经方二妙散对胶原诱发关节炎大鼠脾和关节中 悦匀砸晕粤 苑 表达的影响

【摘 要】 目的 研究不同浓度二妙散对胶原诱发关节炎（悦陨粤）大鼠脾脏和关节中 悦匀砸晕粤 苑 表达的影 响。方法 建立雌性 宰蚤泽贼葬则 大鼠 悦陨粤 模型，不同浓度二妙散对 悦陨粤 大鼠灌胃治疗，研究二妙散对 悦陨粤 大鼠脾脏
和关节 悦匀砸晕粤 苑 表达的影响。通过反转录聚合酶链反应（择砸栽鄄孕悦砸）和蛋白质印迹法分析不同实验组大鼠脾脏 和关节中 悦匀砸晕粤 苑 的表达特性。结果 悦匀砸晕粤 苑 mRNA 在关节中的表达量显著高于脾脏（孕约园援园缘）；在 悦陨粤 大 鼠脾脏中的表达量显著高于其他处理组（孕约园援园缘）；悦匀砸晕粤 苑 mRNA 在 悦陨粤 大鼠关节中的表达量显著高于其他处 理组（孕约园援园缘）；员援缘 早/噪早 二妙散药物组 悦陨粤 大鼠关节中 悦匀砸晕粤 苑 的蛋白表达量显著低于其他处理组（孕约园援园缘）。 结论 不同实验组关节和脾脏中 悦匀砸晕粤 苑 均有不同程度的表达，二妙散治疗 CIA 最适药物浓度为 员援缘 早/噪早， 悦匀砸晕粤 苑 参与 悦陨粤 大鼠炎性反应的形成，介导 CIA 炎性因子的形成和发展。

揖Abstract铱 Objective To investigate the effects of different concentrations of the traditional Chinese herbal formula, Er鄄miao鄄san on CHRNA 7 expression in spleen and joints of collagen鄄induced arthritis 渊CIA冤 rats. Methods In the study, we established a CIA model with female Wistar rats, and Er鄄miao鄄san of different concentrations was ad鄄 ministered to CIA rats by gavage. The expression characteristics of CHRNA 7 in the spleens and joints of the rats in different experimental groups were analyzed using qRT鄄PCR and western blotting. Results The expression of CHRNA 7 mRNA was significantly higher in the joints than that in the spleens (P<0.05), and the expression in the spleens and joints of CIA rats was significantly higher than that in the spleens of other treatment groups (P<0.05). The expression of CHRNA 7 in the joints of rats treated with 1.5 g/kg of Er鄄miao鄄san was significantly lower than that in other treatment groups (P<0.05). Conclusion CHRNA 7 was expressed to different extents in the joint and spleen tis鄄 sues of rats in the different experimental groups. The optimal concentration of Er鄄miao鄄san for CIA is 1.5 g/kg. CHRNA 7 is involved in the generation of the inflammatory reaction in CIA rats via mediating the formation and de鄄 velopment of inflammatory factors related to CIA.
