1.安装软件(Installation & Configuring Components)1. 选择“Launch CHROMELEON Setup”,根据提示安装软件;2. Server Monitor”;3. 点击“Start”,等待变色龙图标变成灰色,关闭“Server Monitor”;4. Server Configuration”;5. 双击计算机名称;6. 选择“Dongle”,在“Key Code”处输入软件密码;点击“确定”;7. 点击“Add Sharable device”;8. 选择“Dionex LAN Card”,点击“OK”;9. 选择网卡,点击“Bind to DX-LAN”;10. 点击“是(Y)”;11. 点击“确定”;12. 关闭“Server Configuration”,保存设置,接通仪器电源,重新启动计算机;13. 再次进入“Server Configuration”,点击“Add timebase”;14. 为新建立的“timebase”命名;15. 点击“Add device”;16. 选择需要添加的设备,点击“OK”;17. 取消“Demo Mode”,打开“Device ID”,选择“GP50:xxxxxx”,点击“确定”,此时GP50面板左下角的“LOCAL”变成“LOCKED RMT”;17-1. 如果安装EG40,点击“Eluent Generator”,选择“Installed”,打开“ID”,选择“EG40:xxxxxx”,将“Offset V olume”改为0µl,在“Cartridge Information”的“Serial”处输入KOH/MSA淋洗液储罐的序列号(S/N),点击“确定”;18. 重复“15”和“16”的操作,选择CD25A,点击“OK”;19. 关闭“Demo Mode”,打开“Device ID”,选择“CD25A:xxxxxx”,点击“确定”,此时CD25A面板左下角的“LOCAL”变成“LOCKED RMT”;20. 重复“15”和“16”的操作,选择“Remote Inject”,点击“OK”;21. 如图所示进行选择后,点击“确定”;22. 关闭“Server Configuration”,保存设置。

对于待检测的细胞或组织样品,经过生物素标记和后续的DAB 显色等步骤,即可在普通光学显微镜下观察到凋亡细胞。

细胞在发生凋亡时,会激活一些DNA 内切酶,这些内切酶会切断核小体间的基因组DNA 。

细胞凋亡时抽提DNA 进行电泳检测,可以发现180-200bp 的DNA ladder 。

基因组DNA 断裂时,暴露的3'-OH 可以在末端脱氧核苷酸转移酶(Terminal Deoxynucleotidyl Transferase, TdT)的催化下加上生物素(Biotin)标记的dUTP(Biotin-dUTP),随后和辣根过氧化物酶(HRP)标记的Streptavidin (Streptavidin-HRP)结合,最后在HRP 的催化下通过DAB 显色来显示凋亡细胞,从而可以通过普通光学显微镜检测到凋亡的细胞,这就是TUNEL(TdT-mediated dUTP Nick-End Labeling)法检测细胞凋亡的原理。

本试剂盒有如下优点。

(1) 高灵敏度:背景染色极低,阳性染色强,可以在单细胞水平检测到细胞凋亡,同时由于凋亡早期就有DNA 断裂,可以检测到早期的细胞凋亡。

(2) 特异性好:TUNEL 检测时通常更容易标记凋亡细胞,而不容易标记坏死细胞。

(3) 快速:仅需约2-3个小时即可完成。

(4) 应用范围广:可以用于检测冷冻或石蜡切片中的细胞凋亡情况,也可以检测培养的贴壁细胞或悬浮细胞的凋亡情况。

當我們進入軟體後,若發現電腦和儀器並未連線,可按下鍵進行連線的工作。

當我們要開始新的實驗,先按下鍵,建立新的方法。

依照您的機型選擇所要設定的模式。

選擇F-2500/F-4500Method進入後,分頁依序為一般選項(General),在測定方法(Measurement)部分可分為:波長掃描(Wavelength scan)時間掃瞄(Time scan)吸光度測定(Photometry)Operator:使用者姓名Instrument:儀器名稱Sampling:除非您有使用HITACHI其他的配備,例如Sipper…等,不然請選擇NoneComment:可以記錄此實驗說明或是實驗內容。

Use Sample Table:是否要使用Sample Table來設定樣品的數據和參數。

波長掃描(Wavelength scan)在選擇波長掃描後,點選儀器參數設定(Instrument),在掃描模式(Scan mode)中選擇所要改變的是激發光(Excitation)或是放射光(Emission)。

數據模式(Data mode)設定測量方式是為螢光(Fluorescence)或冷光(Luminescrence)並設定激發光及放射光的波長範圍。

掃瞄速度(Scan speed):決定掃描時間的長短,但掃描速度愈慢,解析度愈好。

延遲時間(Delay):需要穩定的試樣,設定一段延遲時間才開始測量。

激發光光柵(EX Slit)放射光光柵(EM Slit):設定光柵大小。

光電倍增管通電電壓(PMT Voltage):可由此調整感度,值愈,感度愈好,但雜訊愈高。

應答速度(Response):光電倍增管每隔多少秒偵測一次,一般設Auto。

pigment is inorganic in nature and is composed of three common raw materials: aluminum, magnesium fluoride and chromium. Propylene glycol n-propyl ether is used as a wetting agent to reduce dusting during weighing and mixing of the pigment.All eight standard colors of ChromaFlair pigment are manufactured from the same three raw materials.All colors have an aluminum metal core. This highly reflective layer serves two purposes. It provides the reflective surface necessary for light interference, which produces the highly chromatic color shift characteristic of ChromaFlair pigments. Secondly, the reflective aluminum layer provides hiding by reflecting light prior to reaching the substrate.DispersionThe shape and features of the ChromaFlair pigment flake need to be considered when dispersing to form a pigment paste or slurry. Milling or crushing of the flake can lead to breakdown in the flake and/or distortionof the planar surface of the flakes. The recommended dispersion method is to incorporate the pigment into the dispersion medium with a high speed, low shear disperser. Sand or media mills are not recommended for processing. Blending will be sufficient to disperse and wet the individual flakes. ChromaFlair pigment is readily dispersible in most vehicle systems. Once dispersed, ChromaFlair pigment remains in a stable suspension. Settling may occur over time, as with other pigment dispersions, due to the fact that the specific gravity is soft and loose and is easily reincorporated into the dispersion vehicle with minimal agitation.Settling may be improved through several methods: increased system viscosity, dispersants, thixotrophs and other settling aids. Increased system viscosity provides greater resistance to the low shear forces acting on the pigment during storage. Dispersants are effective due to their structure. One end of the dispersant is attracted to the surface of the pigment particle while the other end, usually a long chain polymer, provides steric hindrance. Thixotrophic agents such as clay, silica and cellulose acetate butyrate form three-dimensional structures through hydrogen bonding, thus providing steric hindrance.LoadingT he hue shifts and chromaticity of ChromaFlair pigment depend on the interaction of light with the pigment surface. Most paint formulations are blends of several colors and types of pigments to produce the desired color and/or appearance effect. ChromaFlair pigmentis often referred to as a “stir in” pigment and has been found compatible in a wide range of standard formulations. In an equal weight of sample, Gold/Silver 080 will contain 2.6 times as many individual pigment particles. The larger number of flakes translates to improved hiding power and reduced pigment loading.Loading level of ChromaFlair pigment will dependon several factors including substrate, applied film thickness, hiding requirements and other pigments in the paint system. In general, levels of 1% to 12% by weight (based on total solids) of ChromaFlair pigment are recommended for most formulations.T able 1 lists guideline levels for each of the eight standard ChromaFlair pigment colors for various applications. These levels will produce varied substrate coverage resulting in a range of color effects. A basecoat loading provides opacity and the most spectacular color shift effect. A midcoat loading applied over an opaque basecoat can provide dramatic color shifts at reduced pigment loading. Flemming固定液使用说明Flemming固定液Flemming Fixative Solution编号名称规格单位北京华越洋WG08810Flemming固定液100ml,500ml瓶Flemming固定液用途:Flemming固定液为铬酸-锇酸-醋酸混合固定液,现配现用。

该固定液对脂肪组织既有固定作用,又有染色作用,对有髓神经纤维也有固定兼染髓鞘的作用。

该液不适合做HE的染色。

应用该液固定的组织,切片不能太大过厚,一般1-2毫米。

固定时间1-3天,后经水洗,保存于80%酒精中。

可用于各种植物组织的固定。

保存条件:RT,有效期一年。

Flemming固定液固定时的注意事项:(1)固定液量:20倍于材料的体积,以免固定液被稀释。

(2)选择合适的固定液:渗透力强的固定液既可以迅速进入组织中,又不使组织过度收缩或膨胀。

(3)固定的时间要合适:与材料的大小和温度有关:材料大则时间长,材料小则时间短;温度高则时间短,温度低则时间长。

经固定的材料如不及时使用,可以经过90%酒精换到70%酒精中各半小时,再换入一次70%酒精,在0-4℃冰箱内可保存半年。

经过较长时间保存的材料进行观察前可以换新的固定液再处理一次,效果较好。

Flemming固定液固定的条件:(1)迅速渗入组织,杀死原生质。

在杀死的短时期中,细胞形态不致有所变化。

(2)必须具有渗透力,对组织各部分的渗透力相等,可使组织内外完全固定。

(3)尽可能避免使组织膨胀或收缩。

(4)使细胞内的成分凝固或沉淀。

(5)增加细胞内含物的折光程度,易于鉴别。

(6)增加媒染作用和染色能力。

(7)使组织变硬,具有一定的硬度。

(8)固定以后能起到保存的作用。

(9)在凝固原生质以后,增加细胞对于各种穿透的抵抗力,不致于因以后的处理而使固定的原生质变形。

Flood screen and frame thoroughly with garden type hose, then dry.MIXMix emulsion and sensitizer according to instructions onbottle. Let emulsion stand at least two hours before using.COATFill scoop coater with room temperature emulsion. Slowly apply first coat to print side. Then coat squeegee side with one to three coats depending upon thickness required. If thicker stencil is required, additional coats may be applied to print side after initial drying of stencil. Be sure to dry thoroughly between coats. DRYThoroughly dry screen in horizontal position, print side down, using a dark, clean drying cabinet. Temperatureshould not exceed 110o F (43oC).EXPOSEPlace emulsion side of photopositive in contact with print side of screen. DEVELOPGently spray both sides of screen with tepid water, wait 30 seconds then gently wash print side of the screen until image is fully open. Rinse both sides thoroughly. Dry screen completely and you are ready to print.RECLAIMApply Chroma/Strip™ screen reclaimer to both sides of screen. Scrub area to be reclaimed with a stiff nylon brush to ensure entire surface is wet and let it work a few moments until stencil begins to dissolve. Remove stencil residue with pressure washer, thenrinse with garden type hose, thoroughly flooding screen and frame. EXPOSURE GUIDELINESNote : Exposure times are suggested only as a guide. Use the Chromaline Exposure Calculator to determine optimal exposure times. Individual exposure times may vary depending upon equipment used, bulb age, and other shop conditions.SUGGESTED M INIMUM E XPOSURE G UIDELINES Exposure times were determined by using the Chromaline Exposure Calculator and the Chromaline UV Minder. Exposure times were set for a 5KW unit at 40" from the frame. All screen mesh was yellow in color. Screens were coated wet on wet, once on print side and twice on squeegee side.AVOID FAILURE: Dual cure emulsions have a very wide exposure latitude. Underexposed stencils often appear acceptable, but premature breakdown can occur on the press. When determining exposure speed, always overexpose your test stencil, then reduce exposure time until acceptable image quality is achieved. Flicker如何發生?
1.液晶分子的特性液晶分子無法長時間固定在某 個電位之下,一但長時間固定於 某個電位下,即使將電位取消, 液晶分子會因特性被破壞而無法 在因應電位的改變來轉動。
2.液晶分子的驅動電路因液晶分子的特性關系,所以在 設計液晶驅動電路時,常會以正、 負極性的電位去驅動相同的灰階 畫面,但此種驅動電路的參考電 位只要有點誤差,就會導致正、 負極性的亮度不同,也就會產生 Flicker。

JEITA量測模式(二)
JEITA 量測方式:
步驟1.量測LCD面板,將LC D亮度值轉換成數位訊號.
步驟2.將待測光源的數位訊號經 由FFT轉換成每個頻率的 量能強度。
步驟3.在將每個頻率的量能強度 乘上Flicker weighing. 步驟4.依據下方JEITA 標準所提 供的公式代入,求出最大 的Flicker 量。 Flicker amount = 10*log (Px/P0)[dB]

Flicker Weighting Factors
根據研究,人眼對flicker的感受,從30Hz開始下降, 當頻率超過60Hz時,人眼對flicker的感受,會大幅度 的下降也就無法感受到flicker的存在。

假如液晶顯示器的亮度變化如右 圖所顯示,亮度的AC成份等於 Vmax-Vmin, 亮度DC成份等於 (Vmax+Vmin)/2。
