新生大鼠成骨细胞体外培养经验总结摘要] 目的:体外培养成骨细胞技术不仅仅在研究其生物学特征及功能方面有重要作用, 而且在研究代谢性骨疾病和重建骨组织方面异常重要。
高效便捷的体外培养方法,是开展体外实验研究的基础。
方法:采用0.25%胰蛋白酶和Ⅱ型胶原蛋白酶联合的二次酶消化法对新生SD大鼠成骨细胞进行原代提取、培养、传代。
通过观察成骨细胞的形态、碱性磷酸酶(ALP)染色和茜素红矿化结节染色法鉴定,确定其细胞活性及增殖情况。
结果:原代成骨细胞多呈混杂细胞,经过3次传代可获取较纯细胞。
成骨细胞呈三角形、梭形、多边形,胞体向外伸展,胞浆丰富,胞核呈卵圆形见,碱性磷酸酶和钙结节染色阳性。
结论:通过体外培养成骨细胞,可获取符合体外细胞生物学对细胞的要求。
为成骨细胞生物学特征及功能、代谢性骨疾病和重建骨组织方面等研究提供基础。

Efficient and convenient in vitro culture method is the basis of in vitro experimental research.Methods: Primary extraction, culture and subculture of osteoblasts from neonatal SD rats were carried out by secondary enzyme digestion with 0.25% trypsin and type II collagenase. By observing the morphology of osteoblasts, alkaline phosphatase (ALP) staining and alizarin red mineralized nodule staining, the cell activity and proliferation were determined.Results: Most of the primary osteoblasts were hybrid cells, and the more pure cells could be obtained after 3 passages. Osteoblasts were triangular, fusiform and polygonal in shape. The body of osteoblasts extended outwards. The cytoplasm of osteoblasts was abundant. The nucleus of osteoblasts was oval. Alkaline phosphatase and calcium nodule were positive.Conclusion: Osteoblasts cultured in vitro can meet the requirements of cell biology in vitro. It provides the basis for the study of osteoblast biological characteristics and functions, metabolic bone diseases and reconstruction of bone tissue. 摘要背景:近几年肥胖尤其是腹型肥胖与骨密度的关系备受关注,有研究认为内脏脂肪面积是骨密度降低的危险因素,也有一些研究认为年龄和内脏脂肪面积及骨密度之间的关系存在交互作用。
目的:探讨人体成分各指标与骨密度的相关性,并预测低骨量的内脏脂肪面积风险切点。
方法:回顾性分析在郑州大学第一附属医院健康管理中心进行定量CT 和生物电阻抗测定的受检者的内脏脂肪面积及骨密度,采用Pearson 相关、多因素线性回归与Logistic 回归分析法分析人体成分各指标和骨密度及低骨量的相关性。
绘制受试者工作特征曲线,确定预测低骨量的内脏脂肪面积切点值。
结果与结论:①随着年龄的增加,骨密度逐渐下降,内脏脂肪面积逐渐增加,基础代谢率逐渐下降;②多因素线性回归分析显示年龄、内脏脂肪面积与骨密度呈负相关;③多因素Logistic 回归分析显示,绝经后女性内脏脂肪面积和低骨量相关;④受试者工作特征曲线预测绝经后女性低骨量的内脏脂肪面积切点值为117.85 cm 2;⑤提示内脏脂肪面积是除了年龄外预测低骨量的危险因子,有望在今后骨矿物质健康管理中发挥重要作用,应对达到内脏脂肪面积风险切点的绝经后女性早期加强减脂管理,以减少低骨量的发病率及医疗和经济负担。

尼可地尔联合麝香保心丸对冠心病心绞痛患者高敏C反应蛋白及基质金属蛋白酶-9的影响张洪霞;李文杰【摘要】目的:观察尼可地尔联合麝香保心丸对冠心病心绞痛患者血清中高敏C 反应蛋白(hs-CRP)及基质金属蛋白酶-9(MMP-9)的影响。
方法选择60例经确诊为冠心痛心绞痛的患者,分为2组,对照组给予正规西药治疗;治疗组在正规西药治疗的基础上加尼可地尔及麝香保心丸治疗。
所有患者均于入院时以及治疗后8周测高敏C反应蛋白(hs-CRP)、基质金属蛋白酶-9(MMP-9)。
结果治疗组治疗后hs-CRP为(114.99±31.84)mg/L、MMP-9为(188.04±121.01)ng/mL,各项水平明显低于对照组(P<0.05)。
结论尼可地尔联合麝香保心丸通过抗炎机制与西药联合对冠心痛心绞痛的治疗具有肯定的疗效。
%Objective To observe the effect of nicorandil combined with Shexiang Baoxin Pill on coronary heart disease angina pectoris of serum C reactive protein (hs-CRP) and matrix metalloproteinase-9 (MMP-9) effect.Methods 60 cases of hospitalized patients with -12 in the Department of internal medicine in January 2013 were selected. Diagnosed as coronary heart disease and angina pectoris weredivided into two groups, the control group was given regular treatment of Western medicine; treatment group given regular western medicine plus nicorandil and musk to guarantee the heart pill treatment. Serogrouping showed that 52% of the isolates belong to one of the serotypes O78,O2 and O1. Antibiograms revealed a high level of resistance to oxytetracycline, trimethoprim-sulfmethoxazole and enrofloxacin (87, 70 and 45%, respectively). A low percentage of strains were resistant to gentamycin and nitrofurane (3 and 2%, respectively). 本研究以实验室威百亩熏蒸的土壤为材料,通过高通量测序技术,研究威百亩熏蒸对土壤细菌群落的影响及熏蒸后土壤微生物群落重建及功能修复的机制。
结果表明:高通量测序总共测得1 062 241个高质量序列,共比对出5 882个细菌分类单元(OTUs )。
熏蒸处理后不同阶段土壤细菌群落结构在门水平上较为类似,但是在种水平差异显著。
威百亩熏蒸处理对微生物群落结构和功能造成很大影响,使得细菌群落a 多样性、群落构建的驱动因素、共发生网 络关系以及碳水化合物代谢、氨基酸代谢、能量代谢等代谢活性在熏蒸处理后的初、中期阶段表现出上升或者下降的趋势,并在处理后的中后期迅速恢复并趋于平稳。
以上结果表明,威百亩处理对细菌群落造成强烈的“生态扰动",导致细菌群落表现出“抑制-激活-恢复"的阶段性特征。
微生物群落的稳定性与群落的抵抗力和恢复力密切相 关。

代谢控制发酵名词解释代谢控制发酵是在发酵过程中通过调控代谢途径,使得微生物产物合成提高、副产物减少或消失的一种控制策略。
代谢(Metabolism)指的是一个生物体对外界物质的摄取、吸收、转化和排泄等一系列化学反应的总和。
代谢可分为两个主要部分:合成代谢和能量代谢。
合成代谢是指生物体从较简单的物质合成复杂的有机物质,如蛋白质、核酸、多糖等。
能量代谢则是指生物体通过氧化降解有机物质释放能量。
在酿酒或发酵过程中,发酵菌通过能量代谢将碳源转化为酒精和二氧化碳,同时合成一些副产物或其他附属物质。

胰岛素与骨代谢的关系韩松梅;陈莉丽【摘要】糖尿病性骨质疏松是由于糖尿病晚期机体胰岛素绝对不足或相对缺乏,引起糖、脂肪、蛋白质三大物质在体内代谢紊乱及钙、磷、镁等元素代谢途径异常,引起机体骨组织显微结构破坏,致其生物力学性能受损,长骨脆性增加和骨基质含量减少,最终发生骨折,是一种全身性骨代谢异常性疾病.在由糖尿病引起的常见慢性并发症中,糖尿病性骨质疏松的发病率逐年递增,然而其具体的发生机制并未完全明确.胰岛素对维持骨基质含量、减少骨量丢失及预防骨折作用显著,早期补充外源性胰岛素可纠正糖尿病性骨质疏松引起的骨基质含量降低,逆转骨重建负平衡.本文通过研究糖尿病后骨代谢指标的变化,综述胰岛素与骨质疏松的关系及其研究现状.%Diabetic osteoporosis is a systemic metabolic disease, in which absolute or relative lack of insulin causes metabolic disturbance of sugar, fat, protein, calcium, phosphorus, magnesium and other elements, and then leads to reduced bone mass, destruction of bone micro-structure and reduced bio-mechanical properties. Finally, the bone fra-gility increases and fracture is prone to occur. Early insulin therapy can correct bone loss and negative balance of bone reconstruction induced from diabetes. 那么,对于有妊娠需求的女性服药者,停药后多久才适合受孕 呢?该文结合双光子荧光成像与三维重构技术研究此问题。
通过单次腹腔注射生理盐水及5 m g/k gM T X建立了对照组和M T X组小鼠,随后建立了M T X注射后代谢5、10、15、20、25及30天组小鼠。
研究发现,小鼠体内成熟卵母细胞的纺锤体正常形态比率与染色体正常空间排布比率,分别于代谢 15天和25天后,显著高于M T X组,并恢复至对照组水平。
表征卵母细胞功能的体外受精率、二细胞 胚胎率及囊胚率,发现分别于代谢15、20、25天后,恢复至对照组水平。
该文结果表明,自身代谢可 恢复M T X所引起的卵母细胞质量受损及功能下降,所需时间约为小鼠的5个生理周期,即25天。

Automated Metabolic Reconstructionat the Molecular LevelMasanori Aritaari@ims.u-tokyo.ac.jpHuman Genome Center,Institute of Medical Science,University of Tokyo4-6-1Shirokanedai,Minato-ku,Tokyo108-8639,Japan1IntroductionThe analysis of metabolism in bacteria facilitates a new application area such as drug synthesis or toxin degradation,using genetically engineered bacteria.However,the understanding of bacterial metabolism is still premature,because in a newly sequenced bacterium,gene functions are determined for half its total ORF s.Only from sequence similarity,it is impossible to decide functions of the remaining ORF s.It seems speculative to estimate metabolism from only half of its component,but the good news is basic metabolism is conserved throughout species.Therefore,the basic strategy for predicting pathways is to(1)map known functions to the pathways we already have1;and to(2)fill gaps by inserting a missing piece of a metabolic jigsaw puzzle.The latter step corresponds to the prediction of ORF functions,and not only sequence similarity but substrate motif becomes available in this analysis.For this purpose,we designed and implemented Automated Metabolic Reconstruction(AMR) system,which includes the database of chemical molecules,the database of enzymatic functions, and the deduction engine for computing putative pathways.2MethodInformation on molecular structures is essential for the pathway reconstruction,in order to check the validity of pathways and the similarity of molecules.AMR considers atomic structures only,and does not introduce any abstraction at the level of chemical groups or components.The system is written in C++using LEDA library package[4].Database for Compounds:Molecular structures are input in SMILES notation[6]with chiral information.Input data is internally converted and stored in graph format.Aromatic rings are automatically detected at the same time.Many algorithms have been proposed for the normalization of molecular graph structures2,but AMR supports an original efficient algorithm.Database for Enzymes:Given a reaction formula,AMR computes which atoms correspond to which,between both hand-sides of the reaction.This mapping information is pre-computed and stored in the database.The structural matching of graphs is performed by an original efficient algorithm.Data of compounds and enzymes are input in an original language,which can describe a hierarchical structure of cellular compartments.Therefore,the system can treat the same compound differently according to the compartment where it is located.Deduction Engine:The information in the above two databases is considered a graph,in which atoms in compounds and the mapping of enzymes correspond to nodes and edges,respectively.Suchedges are called enzyme edges.A metabolic pathway from compound S to T corresponds to a sequence of edges from an atom in S to T.In addition to these nodes and edges,the graph is augmented with hypothetical edges and nodes,generated from about20basic reactions representing dehydrogenase, kinase,and other common enzymes.All edges are assigned weights,whose biological interpretation is a likelihood of the corresponding reaction.The computation of pathways in this graph is realized with k shortest paths algorithm by Eppstein[1].3Result and DiscussionCurrently,over700compounds are stored in the database,and the search process can efficiently compute pathways.For example,it can reproduce glycolysis and amino acid syntheses in bacteria. The advantage of our system is the enumeration of all the logically possible pathways,including hypothetical reactions.The overall strategy resembles that of an expert system forfinding synthetic paths of chemical compounds[5],but ours has several advantages over previous systems.First,because of the graph matching procedure,the enzyme database is generated only from reaction formulas.Enzyme data is different for each bacterium,but there is no laborious data-input process.Second,the graph theoretic representation of metabolism enabled us the search of much longer pathways,compared to a few steps in previous systems.Finally,a chemical molecule is merely an instance in our framework,and the deduction engine is applicable to other datatypes.Therefore,other biological cascades can be written in our system,provided that appropriate hypothetical edges and their weights can be defined.If a hypothetical edge(s)can recover an important metabolism,the corresponding enzyme(s)is likely to be coded in the ORF s of unknown function.The prediction of gene functions with substrate motifs is an immediate application of this system.However,the output of the system depends sorely on edge weights,and their adjustment is quite important.The computational learning of these weights from examples is our future work.AcknowledgmentsThe author thanks Prof.Morris,Prof.Green(U.Washington),Prof.Akutsu(U.Tokyo),and Prof.Dan-dekar(EMBL Heidelberg)for their continuous help and encouragement.This work is supported by the Research Fellowship of the Japan Society for the Promotion of Science for Young Scientists.
