在日本,韩国和中国,蘑菇类多糖中的香菇多糖,裂褶菌多糖和云芝多糖已被认可为具有免疫调节活性的。
多糖的活性是通过其构象,组成和大小决定的。
在来源广泛的真菌中,担子菌以及从子实体和菌丝体培养是药用的具有生物活性的多糖的较高来源。
大多数具有生物活性蘑菇多糖是β-(1-3)(1-6)-葡聚糖,有些是杂多糖。
在担子菌发现活性葡聚糖α-和β-构都含有。
一些葡聚糖在蛋白质或肽残基中,以蛋白多糖的形式存在。
本研究的目的是评估和比较从药用真菌物种中热水提取的多糖抗氧化性能和免疫调节作用,包括双孢菇,姬松茸,灵芝,和桑黄多糖。
该多糖提取物的结构是由红外光谱(FT-IR)和糖成分被酸水解,高效液相色谱法(HPLC)分析后确定的。
研究其抗氧化活性采用方法有:共轭二烯法,还原力, DPPH 法和亚铁离子螯合能力法。
免疫调节是在体外进行测试,通过用酶联免疫吸附测定法(ELISA)测量细胞因子的合成。
2.材料和方法2.1.化学制品1,1 - 二苯基-2 - 苦基苯肼(DPPH),硫酸亚铁,氢,铁氰化钾,氯化铁,亚油酸,牛血清,白蛋白(BSA),考马斯亮蓝G-250,三氟乙酸,(TFA),吐温20,氯化亚铁,菲洛嗪,标准试剂例如α-生育酚,抗坏血酸,合成抗氧化剂叔丁基羟基甲苯(BHT),柠檬酸和乙二胺四乙酸(EDTA)购自Sigma化学公司(圣路易斯购买, MO,USA)和Merck公司(达姆施塔特,德国)。
无水甲醇(甲醇Optigrade)由德国LGC Promochem提供。
干扰素-γ(IFN-c),链霉亲和素-HRP,佛波醇 12 - 肉豆蔻酸酯13 - 乙酸酯(PMA)和钙离子载体从BioSourceTM(美国)购买。
含有L-谷氨酰胺和碳酸氢钠RPMI-1640培养基是从西格玛化工有限公司(圣路易斯, MO,USA)购买。
分析蘑菇β-葡聚糖试剂盒购自Megazyme (爱尔兰Wicklow)。
2.2.样品制备野生型桑黄的热水提取物是由奇异恩典健康产业(泰国曼谷)提供的。
A级灵芝孢子是从福建仙芝楼生物科学与技术有限公司(中国)提取的。
姬松茸和双孢蘑菇子实体是从蘑菇实验站所得到(霍斯特,荷兰)。
粗多糖提取物通过热水提取。
多糖在65%的乙醇中进行醇沉和反复洗涤以除去过量的甘露醇和微溶于乙醇的酚类化合物。
该沉淀物在42℃干燥,在真空条件下储存以供进一步使用。
2.3.多糖分析多糖提取物使用一个美国尼高力仪器公司6700型分光光度计记录的FT-IR光谱。
分光光度计配备DTGS TEC探测器和OMNIC7.3软件。
光谱采用KBr压片法记录在 400-4000cm-1范围内。
2.4 .葡聚糖含量测定总量和α-葡聚糖的测定用蘑菇多糖提取物和酵母β-葡聚糖测定方法K - YBGL 09/2009 ( Megazyme的诠释。
)酶试剂盒,包含exo-1,3 - β-葡聚糖酶,β-葡糖苷酶,淀粉葡糖苷酶,蔗糖,葡萄糖测定用试剂( GOPOD -葡萄糖氧化酶,过氧化物酶, 4 - 氨基安替比)和葡萄糖标准溶液。
估算多糖样品中总葡聚糖含量时在60%的硫酸(v/v)100℃下水解1h和2h后冷却。
中和后,在醋酸钠缓冲液(pH 4.5 )中40℃下水解1h。
葡萄糖利用外切β- (1→ 3 )-D-葡聚糖酶加β-葡糖苷酶的混合物。
为了测量总葡聚糖含量,加入葡萄糖氧化酶/过氧化物酶试剂。
所有的溶液510 nm处测吸光值。
经淀粉葡萄糖苷酶与蔗糖酶水解,估计α-葡聚糖含量。
发色团的发现用葡萄糖过氧化酶试剂根据总葡聚糖含量法进行测定。
该β-葡聚糖含量的计算方法是从总葡聚糖含量中减去α-葡聚糖含量。
葡聚糖全部值与每100g干物质的葡萄糖等量。
2.5 .多糖含量和组成的测定所提取的多糖的总多糖含量用苯酚 - 硫酸法测定,与D-葡萄糖作为参考。
进行单糖组分的分析如(Smiderle等,2010)发表的方法。
冻干样品进行水解,用2Mmol的TFA在100 ℃过夜,然后蒸发至干。
残余TFA用0.5毫升甲醇中除去,重复两次,并最终溶于0.5毫升水中。
单糖稀释100倍后,使用Pac PA- 1柱( 4 - 250mm)进行HPLC分析,并用20mM 的NaOH等度洗脱25μL样品溶液( 1毫升/分钟)。
ED40电化学检测器配有脉冲安培电池。
以葡萄糖和半乳糖为标准。
2.6.蛋白质测定蛋白质浓度用Bradford 法( 1976)测定,以BSA为标准。
提取物总蛋白含量与每100克BSA的干重等值。
2.7.脂质过氧化作用的抑制测定各多糖粉末(0.1至20mg/ml, 100Ⅱ)在纯水中用2ml10mM的亚油酸混合,在0.2M磷酸钠缓冲液(pH6.5)乳化。
溶液中加入6.5mM吐温20,使乳化稳定,在黑暗中在37 ℃摇动至加速氧化。
培养15h后加入0.2ml抗氧化剂与6毫升无水甲醇的混合物。
使用紫外/可见分光光度计在234nm处测混合物上清液的吸光度,空白样是使用方案中所有其它试剂,但没有提取样品。
抗氧化性由下式计算:其中A0为对照反应, A1为样品吸光度。
以抗坏血酸和α-生育酚用作阳性对照。
100 %的值表示最强的抑制能力。
2.8 .三价铁还原力抗氧化能力测定还原力测定根据Oyaizu法( 1986)测定。
各多糖粉末中加入纯水(0.1至20mg/ml,2.5毫升),加入2.5ml的0.2M磷酸钠缓冲液(pH混合6.6 )和2.5ml的1%铁氰化钾。
将混合物混匀,在50℃水浴 20分钟。
然后加入2.5ml 10%(重量/体积)的三氯乙酸。
将混合物在2000转离心10分钟。
上层(5毫升)中,用5ml的纯水和1ml的0.1%氯化铁混合。
用紫外/可见分光光度计在700nm处测定吸光度。
空白是方案中所有其它试剂,但没有提取液。
具有较高的吸光度表示较高的还原能力。
以抗坏血酸作阳性对照。
2.9. DPPH自由基清除活性该测定方法是根据Bilos方法(1958)进行了修改。
将第一系列的每种多糖粉末( 0.1-10mg/ml,2毫升)溶于纯水中,加入1ml现配置的含有0.2毫升DPPH的DMSO 溶液。
在第二系列中的每个样品,用1ml DMSO溶液混合。
剧烈搅拌混合物1分钟,在避光条件下20 ℃中水浴 40分钟。
将所得溶液在517nm处用紫外/可见分光光度计测定吸光值。
DPPH自由基清除活性的计算方程:其中Ai是2毫升样液与1毫升DPPH溶液混合的吸光度, Aj是2毫升提取物样液用1ml DMSO溶液混合的吸光度,Ac是空白样,2毫升DMSO中,用1ml的DPPH溶液混合的吸光度。
以抗坏血酸, BHT和α-生育酚溶解在DMSO中用作阳性对照。
2.10 .亚铁离子螯合能力的测定每种多糖粉末溶于纯水(0.1至20mg/ml),取1毫升。
与3.7毫升的甲醇和0.1ml的2mM的氯化亚铁溶液混合。
加入0.2毫升5mM的ferrozine试剂后在室温下反应10分钟,在562nm处测定混合液的吸光值。
空白样加入所有其它试剂,但没有提取样品。
较低的吸光度表示较高的螯合能力。
EC 50值(毫克提取物/毫升)是亚铁离子50%被螯合的有效浓度。
柠檬酸和(EDTA )用于对照。
2.11 .体外测定免疫调节作用2.11.1 .细胞外周血单核细胞(PBMC)从人血的血沉棕黄层制备(由荷兰输血服务中心提供)。
血沉棕黄层用磷酸盐缓冲盐水(PBS , pH 7.2)进行1:1稀释,并在一层Histopaque 1077 淋巴细胞分离液(Sigma公司)上2500转离心15分钟。
精心收集细胞且用PBS洗涤。
细胞进行计数并重新悬浮在5-10 * 106/ml的含10%的胎牛血清( FCS)、1%青霉素和1%链霉素的RPMI-1640培养基中。
2.11.2 .免疫调节活性多糖溶液( 50 LG /毫升)在95℃加热 20分钟预热。
不同多糖提取液的免疫调节活性由1毫微克/毫升的PMA和0.5毫克 /毫升钙离子载体刺激外周血单个核细胞测试。
在37 ℃,5% CO 2中培养48和72小时后,培养基是根据厂商协议通过夹心酶连免疫分析法检测过IFN-γ干扰素的。
IFN –γ的检测范围是为7.81-1000Pg/ml。
2.12 .统计分析所有抗氧化活性进行了三次重复试验,免疫调节活性进行了四次重复试验。
该结果被报告为SEM的平均值±标准误差。
抗氧化活性采用单因素模块(ANOVA )进行方差分析,进一步的比较采用Fisher最小显著差异( LSD )P = 0.05 。
IFN -γ平均值的比较采用单因素方差分析(ANOVA ), Dunnett检验,然后用来比较所有治疗组与对照组(p = 0.05 )。
统计分析采用MS Excel (微软Office 2007专业版)。
EC 50值的计算是通过使用在线免费统计回归计算进行线性回归进行分析( /statistics/regression.php )。
多糖,蛋白质和抗氧化剂之间的相关系数r,使用MS Excel进行分析。
