四氯化碳诱导大鼠肝纤维化模型改良的研究

摘要:目的 构建肝纤维化大鼠模型,并对经典四氯化碳(CCl 4)复制模型方法进行适当改进。方法 取Sprague -Daw ley(SD)大鼠40只,雌雄各半,按随机数字表法分为A 组10只、B 组14只和C 组16只。B 组给予大鼠腹腔注射40%CCl 4花生油溶液2mL kg -1,2次 周-1,同时给予普通全价饲料喂养(改良方法);A 组为正常对照组,给予等量花生油腹腔注射,2次 周-1,同时给予普通全价饲料喂养;C 组给予大鼠腹腔注射40%CCl 4花生油溶液5mL kg -1,2次 周-1,同时给予添加10%猪油及2%胆固醇的饲料喂养(经典方法)。实验第8周末处死各组剩余大鼠:采用酶联免疫吸附法检测血清肝功能(A L T 、AST 、A L B 、T P 、A/G)水平;H E 及M asson 染色观察大鼠肝组织病理学变化。结果 实验第8周B 、C 组均可见明显肝功能损害表现。B 组可见部分标本肝脏假小叶形成,达到早期肝硬化,肝细胞少量脂肪变性,死亡率28.6%。C 组均已形成明显肝硬化,弥漫性肝细胞脂肪变性,死亡率43.7%。结论 低剂量CCl 4诱导并给予普通饲料喂养可成功建立大鼠肝纤维化模型,并明显降低大鼠死亡率,提高了模型质量及实验效率。

ABSTRACT:Objective To establish a rat m odel of liver fibrosis,and to improv e the classical CCl 4induction method.Methods Forty SD r ats (20female and 20male)w ere r ando mly divided into three gro ups.Gro up B (n =14)w as given intraperitoneal injection of 2mL kg -1of 40%CCl 4solution (a mix ture of CCl 4and peanut oil)tw ice a w eek and the feeding of no rmal complete diet.Gro up A (n =10)w as given intraperitoneal injection of 2mL kg -1of peanut oil tw ice a w eek and the feeding of nor mal com plete diet.Gro up C (n =16)w as given intr aper ito neal injec -tion of 5mL kg -1of 40%CCl 4solution tw ice a w eek and the feeding of diet containing 10%lar d and 2%cholesterol.Rats w ere killed at the end of 8w eeks,and the levels o f ser um ALT ,AST ,Alb,T P and A/G w ere determined by ELISA.The patho logical changes in liver tissue w ere ob -served by H E and M asso n staining.Results Liv er function w as damag ed apparently by CCl 4in -jectio n.Some specimens in g roup B pro duced false flo cculus and achieved the ear ly stage of liver fibro sis,w ith fatty deg ener ation in a few liver cells.A ll specimens in gr oup C had cirrhosis w ith diffuse hepatic steato sis.The m ortality w as 28.6%and 43.7%in gro up B and g roup C,respec -tively.Conclusion Liv er fibrosis can be induced by injection o f low do ses of CCl 4and feeding of norm al co mplete diet in rats.In addition,the improved m ethod can reduce the mortality and im -40南昌大学学报(医学版)2011年第51卷第7期 Jou rnal of Nanch ang University(M edical Science)2011,Vol.51No.7
