芦荟提取物本品为百合科植物库拉索芦荟Aloe barbadensis Miller、好望⾓芦荟Aloe feroxMiller或其他同属近缘植物叶的汁液浓缩⼲燥物。

库拉索芦荟习称“⽼芦荟”,好望⾓芦荟习称“新芦荟”。

【性状】库拉索芦荟呈不规则块状,常破裂为多⾓形,⼤⼩不⼀。

表⾯呈暗红褐⾊或深褐⾊,⽆光泽。

体轻,质硬,不易破碎,断⾯粗糙或显⿇纹。

富吸湿性。

有特殊臭⽓,味极苦。

好望⾓芦荟表⾯呈暗褐⾊,略显绿⾊,有光泽。

体轻,质松,易碎,断⾯玻璃样⽽有层纹。

【鉴别】 (1) 取本品粉末0.5g,加⽔50ml,振摇,滤过,取滤液5ml,加硼砂0.2g,加热使溶解,取溶液数滴,加⽔30ml,摇匀,显绿⾊荧光,置紫外光灯(365nm) 下观察,显亮黄⾊荧光;再取滤液2ml,加硝酸2ml,摇匀,库拉索芦荟显棕红⾊,好望⾓芦荟显黄绿⾊;再取滤液2ml,加等量饱和溴⽔,⽣成黄⾊沉淀。

(2) 取本品粉末0.1g,加三氯化铁试液5ml 与稀盐酸5ml,振摇,置⽔浴中加热5 分钟,放冷,加四氯化碳10ml,缓缓振摇 1分钟,分取四氯化碳层6ml,加氨试液3ml,振摇,氨液层显玫瑰红⾊⾄樱红⾊。

(3) 取本品粉末0.5g,加甲醇20ml,置⽔浴上加热⾄沸,振摇数分钟,滤过,滤液作为供试品溶液。

另取芦荟苷对照品,加甲醇制成每 1ml含5mg 的溶液,作为对照品溶液。

照薄层⾊谱法(附录Ⅵ B)试验,吸取上述两种溶液各 5µl,分别点于同⼀硅胶G薄层板上,以醋酸⼄酯-甲醇-⽔(100:17:13) 为展开剂,展开,取出,晾⼲,喷以10%氢氧化钾甲醇溶液,置紫外光灯(365nm) 下检视。

供试品⾊谱中,在与对照品⾊谱相应的位置上,显相同颜⾊的荧光斑点。

【检查】⽔分照⽔分测定法(附录Ⅸ H 第⼀法)测定,不得过12.0%。

总灰分不得过4.0%(附录Ⅸ K)。

【含量测定】取库拉索芦荟粉未(过五号筛)约0.15g(或好望⾓芦荟粉末约0.2g),精密称定,置500ml 量瓶中,⽤甲醇 1ml湿润,加60℃⽔ 5ml混匀,再加60℃⽔75ml,振摇30分钟,放冷,加⽔⾄刻度,振摇,滤过,精密量取续滤液10ml,置盛有60%三氯化铁溶液1ml 与盐酸6ml 的烧瓶中,加热回流 4⼩时,放冷,转移⾄分液漏⽃中,⽤1mol/L氢氧化钠溶液4ml 和⽔4ml,依次洗涤烧瓶,洗液并⼊分液漏⽃中,⽤四氯化碳提取3 次,每次20ml,合并四氯化碳液,⽤⽔洗涤2 次,每次10ml,弃去⽔液,提取液置100ml 量瓶中,加四氯化碳⾄刻度,摇匀,精密量取20ml,置⽔浴上⼩⼼蒸⼲,精密加⼊0.5%醋酸镁甲醇溶液10ml使溶解,照分光光度法(附录Ⅴ B),在512nm 的波长处测定吸收度,按芦荟苷(C21H22O9)的吸收系数(E1% 1cm)240计算,即得。

芦荟提取技术流程如下:1、表面净化。

首先把鲜叶用清水飘洗干净,最好用深井水,不能用含漂白剂的自来水。

因漂白剂可导致芦荟液变色、变质。

2、外线辐射杀菌。

洗干净的鲜叶晾干后,运入无菌室(室内裴有杀菌消毒设备)进行紫外线辐射杀菌,一般5~10分钟即可。

3、去掉叶皮。

在无菌室内去掉芦荟叶皮,用消毒的刀片割开外皮,并将芦荟凝胶取出。

将凝胶和叶皮分别装入塑料桶或缸内。

4、高速粉碎与沉淀。

用高速粉碎机把叶皮组织分别捣碎,成为稀浆液。

然后用高速沉淀机进行沉淀处理。

之后过滤或真空过滤,即成为芦荟纯原汁。

滤取芦荟皮汁后的滤渣数量不多,约为芦荟鲜叶重量的2%~4%。

此渣可加水煮1小时,滤取其汁,该方法生产产品为芦荟炼汁。

芦荟炼汁直接用火浓缩至饴糖样,冷却后凝成树脂状,即为中药芦荟。

凝胶和叶皮处理后的原汁用途不同。

凝胶原汁主要用于饮料、食品或添加剂等。

叶皮原汁则主要用于护肤、护发化妆品和制造药品等。

芦荟干粉,在使用时,首先要把粉料溶于水,也就是恢复水相成分,扩张系数为1.199。

2、液态芦荟。

把收割来的芦荟鲜叶经过一种特殊混合器进行均化,温度控制在4℃,以每分钟2500转离心机粉碎,约15分钟,待离心液沉淀,即可获浅色清液。

3、Freedom超级芦荟。

国际著名的芦荟权威高比尔(BillCoat)博士与美国达拉斯(国际芦荟科学研究会所在地)的药物研究专家龚查理(CharlieQaeen)合作研究,发明了一种Goldprocesspurificationformula净化方法,用来制造一种不含泻剂成分的芦荟新产品;高比尔/千克的芦荟素,这是最纯正及最有效的芦荟产品。

膜分离技术浓缩纯化芦荟提取液解读芦荟是具有相当医药、保健、美容价值的多功能植物,芦荟产品的开发已成为市场的热点,据国际芦荟产业协会的统计96年美国的芦荟产值就达上百亿美圆。

众所周知,芦荟的加工提取工艺主要是表面净化、杀菌、去皮取胶、粉碎沉淀、过滤浓缩、稳定化或冷冻干燥,基本工艺简单。

在过滤浓缩工艺中采用传统蒸发浓缩的方法浓缩芦荟提取汁一方面蒸汽耗
用量大、浓缩成本高,另一方面升温浓缩破坏活性成分、产品质量下降,因而有条件的企业采用冷冻浓缩,保证浓缩过程芦荟提取汁不受热,有利于其营养物质、活性物质的保存。

然而采用冷冻浓缩法要得到较高的浓缩倍数需要极低的温度,效率很低,在冰晶分离时夹带少量有效成分导致产品损失,并且设备投资巨大。

纳滤膜分离技术在芦荟应用中有一定的优势。

2、膜耐受的条件范围宽,浓缩倍数高。
