支气管哮喘(简称哮喘)是指由多种细胞(如嗜酸性粒细胞、

T 淋巴细胞、肥大细胞、中性粒细胞等)和细胞组分参与形成的以气道慢性炎症为特征的异质性疾病[1]。目前西医治疗多以糖皮质激素、β2受体激动体、白三烯受体拮

抗剂、茶碱类等为主,但部分患者存在耐药性、用药依从性差等问题导致复发率高,临床疗效存在一定局限性。哮喘属于中医“哮病”和“喘病”范畴,认为与宿痰伏肺、痰阻气道、肺失宣降密切相关,通过辨证结合辨病治疗,具有标本兼治的优势。定喘汤出自《摄生众妙方》,是中医临床治疗哮喘的常用方,适宜治疗哮喘属风寒外束、痰热内蕴型喘证者。但是由于成分复杂,其治疗哮喘的作用机制尚不完全明确。网络药理学是从整体观角度认识药物与机体的相互作用并指导新药发现,可提高新药临床试验的成功率,与复杂中药成分治疗疾病理念相一致。因此,本文采用网络药理学方法预测分析定喘汤中的药物成分与靶蛋白、靶蛋白之间、靶蛋白参与通路等情况,探讨其治疗哮喘的作

摘要:目的采用网络药理学方法探讨定喘汤治疗支气管哮喘的作用机制。方法利用TCMSP 筛选定喘汤药物的有效

活性成分,并通过GeneCards 等数据库进行检索获得支气管哮喘相关的疾病靶点。采用DAVID v.6.8数据库进行GO 富集分析及KEGG 通路富集分析。结果经数据库分析,定喘汤与支气管哮喘的共同靶基因为286个。GO-BP 功能分析结果显示,共同基因主要作用于正向调控RNA 聚合酶Ⅱ启动子的转录、血管生成、ERK1和ERK2级联反应等,负向调控凋亡过程等。KEGG 富集分析结果显示,共同靶点可以与哮喘相关的T 细胞受体信号通路、Fc εRI 信号通路、B 细胞受体信号通路等多条信号通路发挥作用。通过与常用西药相关蛋白比较发现,定喘汤可发挥β2受体激动剂和吸入皮质激素的作用。结论定喘汤从多成分、多靶点、多通路的角度治疗支气管哮喘的作用机制对后续深入研究具有重要意义。关键词:定喘汤;支气管哮喘;网络药理学;信号通路中图分类号:R256.12文献标志码:A 文章编号:2096-1413(2021)10-0006-04

ABSTRACT:Objective To explore the mechanism of Dingchuan deco ction in the treatment of bronchial asthma by

network pharmacology method.Methods The active ingredients of Dingchuan decoction were screened by TCMSP,and the disease target associated with bronchial asthma were obtained by searching GeneCards and other databases.GO enrichment analysis and KEGG pathway enrichment analysis were carried out by DAVID v.6.8database.Results Through database

analysis,there were 286common target genes of Dingchuan decoction and bronchial asthma.The GO -BP functional analysis results showed that the common genes were mainly involved in the positive regulation of RNA polymerase Ⅱpromoter transcription,angiogenesis,ERK1and ERK2cascade reaction,and negative regulation of apoptosis and so on.KEGG enrichment analysis results showed that the common target could play a role with asthma related T cell receptor signaling pathway,Fc εRI signaling pathway,B cell receptor signaling pathway and other signaling pared with common western medicine related proteins,Dingchuan decoction can play the role of β2receptor agonist and inhaled

corticosteroids.Conclusion The mechanism of Dingchuan decoction in the treatment of bronchial asthma from the perspective of multi-component,multi-target and multi-channel is of great significance for further research.KEYWORDS:Dingchuan decoction;bronchial asthma;network pharmacology;signaling pathway
