分析检测基于LC-MS/MS的蔬菜农药残留基质效应分析单晓丽1,周 峰1,李 东1,王国洋2(1.安丘市检验检测中心有限公司,山东潍坊 262100;2.山东柠檬生化有限公司,山东潍坊 262100)摘 要:为明确蔬菜农药残留对于基质效应的影响,本文选取韭菜、芹菜、茄子3种蔬菜,利用液相色谱-串联质谱法对蔬菜中6种农药残留进行测定,通过基质和溶剂标准曲线的斜率,判定其基质效应。

试验结果表明,不同蔬菜中农药残留量的基质效应主要为信号抑制作用。

因此,在后期农药检测中,可通过使用基质标准曲线进行定量测定,从而提高检测结果的准确性,对于保障食品安全具有重要意义。

The six pesticide residues in vegetables were determined by liquid chromatography-tandem mass spectrometry. The matrix effect was determined by the slope of the matrix and solvent standard curve. The results showed that the matrix effect of pesticide residues in different vegetables was mainly signal inhibition. Therefore, in the later pesticide detection, the matrix standard curve can be used for quantitative determination, so as to improve the accuracy of the test results, which is of great significance for ensuring food safety.Keywords: LC-MS/MS; vegetable pesticide residues; matrix effect随着蔬菜农药残留问题的日益突出,农药残留检测已成为当前食品安全研究领域的重点内容。

采用ACE EXCEL 3氐-AR ( 164 mmx4.9 mm , 3 jm )色谱 柱,以0. 3%甲酸水溶液和0. 3%甲酸乙睛为流动相梯度洗脱分离,流速为0. 5。

mL/mm ,柱温为3。

C ,自 动进样器温度为4 C ,设置六通阀切换保护质谱系统。

质谱分析采用ESI 离子源,正离子平行反应监测 (PRM )扫描模式,外标法定量。

NDMA 在1.00~100.00 11/1^范围内线性良好,相关系数()为0.999 7,检 岀限和定量下限分别为0. 24 n/mL 和1. 00 n/mL ,在法莫替丁及其制剂中的平均回收率为98. 5% - 108% , 相对标准偏差(RSD )为2. 3% ~ 6. 7%。

高效液相色谱质谱法测定食品接触材料着色剂中芳香胺类物质花 锦1*,肖利龙2(1.太原海关技术中心,山西太原 030001;2.太原学院,山西太原 030001)摘 要:目的:建立高效液相色谱质谱法测定食品接触材料中着色剂中芳香胺类物质。

方法:用乙腈超声提取试样中芳香胺类物资,改进QuEChERS 法净化,使用ACQUITY HSS T3柱以水和甲醇溶液进行梯度洗脱,采用电离喷雾电离方式(ESI+),通过多反应监测(MRM)定量。

结果:该法测定时间短,在5 min 内出峰,线性关系良好,相关系数r 大于0.99,回收率在67.0%~95.0%,相对标准偏差(RSD )均小于15%。

该法快速、简便、精确度高、准确度高,对食品级着色剂中芳香胺残留有普遍适用性,其他着色剂可参考使用。

关键词:QuEChERS;食品接触材料;芳香胺;着色剂;液相色谱质谱联用食品接触材料通常是指与食物直接接触的物质,在带来生活上便利的同时也带来了食品安全问题。

着色剂是指能给予或改变食品包装材料颜色的物质,通常包括染料和颜料两种,此外还有一些特殊物质如光学增量剂、荧光漂白剂等本身没有颜色但能提高产品光亮的物质,主要用于提高产品的美观。

因此食品接触材料中会存在着色剂,其中偶氮类染料最为常见。

偶氮染料具有色谱范围广,牢度强,着色快,色光好等优势,因此偶氮染料的使用较为广泛,不仅应用于食品接触材料,还应用于皮革、纺织、化妆品、纸张等生活必需品[1]。

早在20世纪,有日本人发现偶氮染料中的溶剂黄可导致老鼠的肝细胞发生癌变,之后1905年德国也从染料品红、萘胺等物质中发现了偶氮染料的致癌性[2]。

偶氮染料主要通过分解为芳香胺来释放有毒有害物质,分解过程见 图1。

图1 芳香族偶氮染料分解为芳香胺芳香胺类物质对人类健康和环境具有危害性,尤其是2,4-二甲基苯胺、2-萘胺、对氯苯胺等,进入人体之后会产生高铁血红蛋白引起一系列疾病[3]。

由于食品安全方面芳香胺的危害较大,而我国目前对于食品接触材料中芳香胺的检测技术还不够全面,只对接触材料进行了研究,对其中着色剂没有深入探讨,因此芳香胺的检测方案急需优化以及扩充。

结果表明:在低盐溶液中,随着水解时间的延长,溶解度显著升高(P <0.05),表面疏水性先降低后升高,乳化性先升高后降低;当水解时间为40 min 时,水解度为4.15%,鸭肉MP 的溶解度和乳化性达到最高,分别达到60.57%和21.2 m 2/g ;表面疏水性最低(40.85 BPB/μg )。

方法:采用高效液相色谱2大气压化学电离2质谱法(HP LC 2APCI 2MS )。

实验条件:正离子检测模式,喷雾电压3000V ,毛细管温度250℃,APCI 源温度400℃,corona 电流4μA 。

鞘气(高纯液氮)压力35K Pa 。

质谱扫描范围为300~900amu ,扫描时间1s ,Q1宽度为017。

液相色谱的固定相为Z orbax Extend C 18柱(416mm ×250mm ,5μm )。

流动相为二氯甲烷2乙腈(35∶65),流速1m L ・min -1。

柱温25℃。

结果:共鉴定出12种甘油三酯。

结论:此测定结果可作为薏苡仁注射液指纹图谱中组分的定性依据。

大豆油是制备乳剂的重要原辅料,其含量直接影响乳剂的成型质量,在氟比洛芬酯注射液中大豆油作为氟比洛芬酯的载体,有利于氟比洛芬酯达到靶向部位,更好地吸收。

但由于大豆油为长链甘油三酸酯,紫外特征不强,常规的紫外检测器无法完成对其的测定。

国内仅有一个厂家生产该品种,现行质量标准[1]中未对大豆油进行含量控制。

笔者采用高效液相色谱法分离,利用蒸发光散射检测器测定氟比洛芬酯注射液中大豆油的含量,有效地解决了大豆油在该注射剂中的检测问题,方法准确,操作简便。
