要:目的:该实验通过电针针刺脾气虚大鼠的“三阴交”和“足三里”穴,观测实验后大鼠的血清铁(SI)、 铁蛋白(Fe)、总铁结合力(TIBC)、转铁蛋白(Tf)和转铁蛋白受体(TfR)的变化,探讨脾气虚对血液的分子生物学 的病理影响,同时尝试研究针刺治疗脾气虚方法和作用机制。方法:应用随机分组的方法,将 SD 大鼠 40 只,平均 分为 4 组,依次为正常组、模型组、经穴组、非经非穴组,以饮食不节、过度疲劳为造模因素进行脾气虚造模。造模成 功后,除正常组进行正常喂养外,模型组每日上午继续造模,下午不给予针刺,每日捆绑 20 min。经穴组和非经穴 组进行针刺治疗。试验结束后,各组大鼠取血清用 ELISA 方法检测大鼠血清中血清铁含量、铁蛋白含量、总铁结合 力。采用荧光定量 PCR 方法检测肝细胞转铁蛋白 mRNA 和转铁蛋白受体 mRNA 的表达,采用 spss 19.0 统计软件 对实验数据进行分析。结果:(1)正常组大鼠血清铁含量与模型组相比显著降低(P<0.01),经穴组血清铁含量与模 型组相比显著降低(P<0.01),经穴组与正常组相比无统计学意义(P>0.05),模型组与非经非穴组比较无统计学意 义(P>0.05)。(2)正常组大鼠铁蛋白含量、总铁结合力与模型组相比显著升高(P<0.01),经穴组铁蛋白含量与模型 组相比显著升高(P<0.01),经穴组与正常组相比无统计学意义(P>0.05),模型组与非经非穴组比较无统计学意义 (P>0.05)。(3)电针双侧“足三里”“三阴交”穴对脾气虚大鼠转铁蛋白 mRNA 的表达量影响模型组大鼠转铁蛋白 mRNA 的表达量、转铁蛋白受体 mRNA 与正常组相比显著降低(P<0.01),经穴组血清转铁蛋白 mRNA 的表达量与 模型组相比显著升高(P<0.01),经穴组与正常组相比无统计学意义(P>0.05),模型组与非经穴组比较无统计学意义 (P>0.05)。结论:(1)脾气虚时大鼠体内的铁代谢出现异常。(2)针刺“足三里”和“三阴交”可以调节脾气虚大鼠 体内的铁代谢,起到益气健脾的作用并治疗脾气虚。(3)转铁蛋白 - 转铁蛋白受体系统很可能是脾主升清和主统血 功能在物质层面的一种表现。(4)铁代谢异常可能与脾气虚的病理本质有关。

group,the acupoint group rose significantly(P<0.01),the difference between the acupoint group and the normal group had no statistical significance(P>0.05),and the difference between the model group and the sham acupoint group had no statistical significance(P>0.05).(3)Compared with the normal group, the model group declined significantly in expression of transferrin mRNA and the expression of transferrin receptor mRNA(P<0.01);compared with the model group,the acupoint group rose significantly(P<0.01) in transferrin content,the difference between the acupoint group and the normal group had no statistical significance(P>0.05),and the difference between the model group and the sham acupoint group had no statistical significance(P>0.05).Conclusions:(1)Iron metabolism of rats became abnormal at spleen Qi deficiency.(2)Acupuncture of“Zusanli”and“Sanyinjiao”can adjust iron metabolism of rats with spleen Qi deficiency,replenish qi to invigorate spleen and treat spleen Qi deficiency.(3)The transferrin transferrin receptor system is likely to reflect the“spleen governing ascending clear”and“spleen governing blood”in physical level. 4. Abnormal iron metabolism may be related to the pathological nature related to spleen-QI deficiency.
脾是人体的一个重要器官,而现代医学和中 医藏象学对其解释却不尽相同。现代医学认为脾 是位于腹腔左上方的一个免疫器官。它的主要 作用是造血和储血,同时清除过滤血液和参与免 疫反应—— B 淋巴细胞就储存在脾脏内。当其功 能出现异常主要会影响人体的免疫功能。而藏象 学说认为脾为后天之本,血气生化之源,位于中 焦五行属土,是重要的消化器官。脾还具有升清、 运化和统血的功能。人所食五谷杂粮由脾胃消化 为水谷之精微,水谷精气在脾内化成血液,再由 脾气升清、发散至四肢百赅,强健肌肉筋膜使内 脏固定而不至脱垂。当脾的功能出现异常会大便 溏泄、面色萎黄、肌瘦无力、面色苍白、少气或懒 言等一些列症状。铁是人体必需的金属元素,它 广泛分布于人体的各个组织和器官中,而铁在血 液中的作用尤为重要。当人体缺乏铁时,最常见 的是缺铁性贫血,人们将会出现有乏力易疲、心 悸气促、面黄无力等症状,免疫力也会下降,同时 神经系统亦会受影响,导致情绪受影响和注意力 不集中。这些症状与脾气虚的症状有很多是相同 的,此次实验以脾气虚大鼠为模型,对大鼠进行 分组电针针刺治疗。通过对血清铁、总铁结合力、 转铁蛋白和转铁蛋白受体等指标进行检测,分析 对比各组之间数据的差异,讨论和研究脾气虚对 铁代谢的影响。 1 材料与方法 1.1 实验材料 1.1.1 实验动物

此次实验的对象为脾气虚模型的大鼠而大鼠的经脉穴位与人类的有所不同脾胃二经只有足三里与三阴交故选此二穴为本实验的最佳选择通过检测血清铁总铁结合力转铁蛋白和转铁蛋白受体这四项指标发现模型组数据与正常组数据相比较均有差异p001说明脾气虚状态下机体的铁代谢会出现异常

Abstract:Objective:In this experiment,electroacupuncture was given to the model rats with spleen Qi deficiency,to observe its influence on serum iron(SI),total iron binding capacity(TIBC),transferrin (Tf)and transferrin receptor(TfR),study the influence of spleen Qi deficiency on hemal molecular biology and explore the methods and mode of action of acupuncture treatment for spleen Qi deficiency. Methods: A total of 40 SD rats were randomly divided into four groups:normal group,model group,acupoint group, and sham acupoint group,10 in each group. Model group,acupoint group and sham acupoint group were modeled for 14 days based on the factors including irregular diet and excessive fatigue. After modeling, except for the normal group that received normal feeding,the model group was modeled every morning, not acupunctured in the afternoon but was bound for 20 min per day;acupuncture group and non acupoint group and acupuncture treatment. After the test,serums of each group of rats were tested with ELISA to detect the content of serum iron and the total iron binding capacity. Fluorescence quantitative polymerase chain reaction(PCR)was used to detect expression of liver cell transferrin mRNA and transferrin receptor mRNA,and the experimental data was analyzed with spss19.0 statistical software. Results:(1)Compared with the model group,the normal group declined significantly in serum iron content(P<0.01),the acupoint group declined significantly(P<0.01),the difference between the acupoint group and the normal group had no statistical significance(P>0.05),and the difference between the model group and the sham acupoint group had no statistical significance(P>0.05).(2)Compared with the normal group,the model group declined significantly in serum ferritin and total iron binding capacity(P<0.01);compared with the model
