尼罗红荧光染色溶液产品说明书(中文版)主要用途尼罗红荧光染色溶液是一种旨在通过与脂类物质的结合并发出荧光检测信号,快速、敏感、可靠地活体定量测定细胞内脂类成分的常用荧光染料。
适用于各种细胞包括细菌细胞,也用于蛋白质电泳染色。
产品严格无菌,即到即用,性能稳定,着色清晰灵敏。
技术背景尼罗红染色剂(9-diethylamino-5H-benzo[alpha]-phenoxazine-5-one;Nile red)是一种亲脂性的恶嗪类荧光染料。
与脂类物质包括腊酯(wax ester)和三酰甘油(triacylglycerol)以及各种脂肪酸结合后,在激发波长543nm的激发下,显示强烈桔红色荧光(散发波长598nm)。
同时在紫外光的照射下显示红色。

Azide compounds should be diluted with running water before discarding. These precautions are recommended to avoid deposits in plumbing where explosive conditions may develop.1. DescriptionThis product is for research use ponents1 mL CD45 (TIL) MicroBeads, mouse:MicroBeads conjugated to monoclonal anti-mouse CD45 antibodies (isotype: rat IgG2b).CapacityFor 10⁹ total cells, up to 100 separations.Product format CD45 (TIL) MicroBeads are supplied in buffercontaining stabilizer and 0.05% sodium azide.StorageStore protected from light at 2−8 °C. Do not freeze. The expiration date is indicated on the vial label.1.1 Principle of the MACS® SeparationFirst, the CD45+cells are magnetically labeled with CD45 (TIL) MicroBeads. Then, the cell suspension is loaded onto a MACS® Column, which is placed in the magnetic field of a MACS Separator. The magnetically labeled CD45+ cells are retained within the column. The unlabeled cells run through; this cell fraction is thus depleted of CD45+ cells. After removing the column from the magnetic field, the magnetically retained CD45+ cells can be eluted as the positively selected cell fraction. To increase the purity, the positively selected cell fraction containing the CD45+ cells must be separated over a second column.1.2 Background informationCD45 (TIL) MicroBeads, mouse have been developed for theisolation of tumor-infiltrating leukocytes (TILs) from single-cell suspensions of solid mouse tumors. To remove dead cells, we recommend using the Dead Cell Removal Kit (# 130-090-101).2.2 Magnetic labeling▲ Cells can be labeled with MACS MicroBeads using the autolabeling function of the autoMACS® Pro Separator. For more information refer to section 2.4.▲ Work fast, keep cells cold, and use pre-cooled solutions. This will prevent capping of antibodies on the cell surface and non-specific cell labeling.▲ Volumes for magnetic labeling given below are for up to 10⁷ total cells. When working with fewer than 10⁷ cells, use the same volumes as indicated. When working with higher cell numbers, scale up all reagent volumes and total volumes accordingly (e.g. for 2×10⁷ total cells, use twice the volume of all indicated reagent volumes and total volumes).▲ For optimal performance it is important to obtain a single-cell suspension before magnetic labeling. Pass cells through 30 µm nylon mesh (MACS SmartStrainers (30 µm), # 130-098-458). Moisten filter with buffer before use.▲ The recommended incubation temperature is 2–8 °C. Higher temperatures and/or longer incubation times may lead to non-specific cell labeling. Working on ice may require increased incubation times.1. Determine cell number.2. Centrifuge cell suspension at 300×g for 5 minutes. Aspiratesupernatant completely.3. Resuspend cell pellet in 90 µL of buffer per 10⁷ total cells.▲ Note: Always use freshly prepared buffer.4. Add 10 µL of CD45 (TIL) MicroBeads per 10⁷ total cells.5. Mix well and incubate for 15 minutes in the dark in therefrigerator (2−8 °C).6. (Optional) Add staining antibodies according tomanufacturer’s recommendations.7. Add buffer to a final volume of 500 μL for up to 5×10⁷ cells.▲Note: If more cells were used, split the sample onto multiple columns duringmagnetic separation.▲Note: For higher cell numbers, scale up buffer volume accordingly.8.Proceed to magnetic separation (2.3).2.3 Magnetic separation▲ Choose an appropriate MACS Column and MACS Separator according to the number of total cells and the number of CD45+ cells. For details refer to the table in section 1.4.▲ Note: MS Columns are recommended for highest purity of CD45+ cells. LSColumns are recommended for highest recovery of CD45+ cells.▲ For optimal performance it is important to obtain a single-cell suspension before magnetic separation. Pass cells through 30 µm nylon mesh (Pre-Separation Filters (30 µm), # 130-041-407) to remove cell clumps which may clog the column. Collectunlabeled cells that pass through and combine with theflow-through from step 3.MS: 3×500 µL LS: 2×1 mL▲ Note:Perform washing steps by adding buffer aliquots as soon as the columnreservoir is empty.5. Remove column from the separator and place it on a suitablecollection tube.6. Pipette the appropriate amount of buffer onto the column.Immediately flush out the magnetically labeled cells by firmly pushing the plunger into the column.MS: 1 mL LS: 3 mL7. Switch on the instrument for automatic initialization.2. Go to the Reagent menu and select Read Reagent. Place sample and collection tubes into the Chill Rack.4. G o to the Separation menu and select the reagent name foreach sample from the Labeling submenu (the correct labeling, separation, and wash protocols will be selected automatically).5. Change the tube and buffer when switching to a different plasmid DNA/siRNA or cell type.•The volume of plasmid DNA or siRNA added to the tranfection reaction should not exceed 10% of the total transfection volume.•Visit for a library of electroporation protocols for a variety of commonly used cell types.Prepare cellsFor the appropriate volume of medium to use based on cell density, or plating volumes for other plate formats, see “Amount of reagents” on page 2.1.Cultivate the required number of cells (70% to 90%confluent on the day of transfection) by seeding a flaskcontaining fresh growth medium 1 to 2 days prior toeletroporation.2.On the day of the experiment, pre-warm aliquots of culturemedium containing serum, PBS (without Ca2+and Mg2+), and Trypsin/EDTA solution to 37°C.3.Rinse the cells with PBS (without Ca2+and Mg2+), thentrypsinize the cells with the Trypsin/EDTA solution.4.Take an aliquot of trypsinized cell suspension, then countcells to determine the cell density.5.Harvest the cells in growth medium containing serum.6.Transfer cells to a 1.5-mL microcentrifuge tube or a 15-mLconical tube, then centrifuge the cells at 100 - 400 × g for 5 minutes at room temperature.7.Wash cells with PBS (without Ca2+and Mg2+) bycentrifugation at 100 - 400 × g for 5 minutes at roomtemperature.8.Aspirate the PBS, then resupsend the cell pellet inResuspension Buffer R (or Resuspension Buffer T forprograms ≥ 1900 V) at a final density of 1.0 × 107 cells/mL for adherent cells or 2.0 × 107 cells/ml for suspension cells.Gently pipette the cells to obtain a single cell suspension.IMPORTANT! Avoid storing the cell suspension for more than 15 to 30 minutes at room temperature. This will reduce cell viability and transfection efficiency.9.Prepare 24-well plates by filling the wells with 500 μL ofculture medium containing serum and supplements, butwithout antibiotics. 肠杆菌科细菌编码微量鉴定管使用中的注意事项于建华,闫文婧(解放军第医院检验科,山东淄博)关键词:肠杆菌;鉴定;编码中图分类号:肠杆菌科的细菌是造成各种感染的常见菌,对于它的鉴定,虽然可用全自动微生物鉴定仪,但大多数中、小医院不具备条件,而按传统方法鉴定必须进行一系列生化反应和血清学试验,不仅造成人力、材料的浪费,而且需要长时间等待结果甚至造成漏检。
采用肠杆菌科编码的方法,简便易行,结果及时可靠。
但该法在使用编码微量鉴定管过程中,发现有一些影响鉴定结果的因素,必须引起注意。
!接种生化鉴定管分离培养出的细菌,首先观察菌落特征。
氧化酶阴性、发酵型的革兰阴性杆菌可按肠杆菌科处理。
取单一菌落或纯的细菌培养物接种生化管,应每接种一管均需灭菌,但微量生化管数量多,耗时长,在实际工作中多采用一次取菌连续接种!管,为了避免生化管成分交叉感染造成假性结果,可合理配组减少影响。
如:()赖氨酸、鸟氨酸、氨基酸;()乳糖、卫茅醇、双糖铁;()硫化氢、尿素;()枸橼酸盐、蛋白胨水。
组间顺序可变,与单一管接种是一致的。
另外,在主要试验和辅助试验接种过程中,苯丙氨酸、葡萄糖酸盐管接种量要大,否则会造成假阴性;枸橼酸盐管接种量要小,避免将培养基成分带入而出现可疑反应;半固定接种应在中间垂直穿入近底部,但管径小,扩散生长并不明显,特别是动力弱的菌株,沿管壁穿刺较易观察,同时参考悬滴法或压片法。
氨基酸脱羧试验一定用灭菌液体石蜡!封口,否则会产生不稳定的胺而被氧化造成假阳性。
"结果判断肠杆菌科的细菌多属于快速生长菌,采用肠杆菌微量鉴定管时多数结果,!时观察结果。
如糖、醇类(迟缓分解除外)、苯丙氨酸、葡萄糖酸盐、靛基质、硫化氢、氨基酸脱羧试验等。
但也有特殊者,观察不妥,如:()甲基红试验:克雷伯菌、肠杆菌属的常见菌,如后加试剂观察呈阳性(红),而后变为阴性(黄);()尿素酶试验,!呈阳性(红色),不变为阴性,当疑为变形杆菌属应分段观察,此菌属!可迅速分解尿素,具有鉴别意义。
