用于治疗高血压、高血脂症等。
卫生部药品标准[WS 3-B -1109-92]对心脉通片没有含量测定指标,为能控制其内在质量,参照《中国药典》一部[1],研究建立关于HP LC 测定心脉通片中葛根素的含量方法,本法操作简便,准确,线性好,可作为控制心脉通片质量的依据之一。
1 仪器与试药TSP -100高效液相色谱仪 (TSP -100泵,TSP -UV100检测器,ANAST AR 色谱工作站)。
葛根素对照品752-200108 (中国药品生物制品检定所),心脉通片样品(广东邦民制药厂有限公司),甲醇(色谱纯)。
2 实验方法及结果2.1 色谱条件 色谱柱:K romasil C 18(4.6mm ×200mm ,5μm );流动相:甲醇-水(30∶70);流速:110ml Πmin ;检测波长:250nm 。
理论板数按葛根素计算不应低于2000。
2.2 阴性对照试验 照处方比例制备葛根的阴性对照样品,依“样品测定”项下制备供试品溶液,进样分析,结果表明,其他成分对葛根素测定无干扰。
2.3 线性试验 精密称取葛根素对照品10mg ,置50ml 容量瓶中,加入30%乙醇溶解并稀释至刻度,摇匀。
分别取1、2、4、8、16ml 置50ml 容量瓶中,加入30%乙醇稀释至刻度,摇匀。
各取10μl 注入液相色谱仪,记录色谱图。
以峰面积为纵坐标,对照品浓度为横坐标,进行回归分析,得回归方程:Y =-18657.7+54112.8X r =0.9990浓度在4~64μg Πml 范围内线性关系良好。

近年来,基因治疗已成为最活跃的研究领域之一。其在遗传疾病, 心血管疾病及肿瘤等方面取得突破性进展。目前的基因治疗载体可分为 病毒型载体和非病毒型载体,病毒性载体具有转导效率高而被较多应用 的优点,但也能诱导宿主免疫反应,装载容量有限等缺点。而非病毒载 体具有安全性高,容易制备和修饰,基因容量大的优势。 聚乙烯亚胺(polyethylenimine,PEI)是至今设计最为成功的一种非 病毒载体聚合物,聚乙烯亚胺含阳离子,具有较强的结合pDNA的能力。 此外PEI几乎在任何pH条件下均具有吸收质子的能力,因此可保护pDNA 免受溶酶体降解,PEI的转染效率较高,但细胞毒性很大,严重限制了 其应用。PEI的毒性与分子量有关,分子量越大,细胞毒性越大,为了克 服毒性问题,人们使用低分子量PEI进行基因转染,虽然其毒性小,但转 染效率却大大降低。 依普罗沙坦(ES)是一种可以与肿瘤细胞表面AT1受体特异性结合的 化学药物,该药物水溶性较差,其分子结构中有两个羧基可与PEI上的氨 基形成偶联物,本研究中ES起着重要作用:首先ES作为一个连接臂,将 低分子量的PEI链接成高分子量的PEI,大大降低毒性的同时提高了转染 效率。其次,ES作为靶向基因可以与某些肿瘤细胞表面高表达AT1受体特

抗生素错用和滥用不仅没有达到治疗细菌感染性疾病的效果,反而会刺激细菌发生DNA损伤修复反应(SOS反应),加剧细菌耐药性的进化和耐药菌的传播。
本文聚焦于耐药菌,简明介绍细菌耐药性与SOS反应,系统概述了质谱技术、微流控技术及其联用技术在细菌检测及细菌耐药机制研究中的应用。
本文为细菌耐药性相关的药物靶点挖掘及新药开发提供理论参考,以期发展细菌耐药性快速检测新方法和抑菌新方法,推动临床细菌感染性疾病的诊断与治疗。

BIOLOGICAL ACTIVITY:
OICR–9429 is a novel small–molecule antagonist of the Wdr5–MLL interaction with IC50 of 5 uM. inhibit proliferation and induce differentiation .

IC 50: 5 uM
In vitro: OICR–9429 inhibit proliferation and induce differentiation in p30–expressing human AML cells. OICR–9429 cause a significant decrease in viability in the majority of patient cells with mutations in the N–terminal part of the CEBPA gene. OICR–9429 displays
exquisite cellular selectivity and specificity in disrupting critical protein–protein interactions between WDR5. [1] reduce the viability of primary human AML cells with N–terminal C/EBPα mutations by about 50% (mean value, n = 8) at 5 μM

In vivo:The reference for OICR–9429 to mice (female NOD–SCID) is 3 mg/kg.
