随后,通过STRING 数据库构建蛋白质-蛋白质相互作用网络,并将STRING 数据库得到的网络数据导入Cytoscape 软件进行分析,计算网络各节点的拓扑特性,筛选出成骨分化关键基因14个,成脂分化关键基因20个。
继而,用Venny 在线工具对成骨分化和成脂分化中的关键基因进行分析,发现7个基因(包括JUNB 、SMAD 2、SMAD 3、MAPK 13、BMP 4、SRC 、RUNX 2)共同参与调控间充质干细胞的成骨分化与成脂分化。
上述筛选出的间充质干细胞发生成骨分化与成脂分化的关键基因,可用于提示间充质干细胞发生异常成骨成脂分化的可能机制,为骨质疏松症等骨与脂肪生成平衡相关临床疾病的发病机制及治疗提供了可能的治疗靶点和新思路。
