IP胶条测序
蛋白混合物进行免疫沉淀反应(IP)后,可以得到与目标蛋白相互作用的靶蛋白,此时一般需要对靶蛋白进行进一步的鉴定,如分子量、含量、翻译后修饰以及一级结构表征等,以全面了解靶蛋白的相关信息。
凝胶电泳是检测分子量最简单的方法,将IP免疫沉淀获得的靶蛋白进行凝胶电泳,电泳结束后蛋白胶条所处的位置对应的Maker的大小就是该靶蛋白的分子质量,此时如还要进行靶蛋白一级结构分析,可以将该蛋白胶条进行回收,然后利用蛋白测序技术对回收的靶蛋白进行序列分析,即IP胶条测序。
常用的测序技术如Edman
降解法N末端测序、C末端测序以及基于质谱的测序技术,质谱技术可以实现蛋白
质的全序列分析、从头测序以及末端测序,对于鉴定与目的蛋白相互作用的未知靶蛋白很有意义。

SDS-PAGE(Sodium Dodecyl Sulfate Polyacrylamide Gel Electrophoresis)十
二烷基硫酸钠聚丙烯酰氨凝胶电泳是一种常用的蛋白质分离纯化技术,它利用十二烷基硫酸钠消除蛋白质结构和电泳的影响,使蛋白质仅根据分子质量差异进行分离,广泛用于蛋白质的分离、纯度鉴定、分子量鉴定以及表达分析等。
蛋白质SDS-PAGE检测包括蛋白质SDS-PAGE电泳和电泳后凝胶图像分析两个部分。
在蛋白质SDS-PAGE电泳过程中,不同分子质量的蛋白质迁移速率不同,相对分子
质量较大的蛋白质在电场的作用下迁移较慢,电泳结束后仍然靠近点样孔;而低分子质量的蛋白质迁移速率较快,电泳结束后处于离点样孔较远的位置。
经银染或考马斯亮蓝等方法染色后,便可以观察到凝胶上蛋白质条带的大小、数量和分布情况。

结冷胶最早由美国Kelco公司从水百合中分离所得,1988年在日本被批准用于食品,1992年被美国FDA批准为食品添加剂,我国于1996年批准结冷胶为食品增稠剂、稳定剂[2]。
结冷胶具有高度的分散性和广泛的复配性,形成的溶液透明度高,有显著的温度滞后性和熔点可调性;结冷胶形成的凝胶能够耐酸、耐高温,还能抵抗微生物及酶的作用[3]。
结冷胶集增稠、悬浮、乳化等功能于一体,在食品中主要用于糖果、果酱和果冻、人造食品、水基凝胶、饮料乳品、宠物食品等[4]。
近年来随着研究的深入,结冷胶在食品以外领域的应用不断扩大。
本文对其在医药领域的研究与应用进行综述。
结冷胶的相对分子质量高达(2~3)×105,分子主链由重复的四糖单元组成,参与形成四糖单元的单糖依次为β(1→3)-D-葡萄糖、β(1→4)-D-葡萄糖醛酸、β(1→4)-D-葡萄糖、α(1→4)-L-鼠李糖。
天然结冷胶每个重复单元中的β(1→3)-D-葡萄糖2位都连有1个O-甘油酰基,6位每隔1个单位连有1个O-乙酰基,又称为高酰基结冷胶。

SDS-聚丙烯酰胺凝胶电泳(SDS—PAGE)实验原理和操作步骤实验原理:SDS—PAGE是对蛋白质进行量化,比较及特性鉴定的一种经济、快速、而且可重复的方法。
该法是依据混合蛋白的分子量不同来进行分离的。
SDS是一种去垢剂,可与蛋白质的疏水部分相结合,破坏其折叠结构,并使其广泛存在于一个广泛均一的溶液中.SDS蛋白质复合物的长度与其分子量成正比。
在样品介质和凝胶中加入强还原剂和去污剂后,电荷因素可被忽略.蛋白亚基的迁移率取决于亚基分子量。
试剂和器材:试剂:1. 5x样品缓冲液(10ml):0.6ml 1mol/L的Tris-HCl(pH6.8),5ml 50%甘油,2ml 10%的SDS,0.5ml巯基乙醇,1ml 1%溴酚蓝,0。
9ml蒸馏水。
可在4℃保存数周,或在-20℃保存数月。

凝胶贮液:在通风橱中,称取丙烯酰胺30g,甲叉双丙烯酰胺0。
8g,加重蒸水溶解后,定容到100ml。
过滤后置棕色瓶中,4℃保存,一般可放置1个月。

pH8。
9分离胶缓冲液:Tris 36。
3g ,加1mol/L HCl 48ml,加重蒸水80ml使其溶解,调pH8。
9,定容至100ml, 4℃保存。

pH6.7浓缩胶缓冲液: Tris 5。
98g ,加1mol/L HCl 48ml,加重蒸水80ml使其溶解,调pH6.7,定容至100ml,4℃保存。
5. TEMED(四乙基乙二胺)原液6.10%过硫酸铵(用重蒸水新鲜配制)7。
pH8。
3 Tris—甘氨酸电极缓冲液:称取Tris 6。
0g,甘氨酸28。
8g,加蒸馏水约900ml,调pH8。
3后,用蒸馏水定容至1000ml。
置4℃保存,临用前稀释10倍。
8. 考马斯亮蓝G250染色液:称100mg考马斯亮蓝G250,溶于200ml蒸馏水中,慢慢加入7.5ml 70%的过氯酸,最后补足水到250ml,搅拌1小时,小孔滤纸过滤。
器材:电泳仪,电泳槽,水浴锅,摇床。

通过拆分法获得艾司西酞普兰原料成本较高,操作条件苛刻,且拆分过程复杂;而采用不对称法可直接合成所要求的异构体,可明显提高药品质量和生产效率,降低终产物的生产成本。
两种方法各有特点,后一种合成法则为优选方法。

These eleven kinds of methods could be divided into two main groups, including resolution methods and asymetric synthesis routes. Escitalopram could be obtained by resolution methods with high cost, harsh conditions and complex workups. Citalopram isomer with an improved quality could be produced through direct asymetric synthesis route by increasing the production efficiency and reducing the cost of the target molecule significantly.
