilized to create a material that can replace steel in the structure of aircraft, improving fuel efficiency, range and reducing weight. 23个碱基对)的dsRNA分子㊂它可以由dsRNA经Dicer酶裂解后在胞内生成,也可以通过人工合成㊂进入胞浆后的siRNA与Ago蛋白形成RNA诱导的沉默复合物(RNA⁃inducedsilencingcomplex,RISC)㊂在RISC中siRNA裂解,随后由反义链对靶mRNA进行碱基配对酶切,抑制了目标RNA翻译为蛋白质,达到基因沉默的目的[5-6]㊂FDA批准Alnylam生产的ONPATTRO投入临床治疗[7],标志着RNA成为继化学和蛋白质疗法之后在制药学的第三个里程碑㊂目前siRNA非病毒载体递送用于肿瘤治疗已成为纳米生物医药的研究热点㊂siRNA可以通过诱导mRNA的降解,以序列特异性的方式抑制致癌基因的表达,在细胞信号转导中发挥重要作用,因此基于siRNA的抗癌药物具有广泛的应用前景㊂但是目前siRNA递送的主要障碍有:全身给药后siRNA在体内的非特异性分布造成的低转染效率,引起免疫反应及毒性;siRNA易受核酸酶的降解以及网状内皮系统的清除;血管内皮壁㊁多重组织的物理屏障阻碍了siRNA药物导入肿瘤细胞;siRNA被细胞摄取和内吞效应低;siRNA无法实现高效的内体逃逸以及其对mRNA的脱靶效应㊂为了克服这些问题,本文将围绕siRNA非病毒载体进行综述,主要包括载体的体内递送过程㊁不同载体用于siRNA递送中的优势和不足,以及它们中一些已经进入临床试验的siRNA载体及制剂,为新载体的设计提供借鉴,推动RNAi疗法向临床的转化㊂1㊀siRNA药物用于治疗的主要障碍及作用靶点㊀㊀通过化学修饰和特定序列设计的RNAi疗法几乎能够降解任何基因mRNA转录物,具有高度特异性㊂同时它依赖ATP的供能,通过天然的调节途径,以催化作用的方式将抑制特定基因表达的效率最大化㊂选择合适的靶点和给药途径非常重要㊂siRNA在消化道内的不稳定性以及其对于肠道上皮细胞的低渗透性都严重阻碍了口服给药的进行㊂同时皮下注射也受到亲脂性和载体大小的限制㊂相比之下,静脉注射是首选㊂静脉注射后裸露的siRNA分子会被血清核酸内切酶降解,最终被肾脏清除[8]㊂同时网状内皮系统(如肝㊁脾)巨噬细胞的非特异性摄取也会对它们造成吞噬破坏㊂在细胞外屏障方面,细胞外基质的复杂性㊁siRNA与细胞膜之间的电荷排斥以及选择性区域(如血脑屏障)的紧密连接都阻碍siRNA对组织的渗透[9-10]㊂在细胞内屏障中,通过内吞作用进入胞内的siRNA分子如果无法实现早期的内体逃逸,则会被溶酶体酸化降解以及胞吐排出[11]㊂逃逸到细胞质中的外源性siRNA还可能会造成靶基因以外的基因表达减少,具有潜在治疗风险㊂此外,siRNA序列中的特殊结构可能会通过激活Toll样受体,产生干扰素(α或β)和炎性细胞因子,触发不必要的免疫反应㊂针对药物和抗体难以抑制的转录因子和某些关键的肿瘤蛋白(如Ras)[12],siRNA可以通过碱基配对识别其靶标㊂目前已有多种靶点被开发出来用于抗肿瘤药物的筛选,包括程序性死亡因子配体-1(programmeddeathligand1,PD-L1)㊁细胞毒性T淋巴细胞相关蛋白4(cytotoxicT-lymphocyte⁃associatedprotein4,CTLA4)㊁表皮生长因子受体(epidermalgrowthfactorreceptor,EGFR)㊁血管内皮生长因子等㊂根据靶标作用机制可以将其分为靶向肿瘤形成调控机制㊁肿瘤微环境㊁肿瘤免疫调节㊁肿瘤生物标志物及肿瘤干细胞㊂EGFR常在非小细胞肺癌中表达异常,其中19号外显因子缺失突变最为常见㊂它激活了EGFR的酪氨酸激酶活性,从而诱导下游的促生长信号通路㊂Nascimento等[13]将构建的针对致癌EGFR突变体的等位基因特异性siRNA递送到肺肿瘤模型中,成功诱导了其凋亡㊂针对PD-L1和CTLA4的免疫疗法已经在治疗黑色素瘤㊁霍奇金淋巴瘤㊁非小细胞肺癌和膀胱癌中获得了进展㊂2㊀siRNA递送载体的主要类型开发合适的载体显得非常必要㊂目前研究主要集中在开发保护siRNA不受核酸酶影响的载体㊂载体通常以静电或共价方式[14]与siRNA结合形成纳米颗粒㊂㊃245㊃北京生物医学工程㊀㊀㊀㊀㊀㊀㊀㊀㊀㊀㊀㊀㊀㊀㊀㊀㊀㊀㊀第42卷2 1㊀脂质体及脂质类似物脂质体制剂具有高度的生物相容性,作为最成熟的RNA载体已成功应用于COVID-19mRNA疫苗的递送,推动了基因治疗的边界㊂脂质体中阳离子脂类最常见,它通过静电吸附的方式显著提高了体外转染效率㊂目前大多数脂质复合物在阳离子脂类的基础上添加了如二油酰磷脂酰胆碱(dioleoylphosphatidylcholine,DOPC)㊁二油酰磷脂酰乙醇胺(dioleoylphosphatidylethanolamine,DOPE)㊁胆固醇[15]和其他一些天然脂类㊂这些中性辅助脂质提高了载体的稳定性和细胞摄取率㊂Hattori等[16]制备了17种由阳离子脂质和DOPE组成的脂质复合物,结果表明不同的阳离子脂质类型对静脉注射后siRNA的生物分布和抑制效率影响显著㊂相比之下,环境响应型脂质是一类新型的脂质传递系统㊂这些脂质具有简单的化学结构,并对肿瘤微环境的变化作出响应(如可电离且pH敏感的可质子化氨基脂)㊂与阳离子脂质不同,可质子化氨基脂中的氨基头基在酸性pH下会电离而带强正电荷,电荷密度控制着颗粒之间的相互作用并抗衡离子的吸附,从而控制着纳米颗粒的稳定性㊂多项研究表明肿瘤微环境响应型脂质疗效高且副作用低,但其在克服免疫系统的清除实现最大递送效率方面还需进一步优化㊂临床上脂质体制剂Atu027和ALN-VSP02已经分别完成了Ⅰ/Ⅱ期和Ⅰ期试验㊂其中Atu027是一种针对蛋白激酶N3的特异性siRNA制剂,它联合吉西他滨在治疗晚期胰腺癌受试者上显示出良好的安全性和耐受性㊂同时Tekmira公司研发出一款靶向于Polo样激酶1的siRNA药物TKM-080301㊂针对该药物开展的Ⅰ/Ⅱ期临床研究在晚期实体瘤患者中进行,结果证明了它的抗肿瘤活性和良好耐受性㊂2 2㊀聚合物聚合物容易大规模合成与生产,且其只依赖于非共价作用,如静电效应或氢键压缩siRNA㊂目前多种阳离子聚合物(cationicpolymer,CP)已被广泛用于核酸传递,它们具有共同的理化特性[17],如阳离子电荷㊁两亲性和融合性㊂具有代表性的CP有聚乙烯亚胺(polyethylenimine,PEI)㊁聚β-氨基酸酯(Polyβ-aminesters,PBAE)㊁聚赖氨酸(poly⁃L⁃lysine,PLL)㊁聚丙交酯乙交酯(polylactide⁃co⁃glycolide,PLGA)以及聚酰胺胺(polyamidoamine,PAMAM)等㊂CP方便进行化学修饰,与阳离子脂质体相比它不含疏水部分而直接溶于水㊂目前CP的转染效率和细胞毒性之间的平衡问题是其用于临床的主要挑战㊂聚乙烯亚胺(Polyethyleneimine,PEI)可将外源基因转染到悬浮细胞或贴壁细胞中㊂它的高缓冲能力可以通过 质子海绵效应 促进内体释放[18]㊂然而PEI通常不易降解,聚合链上的高密度电荷与蛋白质之间的强烈作用也会降低多聚体的血清稳定性㊂Xue等[19]通过三氟乙酸乙酯和全氟丁酰氯的酰胺化反应合成的两个系列的氟化PEI,发现经氟化处理后的PEI细胞毒性显著降低㊂氟化已被证明是降低PEI的细胞毒性和提高siRNA递送效率的有效方法㊂PLL和PBAE与PEI相比生物相容性更好,但是化学键的不稳定性增加了它们进行高效递送的难度[20]㊂PLGA具有粒径小㊁相对无毒和持续释放轮廓的优点㊂表面具有大量活性基团的PAMAM树枝状大分子可以通过修饰不同的官能团(配体)来实现其药物靶向[21]㊂然而高度分支化的PAMAM提供了高效基因转染的同时也具有高度毒性㊂不同于线型结构的CP,分支化的CP通过其多功能末端基团组成的三维结构能更紧密地包裹siRNA,在生理pH下形成多聚复合物㊂与CP相比,天然类聚合物生物相容性更好㊂胶原蛋白是天然类聚合物的代表㊂目前试验中常通过靶向修饰siRNA/胶原蛋白复合物来抑制肿瘤生长㊂基于环糊精聚合物的纳米颗粒CALAA-01是第一个进入癌症临床试验的靶向siRNA传递系统[22],但是Ⅰ期临床试验由于患者出现剂量限制的毒副反应而终止㊂壳聚糖(chitosan,CS)是一种线性多糖,pKa值为6 2 7 0的D-氨基葡萄糖残基弱化了壳聚糖的碱性㊂当pH低于pKa时,伯胺会质子化㊂CS与siRNA结合构成的磷酸骨架转染效率较低㊂Choi等[23]通过加入阳离子谷氨酰胺偶联壳寡糖构建了纳米载体系统,显著提高了转染效率㊂在治疗胰腺癌方面,以聚合物胶束为制剂的NC-6004已经进入到Ⅲ期临床试验㊂而NC-4016也投入到治疗各种实体瘤初步的临床药理学㊁人体安全性评价试验中㊂同时,SilenSeed公司研发出的靶向siRNA药物siG12D-LODER也进入Ⅱ期临床试验㊂它是一种可生物降解的聚合物基质,其Ⅰ期数据显示出与化疗药物(如吉西他滨㊁厄洛替尼以及奥沙利铂)联用对胰腺导管腺癌具有一定疗效㊂㊃345㊃第5期㊀㊀㊀㊀㊀㊀王飞,等:siRNA非病毒载体递送用于肿瘤治疗的研究进展2 3㊀无机纳米颗粒无机纳米粒子(inorganicnanoparticles,INPs)尺寸可控㊁表面易于化学修饰,显示出对酶降解优异稳定性的同时体内示踪效果也较好㊂目前主要需要克服其在体内长期滞留带来的潜在不良反应㊂常用的INPs如下:金纳米颗粒(goldnanoparticles,GNPs)的尺寸很小,能够直接通过核孔复合体进行核靶向㊂此外,GNPs通过改变不同的生物活性配体如肽㊁聚合物和抗体来实现其功能多样性㊂据报道,环状精甘天冬氨酸(arginylglycylasparticacid,RGD)肽修饰的GNPs抗癌效果良好㊂相对于Cu和Ag,S与Au配体的共价性较强㊂利用GNPs传递siRNA的最直接方法是通过Au-S共价将siRNA偶联到其表面㊂例如,Ahwazi等[24]将人类免疫缺陷病毒的反式激活蛋白通过Au-S键偶联到GNPs上来治疗乳腺癌㊂磁性氧化铁纳米颗粒通过磁响应性在控制粒子目标成像的同时还能产生高热来消融组织㊂介孔二氧化硅纳米颗粒(mesoporoussilicananoparticles,MSNs)比表面积大,结合位点多,易于官能化㊂Pinese等[25]将PEI接枝到粒子的表面,设计了一种纤维表面吸附的新型siRNA/MSN-PEI涂层支架,基因沉默效果良好㊂目前已证明用阳离子聚合物包覆的MSNs可以有效地负载siRNA㊂最近,一种用于正电子发射断层扫描光学成像的124I环状RGD多肽无机杂化纳米粒子由于其尺寸极小㊁靶向性好而受到了关注㊂它在Ⅰ期临床试验中对转移性黑色素瘤的整合表达病灶产生了明显的对比㊂2 4㊀细胞穿透肽细胞穿透肽(cell⁃penetratingpeptides,CPP)因病毒跨膜蛋白结构域的研究而被发现[26]㊂在增强CPP靶向特异性方面主要有两种方法:设计以肿瘤细胞为首选和针对癌细胞内特性的工程肽㊂药物可以与肿瘤归巢蛋白㊁膜受体特异性抗体相连接㊂此外,基于CPP的药物可以被设计成只在肿瘤特殊的生理微环境中激活㊂如Ben等[27]通过研究由细胞穿透肽gH625功能化的siRNA纳米载体(称为CS⁃MSN)在三阴性乳腺癌模型中的细胞转运,发现CS⁃MSN转染效率比相同的不含gH625肽的纳米载体高1 7倍㊂同人工合成载体的内吞作用不同,CPP最大的特征在于其可以不依赖能量直接穿膜㊂CPP与siRNA共价或非共价结合成纳米颗粒,但其在体内递送的稳定性还有待提高㊂2 5㊀外泌体外泌体具有长循环的半衰期和可变形的细胞骨架,它在细胞间充当着通信媒介的作用㊂同时部分细胞分泌的外泌体对肿瘤还具有归巢能力[28]㊂Xu等[29]以P21激活激酶(4P21-activatedkinase4,PAK4)为靶点,通过瘤内注射将来自胰腺癌的外泌体包裹PAK4特异性siRNA(PAK4-specificsiRNA,siPAK4)输送到肿瘤细胞中,H&E染色显示siPAK4处理组有明显的组织细胞凋亡,小鼠的存活率显著提高(P<0 Review
abstract
Thrombus formed in blood vessel is a progressive process, which would lead to lifethreatening thrombotic diseases such as ischemic stroke. Unlike other diseases, the recognition of thrombus is usually in the late stage where blood vessels are largely blocked. So acute thrombotic diseases have a narrow therapeutic window, and remain leading causes of morbidity and mortality, whereas current thrombolysis therapy has limited therapeutic effects and bleeding complications. Thrombolytic agents in unwanted sites would cause hemorrhage due to the activation of plasminogen. Moreover, untargeted thrombolysis therapy require large amounts of thrombolytic agents, which in return would enhance hemorrhage risk. To improve the efficiency while minimizing the adverse effects of traditional thrombolysis therapy, novel drug delivery systems have been investigated. Various targeting strategies including ultrasound and magnetic field directed targeting, and specific binding, have been designed to deliver thrombolytic drugs to the thrombotic sites. These strategies demonstrate promising results in reducing bleeding risk as well as allowing less dosage of thrombolytic drugs with lowered clot lysis time. In this review, we discuss recent progress on targeted delivery of thrombolytics, and summarize treatment advantages and shortcomings, potentially helping to further promote the development of targeted thrombolysis.
