In this paper; we report an unexpected; acute effect of GDNF on A-type potassium channels; leading to a potentiation of neuronal excitability; in the dopaminergic neurons in culture as well as in adult brain slices. Further; we show that GDNF regulates the K+ channels through a mechanism that involves activation of MAP kinase. Thus; this study has revealed; for the first time; an acute modulation of ion channels by GDNF. Our findings challenge the classic view of GDNF as a long-term survival factor for midbrain dopaminergic neurons; and suggest that the normal function of GDNF is to regulate neuronal excitability; and consequently dopamine release. These results mayalso have implications in the treatment of Parkinson’s disease.Due to a direct competition and conflict of interest; we request that Drs. XXX of Harvard Univ.; and YY of Yale Univ. not be considered as reviewers. With thanks for your consideration; I amSincerely yours;case2Dear Editor; We would like to submit the enclosed manuscript entitled "Ca2+-binding protein frequenin mediates GDNF-induced potentiation of Ca2+ channels and transmitter release"; which we wish to be considered for publication in Neuron. We believe that two aspects of this manuscript will make it interesting to general readers of Neuron. First; we report that GDNF has a long-term regulatory effect on neurotransmitter release at the neuromuscular synapses. This provides the first physiological evidence for a role of this new family of neurotrophic factors in functional synaptic transmission. Second; we show that the GDNFeffect is mediated by enhancing the expression of the Ca2+-binding protein frequenin. Further; GDNF and frequenin facilitate synaptic transmission by enhancing Ca2+ channel activity; leading to an enhancement of Ca2+ influx. Thus; this study has identified; for the first time; a molecular target that mediates the long-term; synaptic action of a neurotrophic factor. Our findings may also have general implications in the cell biology of neurotransmitter release.0630投稿写作某杂志给出的标准Sample Cover Letterthe example used is the IJEBCase 3Sample Cover Letterthe example used is the IJEBDear Editor of the please type in journal title or acronym:Enclosed is a paper; entitled "Mobile Agents for Network Management." Dear Editor of the 此处写期刊英文名称:Thank you for your letter and thanks for your careful review to our manuscript entitled "论文题目". We would like to submit the revised manuscript entitled "论文题目(修改后的)". In our revised manuscript, the advices of reviewers were accepted and we make some revise. The list of revised items was upload as supporting information (filename: list of revised items). The following is the answer for the reviewer’s comment s. Problems Answered:1. On Page 3 (lines 50-55), the authors said that preliminary studies have shown that 4 hours were sufficient for attaining equilibrium. Which are these preliminary studies?, Y ou should give the references.Answer: We made a mistake. We want to say that according to our previous experiments, four hours were sufficient for attaining equilibrium. Our English is not so good. In fact, the composition of two phases remains the same after two hours. In order to make sure that the equilibrium is attained, four hours are given. The time used here for equilibrium and phase splitting was fixed according to results from our previous experiments.2. On Table 2 is expected that: w1,II + w2,II + w3,II + w4,II be equals to 1. However, using your results for the last series I obtain the following result: 0.327 + 0.658 + 0.016 + 0.002 = 1.003. Why?Answer: According to the original data, it is a mistake that 0.327 should be 0.324.3. In the Abstract and in the page 5 (line 24) the authors stated that “the reliability of the experimental data were taste using the Othmer –Tobias correlation”. Good correlation results with the empirical Othmer –Tobias equation are not criteria accuracy of the fitting data. This is wrong. Good representation of the experimental data with the empirical equation do not means that the data accurate. Even wrong data can be accurately correlates with any empirical equations.Answer: We think you are right and we revised it. But Reference [16], [17] also said so. The abstract of Reference [16] is following:Liquid-liquid equilibrium (LLE) data for four ternary systems comprising cyclohexane + (benzene, toluene, ethylbenzene, or cumene) + sulfolane have been measured at 303.15 K and at atmospheric pressure. The LLE data for a six-component system including (cyclohexane + benzene + toluene + ethylbenzene + cumene + sulfolane) has also been measured at the above conditions. The reliability of the experimental data was testedusing the Othmer-Tobias correlation. The LLE data were then analyzed using a UNIFAC model with group-interaction parameters extracted from the UNIFAC-LLE data bank and a NRTL version with temperature-dependent binary parameters determined from the experimental LLE data (NRTL/2), both as programmed by the Aspen Plus simulator. Based on the analysis of these data, both models represented the experimental data with sufficient accuracy as revealed from the very small values of the root mean square error and the average absolute deviation in composition.4. Please give some comment how extraction of the samples from each phase is effected on the uncertainty of the measurements. Because, when samples extracting from the phase the equilibrium condition can be slightly changed.Answer: We tried our best to get the samples of two phases at the same time and the amounts of samples are much less compared with that of the material of two phases. So the uncertainty that extraction of the samples brought up can be neglected reasonably. We have reviewed the final version of the manuscript and approve it for publication. All authors have read the manuscript and approved to submit it to your journal. I have not yet received a reply and am wondering whether you have reached a decision. I should appreciated your letting me know what you have decided as soon as possible.四、关于论文的总体审查意见1. This is a carefully done study and the findings are of considerable interest. A few minor revision are list below.2. This is a well-written paper containing interesting results which merit publication. For the benefit of the reader, however, a number of points need clarifying and certain statements require further justification. There are given below.3. Although these observation are interesting, they are rather limited and do not advance our knowledge of the subject sufficiently to warrant publication in PNAS. We suggest that the authors try submitting their findings to specialist journal such as –4. Although this paper is good, it would be ever better if some extra data were added.5. This manuscript is not suitable for publication in the journal of –because the main observation it describe was reported 3 years ago in a reputable journal of - .6. Please ask someone familiar with English language to help you rewrite this paper. As you will see, I have made some correction at the beginning of the paper where some syntax is not satisfactory.7. We feel that this potentially interesting study has been marred by an inability to communicate the finding correctly in English and should like to suggest that the authors seek the advice of someone with a good knowledge of English, preferable native speaker.8. The wording and style of some section, particularly those concerning HPLC, need careful editing. Attention should be paid to the wording ofthose parts of the Discussion of and Summary which have been underlined.9. Preliminary experiments only have been done and with exception of that summarized in Table 2, none has been repeated. This is clearly unsatisfactory, particularly when there is so much variation between assays.10. The condition of incubation are poorly defined. What is the temperature? Were antibody used?五、给编辑的回信1. In reply to the referee’s main criticism of paper, it is possible to say that –One minor point raised by the referee concerns of the extra composition of the reaction mixture in Figure 1. This has now been corrected. Further minor changes had been made on page 3, paragraph 1 (line 3-8) and 2 (line 6-11). These do not affect our interpretation of the result.2. I have read the referee’s comments very carefully and conclude that the paper has been rejected on the sole grounds that it lake toxicity data.I admit that I did not include a toxicity table in my article although perhaps I should have done. This was for the sake of brevity rather than an error or omission.3. Thank you for your letter of –and for the referee’s commentsconcerning our manuscript entitled “”. We have studied their comments carefully and have made correction which we hope meet with their approval.4. I enclosed a revised manuscript which includes a report of additional experiments done at the referee’s suggestion. You will see that our original findings are confirmed.5. We are sending the revised manuscript according to the comments of the reviewers. Revised portion are underlined in red.6. We found the referee’s comments most helpful and have revised the manuscript7. We are pleased to note the favorable comments of reviewers in their opening sentence.8. Thank you for your letter. I am very pleased to learn that our manuscript is acceptable for publication in Cancer Research with minor revision.9. We have therefore completed a further series of experiments, the result of which are summarized in Table 5. From this we conclude that intrinsic factor is not account.10. We deleted the relevant passage since they are not essential to the contents of the paper.11. I feel that the reviewer’s comments concerning Figures 1 and 2 result from a misinterpretation of the data.12. We would have include a non-protein inhibitor in our system, as a control, if one had been available.13. We prefer to retain the use of Table 4 for reasons that it should be clear from the new paragraph inserted at the end of the Results section.14. Although reviewer does not consider it is important to measure the temperature of the cells, we consider it essential.15. The running title has been changed to “”.16. The Materials and Methods section now includes details for measuring uptake of isotope and assaying hexokinase.17. The concentration of HAT media (page12 paragraph 2) was incorrectly stated in the original manuscript. This has been rectified. The authors are grateful to the referees for pointing out their error.18. As suggested by both referees, a discussion of the possibility of laser action on chromosome has been included (page16, paragraph 2).19. We included a new set of photographs with better definition than those originally submitted and to which a scale has been added.20. Following the suggestion of the referees, we have redraw Figure 3 and 4.21. Two further papers, published since our original submission, have been added to the text and Reference section. These are:22. We should like to thank the referees for their helpful comments and hope that we have now produced a more balance and better account ofour work. We trust that the revised manuscript is acceptable for publication.23. I greatly appreciate both your help and that of the referees concerning improvement to this paper. I hope that the revised manuscript is now suitable for publication.24. I should like to express my appreciation to you and the referees for suggesting how to improve our paper.25. In this paper, we repo rt an unexpected, acute effect of GDNF on A-type potassium channels, leading to a potentiation of neuronal excitability, in the dopaminergic neurons in culture as well as in adult brain slices. Further, we show that GDNF regulates the K+ channels through a mechanism that involvesactivation of MAP kinase. Thus, this study has revealed, for the first time, an acute modulation of ion channels by GDNF. Our findings challenge the classic view of GDNF as a long-term survival factor for midbrain dopaminergic neurons, and suggest that the normal function of GDNF is to regulate neuronal excitability, and consequently dopamine release. These results may also have implications in the treatment of Parkinson’s disease.Due to a direct competition and conflict of interest, we request that Drs. XXX of Harvard Univ., and YY of Yale Univ. not be considered as reviewers. With thanks for your consideration, I amSincerely yours,case2Dear Editor,We would like to submit the enclosed manuscript entitled "Ca2+-binding protein frequenin mediates GDNF-induced potentiation of Ca2+ channels and transmitter release", which we wish to be considered for publication in Neuron. We believe that two aspects of this manuscript will make it interesting to general readers of Neuron. First, we report that GDNF has a long-term regulatory effect on neurotransmitter release at the neuromuscular synapses. This provides the first physiological evidence for a role of this new family of neurotrophic factors in functional synaptic transmission. Second, we show that the GDNF effect is mediated by enhancing the expression of the Ca2+-binding protein frequenin. Further, GDNF and frequenin facilitate synaptic transmission by enhancing Ca2+ channel activity, leading to an enhancement of Ca2+ influx. Thus, this studyhas identified, for the first time, a molecular target that mediates the long-term, synaptic action of a neurotrophic factor. Our findings may also have general implications in the cell biology of neurotransmitter release.[0630][投稿写作]某杂志给出的标准Sample Cover Letter[the example used is the IJEB]Case 3Sample Cover Letter[the example used is the IJEB]Dear Editor of the [please type in journal title or acronym]:Enclosed is a paper, entitled "Mobile Agents for Network Management." In this paper, we report an unexpected, acute effect of GDNF on A-type potassium channels, leading to a potentiation of neuronal excitability, in the dopaminergic neurons in culture as well as in adult brain slices. Further, we show that GDNF regulates the K+ channels through a mechanism that involves activation of MAP kinase. Thus, this study has identified, for the first time, a molecular target that mediates the long-term, synaptic action of a neurotrophic factor. Our findings may also havegeneral implications in the cell biology of neurotransmitter release.[0630][投稿写作]某杂志给出的标准Sample Cover Letter[the example used is the IJEB]Case 3Sample Cover Letter[the example used is the IJEB]Dear Editor of the [please type in journal title or acronym]:Enclosed is a paper, entitled "Mobile Agents for Network Management." JNK2 mRNA levels are relatively higher in SW1116 cells, COLO205 and AGS cells, when compared to the other cell lines tested. However, Similarly JNK proteins levels are high in the three cell lines. However, is low in SW1116 cells, compared to the other two. In addition, the SP compound induced higher levels of apoptosis in AGS and COLO205 cells. It seems that inhibition of phosphorylated JNK may be the mechanism by which the SP compound is acting in these cells. The authors should discuss this potential possibility in the Discussion section.Answer: Several sentences have been changed in the Discussion (page 12, paragraph 1) in the revised version to address this issue.2. The author should mention why they chose the six cell lines for their study.Answer: AGS, HT-29 and COLO205 are widely used gastrointestinal cell lines for MAPK pathway study. And the rest three cell lines are commonly investigated in our Lab.3. Surprisingly, inhibition of MKN-45 cell proliferation did not occur until 72 h. Could this be due to slower growth of the cells in the first place?Answer: Actually, we didn’t find MKN-45 cell grow obviously slower than the other cell lines in our study.DisclaimerThe templates are prepared by Medjaden Bioscience Limited (, medjaden@) as references, for our fellow Chinese investigators to publish their research articles in English journals. In the studies with the caspase inhibitor, the authors have only mentioned their results in COLO205. It would be expected that a similar result would have been obtained for AGS cells as well. If so, for the sake of completeness of the report they could add a line stating so.Answer: The statement of AGS has been added in Results (page 10, paragraph 1, line 4) in the revised version.Additional Minor Comments:Materials and Methodsa. 2.1 Cell cultures.-In the first line, MKN-45 cell line is not stated.Answer: MKN-45 has been added in the revised version.b. 2.4 Detection of Caspase-3 activity- typographical error -pH (line 10)Answer: Correction has been made in the revised version.ResultsFirst line- page 10 typographical error- caspase-9Answer: Correction has been made in the revised version (page 10, paragraph1, line7).Referencesa. Can the authors describe clearly the relations between phosphorylated JNK p38 p54, p56 and JNK1 and JNK2, and the significance of their expression?Answer: Several sentences have been added in the Introduction (page 3, paragraph 1) and Results (page 8, paragraph 3 and page 9, paragraph 1) in the revised version to address this issue.2. The experiments are extensive in the present study. However, the author may wish to perform a further functional study to prove that the differential response in cells to SP-600125 is associated with their basal JNK2 expression.Answer: Yes, further functional study will be conducted to prove the correlation between basal JNK2 expression and cell response to SP-600125, eg. transfection of JNK2 siRNA or antisense.3. There appear some illegal symbols, such as "20?, 37?, etc" in the Methods section.Answer: Corrections have been made in the revised version.4. It is unclear why the author chose to use the dose of 20然 of SP-600125 for treatment of all cancer cell lines. It is recommended that the authors shows a dose response curve for the different cell lines.DisclaimerThe templates are prepared by Medjaden Bioscience Limited (, medjaden@) as references, for our fellow Chinese investigators to publish their research articles in English journals. Medjaden encourages authors to use the templates in a creative way because the situation of every article is unique and is different from each other. Copy/paste of thisAnswer: Hideshima T et. al state in their paper “Biologic sequelae of c-Jun NH2-terminal kinase (JNK) activation in multiple myeloma cell lines” that SP600125 inhibits cell growth in a dosage dependent manner (2.5~20uM) in myeloma cell lines. SP600125 20uM in our experiment were found effectively exert its inhibitory effect without cytotoxicity. This reference has been quoted as ref.16 in the revised version.5. Figure 3 would be improved by including the distribution of all phases of the cell cycle (e.g. G0/G1, G2/M and S phase).Answer: The phases of the cell cycle distribution of AGS and COLO205, the two most responsive cell lines with or without treatments, have been listed in Table-1. The G2/M arrest data have been highlighted.6. The results of apoptosis should be included in table 1.Answer: Since apoptosis is not routinely assessed by single PI staining, we used double staining with Annexin V and PI to detect apoptosis more sensitively. The result was shown in Fig.3.7. Page 11 line 5, "SP-600125 induced 1.5-4.5 more times apoptotic cells in 4 out of 6 cancer cell lines". The authors should be more specific and provide the raw data for all the cell lines in the results section.Answer: Several sentences have been added in the Results (page 9, paragraph 3; page 10, paragraph 1) to address this issue.DisclaimerThe templates are prepared by Medjaden Bioscience Limited (, medjaden@) as references, for our fellow Chinese investigators to publish their research articles in English journals. The publication fees (or costs for printing the color figures, or whatever you are charged) appear too expensive for me at the moment. Although this project was supported by a grant from ****, but the total amount of the grant was RMBXXXX (about $XXXX). Cover Letter 范例1- 投稿《欧洲心血管病杂志》1) Dear editorial board of European Journal of Cardiology,Please find enclosed the manuscript: “The angiotensin-converting enzyme is not a risk factor for myocardial infarction in French individuals”, by Sarah H., et al., t o be submitted as a Short Communication to the European Journal of Neurology for consideration of publication. All co-authors have seen and agree with the contents of the manuscript and there is no financial interest to report. We certify that the submission is original work and is not under review at any other publication. In this manuscript, we report the results of the first study on the genetic and functional roles of the ACE on the risk of suffering a myocardial infarction in the French population. Indeed, we genotyped the rs4341 polymorphism in 531 IS cases and 549 healthy controls, and then performed functional studies by measuring serum ACE protein level and activity in healthy controls, stroke patients at baseline and stroke patients 24h after stroke symptoms onset. The results from our study did not reveal any association of the ACE variant with myocardial infarction, although it affected ACE protein level, and ischemic stroke patients showed lower ACE level than controls in the acute phase but not in the chronic phase. We believe that our findings could be of interest to the readers of European Journal of Cardiology because they bring new and strong evidence that the ACE gene and protein are not a risk factor for myocardial infarction. We hope that the editorial board will agree on the interest of this study.Sincerely yours,Sarah H. on behalf of the authors.Corresponding author: Sarah Hamilton at Cardiovascular Research Laboratory, Marie Curie Research Institute, 75000, Paris, France, xxx@mariecurie.fr, phone number: +33582246xxx, fax number: +33582246xxx.Cover Letter 范例2-《动脉粥样硬化》的修改稿回复2)Dear Dr Enzo Montanero,Thank you for considering the revised version of our manuscript ACE variants and risk of Cardiovascular Disease, by Sarah H. et al. for publication in Atherosclerosis. We are thankful to the referees and the Editor for pointing out some important modifications needed in the report. We have thoughtfully taken into account these comments. The explanation of what we have changed in re sponse to the reviewers’ concerns is given point by point in the following pages. We believe that the comments have been highly constructive and very useful to restructure the manuscript. We also believe that the new data included in the article really improved the quality ofboth our genetic and functional analysis. Indeed, we now show that the A allele of the rs10947 SNP of the ACE gene is a risk factor for all etiologies of cardiovascular disease (the association in the overall population resisted correction for multiple testing by Bonferroni). Moreover, we now show that mRNA levels of the ACE gene are higher in MI cases during the acute phase than in healthy controls or MI cases 3 months after the event. and IS cases in the stable condition. We hope that all these changes fulfil the requirements to make the manuscript acceptable for publication in Atherosclerosis.Looking forward to hearing from you soon.Sincerely yours,Sarah H. and Lucas Delphino on behalf of the authors.Corresponding author: Lu cas Delphino at Alzheimer’s Disease Laboratory, Marie Curie Research Institute, 75000, Paris, France, xxx@mariecurie.fr, phone number: +33582246xxx, fax number: +33582246xxx.Cover Letter 范例3- 投稿《老年神经生物学》3)Dear editorial board of Neurobiology of Aging,Please find enclosed the manuscript: ACE variants and risk of Alzheimer’s Disease, by Sarah Hamilton et al., to be submitted as an Original Research Article to Neurobiology of Aging. All co-authors have seen and agree with the contents of the manuscript and there is no financial interest to report. We certify that the submission is not under review at any other publication. In this manuscript, we report the results of a genetic and functional study in a white population of sporadic Alzheimer’s Disease patients on the risk of suffering cognitive impairments. We believe that our findings could be of interest to the readers of Neurobiology of Aging, because they could have a great impact on the diagnostic, prognostic and treatment of patients with Alzheimer’s Disease. Indeed, the ApoE gene is the only well recognized risk factor for Alzheimer’s Disease at the moment (Goder et al. 1998), and we report here evidence that the ACE gene could also be involved in this disease. Moreover, we show that the variant studied modulates ACE levels and increase the predictive value of the ApoE gene. This study could thus have a great pharmacogenetic interest and bring new and important light in the field of Alzheimer’s Disease management and we hope that the editorial board and the reviewers will agree on the interest of this study.Sincerely yours,Sarah H. and Lucas Delphino on behalf of the authors.Corresponding author: Lucas Delphino at Alzheimer’s Di sease Laboratory, Marie Curie Research Institute, 75000, Paris, France, xxx@mariecurie.fr, phone number: +33582246xxx, fax number: +33582246xxx.Cover Letter (投稿信)Dear Dr Broderick,Please find enclosed the manuscript: Association between Estrogen Receptor Alpha (ESR1) genetic variants and risk of Stroke, by Sarah H., Isaac S., Marta L., Marc C. and Julien S. to be submitted as an new article to Clinical Chemistry. In this manuscript, we report the results of a nested case-control study in a French population on the risk of suffering an ischemic stroke associated with genetic variants in the gene coding for the protein Estrogen Receptor Alpha. We believe that our findings could be of interest to the readers of Clinical Chemistry because they replicate partially the results observed by Rexrode et al. in a white American population and published in Clinical Chemistry in October 2007. Indeed, we demonstrate an association of the rs1271673 SNP T variant and a very common haplotype encompassing the rs1271673 variant with an increased risk of ischemic stroke. We hope that the editorial board and the reviewers will agree on the interest of this study. Sincerely yours,Sarah H..Cover Letter 范例5- 投稿《脑血管疾病》5)Dear editorial board of Cerebrovascular Diseases,Please find enclosed the manuscript: Association of a SNP in the ALOX5AP gene with risk of Ischemic Stroke, by Sarah H., et al., to be submitted as a Original Research Article to Cerebrovascular Diseases for consideration of publication. All co-authors have seen and agree with the contents of the manuscript and there is no financial interest to report. We certify that the submission is original work and is not under review at any other publication. In this manuscript, we report the results of the first study on the genetic and functional roles of the ALOX5AP and PDE4D genes on the risk of suffering an ischemic stroke in the French population. We believe that our findings could be of interest to the readers of Cerebrovascular Diseases because they bring new light on the controversial role of the ALOX5AP and PDE4D genes in stroke and replicate some of the results observed by Helgadottir et al. in an Icelandic population and published in Nature Genetics in 2004. Indeed, we demonstrate an association resistant toBonferroni of the SG13S114 (rs10507391) T allele of the ALOX5AP gene with an increased risk of ischemic stroke in white populations through an actualized meta-analysis, and in the never tested before Iberian population through case-control study. Moreover, we investigated and showed for the first time that ALOX5AP mRNA levels depended on the SG13S114 genotypes and stroke patients had higher ALOX5AP mRNA levels than healthy controls. These results thus support a role for the ALOX5AP gene in stroke and uncover diagnostic and therapeutic expectations. We hope that the editorial board will agree on the interest of this study.Sincerely yours,Sarah H. and Lucas D. on behalf of the authors.Corresponding author: Lucas D. at Stroke Laboratory, Marie Curie Research Institute, 75000, Paris, France, xxx@mariecurie.fr, phone number: +33582246xxx, fax number: +33582246xxx.。
