摘要

本实验探讨了普洱茶茶褐素对高脂SD大鼠的降血脂作用及其作用机理,采用生化试剂盒研究普洱茶茶褐素对实验大鼠血清中T

C、TG、HDL-

C、LDL-

C、LCAT、LPL、HL的动态影响,检测了普洱茶茶褐素对肝脏和附睾组织中HSL活性及其mRNA表达量的影响,分析了普洱茶茶褐素对高脂饮食大鼠粪便中胆固醇和胆汁酸含量的影响,通过电子显微镜观察了普洱茶茶褐素对实验大鼠肝脏病理变化的影响。结果显示:

1、经饲喂高脂饮食4周后,模型组大鼠血清T

C、LDL-C显著升高(P﹤

0.01)、TG显著升高(P﹤

0.05),说明本实验所采用的高脂血症大鼠模型是成功的。

2、与高脂模型组相比,治疗组高脂血症大鼠血清T

C、LDL-C水平显著降低(P﹤

0.01),TG显著降低(P﹤

0.05)、HDL-C水平明显升高。说明普洱茶茶褐素具有降血脂作用;预防组高脂饮食大鼠血清中T

C、TG、LDL-C水平升高HDL-C水平降低受到抑制,与空白对照组没有显著性差异(P﹥

0.05),说明茶褐素能减少肝脏脂肪沉积,具有预防实验动物脂肪肝形成的作用;正常试物组大鼠血清T

C、TG、HDL-

C、LDL-C水平与空白对照组相比也没有显著性差异(P﹥

0.05),这说明茶褐素对正常大鼠血脂水平影响不显著。

3、普洱茶茶褐素可以增强高脂血症大鼠HL、HSL活性,其中HL活性上升了

62.50%,肝组织和附睾组织中分别上升了

51.00%和

163.41%。同时,茶褐素能显著提高大鼠肝组织和附睾脂肪组织中HSL的mRNA表达量,表明茶褐素能通过调节脂质代谢关键酶活性来发挥其降脂功效,基本明确了茶褐素降脂作用的酶学和分子生物学作用机制。

4、测定结果表明,茶褐素促使粪便中胆固醇和胆汁酸的含量分别增加了

21.11倍和

4.08倍,与高脂模型组相比,差异极显著(P﹤

0.01)。说明普洱茶茶褐素具有抑制大鼠体内外源性胆固醇的吸收,加速其排泄的作用。

5、对不同发酵阶段茶褐素的降血脂效果进行了研究,结果表明四翻茶褐素的降血脂效果较好。

Abstract

This study investigate at the effects of the reduced blood ester of the TB on the high ester SD rat and its therapeutic mechanism.Standard biochemical kits were used to detect dynamics of lipid contents in serum.HSL mRNA expression and HSL activity were used to detect in liver and the fat of epidymis.The cholesterol of the stool,fecal bile acids were measured. The results include:

1、It shows in this subject that the amount of TC in the blood serum of the rat fed on high

fat diet for 4 weeks and the amount of TC,LDL-C in the blood serum increase remarkably(P﹤

0.01),the amount of TG in the blood serum increase remarkably (P﹤

0.05),which indicates that the rat served as hyperlipdemia animal model aresuccessful.
